    New product development in an emerging economy: analysing the role of supplier involvement practices by using Bayesian Markov chain Monte Carlo technique

    The research question is whether the positive relationship found between supplier involvement practices and new product development performances in developed economies also holds in emerging economies. The role of supplier involvement practices in new product development performance is yet to be substantially investigated in the emerging economies (other than China). This premise was examined by distributing a survey instrument (Jayaram’s (2008) published survey instrument that has been utilised in developed economies) to Malaysian manufacturing companies. To gauge the relationship between the supplier involvement practices and new product development (NPD) project performance of 146 companies, structural equation modelling was adopted. Our findings prove that supplier involvement practices have a significant positive impact on NPD project performance in an emerging economy with respect to quality objectives, design objectives, cost objectives, and “time-to-market” objectives. Further analysis using the Bayesian Markov Chain Monte Carlo algorithm, yielding a more credible and feasible differentiation, confirmed these results (even in the case of an emerging economy) and indicated that these practices have a 28% impact on variance of NPD project performance. This considerable effect implies that supplier involvement is a must have, although further research is needed to identify the contingencies for its practices

    Structure-function relationships of wheat flavone O-methyltransferase: Homology modeling and site-directed mutagenesis

    <p>Abstract</p> <p>Background</p> <p>Wheat (<it>Triticum aestivum </it>L.) <it>O</it>-methyltransferase (TaOMT2) catalyzes the sequential methylation of the flavone, tricetin, to its 3'-methyl- (selgin), 3',5'-dimethyl- (tricin) and 3',4',5'-trimethyl ether derivatives. Tricin, a potential multifunctional nutraceutical, is the major enzyme reaction product. These successive methylations raised the question as to whether they take place in one, or different active sites. We constructed a 3-D model of this protein using the crystal structure of the highly homologous <it>Medicago sativa </it>caffeic acid/5-hydroxyferulic acid <it>O</it>-methyltransferase (MsCOMT) as a template with the aim of proposing a mechanism for multiple methyl transfer reactions in wheat.</p> <p>Results</p> <p>This model revealed unique structural features of TaOMT2 which permit the stepwise methylation of tricetin. Substrate binding is mediated by an extensive network of H-bonds and van der Waals interactions. Mutational analysis of structurally guided active site residues identified those involved in binding and catalysis. The partly buried tricetin active site, as well as proximity and orientation effects ensured sequential methylation of the substrate within the same pocket. Stepwise methylation of tricetin involves deprotonation of its hydroxyl groups by a His262-Asp263 pair followed by nucleophilic attack of SAM-methyl groups. We also demonstrate that Val309, which is conserved in a number of graminaceous flavone OMTs, defines the preference of TaOMT2 for tricetin as the substrate.</p> <p>Conclusions</p> <p>We propose a mechanism for the sequential methylation of tricetin, and discuss the potential application of TaOMT2 to increase the production of tricin as a nutraceutical. The single amino acid residue in TaOMT2, Val309, determines its preference for tricetin as the substrate, and may define the evolutionary differences between the two closely related proteins, COMT and flavone OMT.</p

    Molecular characterization and identification of members of the Anopheles subpictus complex in Sri Lanka

    BACKGROUND: Anopheles subpictus sensu lato is a major malaria vector in South and Southeast Asia. Based initially on polytene chromosome inversion polymorphism, and subsequently on morphological characterization, four sibling species A-D were reported from India. The present study uses molecular methods to further characterize and identify sibling species in Sri Lanka. METHODS: Mosquitoes from Sri Lanka were morphologically identified to species and sequenced for the ribosomal internal transcribed spacer-2 (ITS2) and the mitochondrial cytochrome c oxidase subunit-I (COI) genes. These sequences, together with others from GenBank, were used to construct phylogenetic trees and parsimony haplotype networks and to test for genetic population structure. RESULTS: Both ITS2 and COI sequences revealed two divergent clades indicating that the Subpictus complex in Sri Lanka is composed of two genetically distinct species that correspond to species A and species B from India. Phylogenetic analysis showed that species A and species B do not form a monophyletic clade but instead share genetic similarity with Anopheles vagus and Anopheles sundaicus s.l., respectively. An allele specific identification method based on ITS2 variation was developed for the reliable identification of species A and B in Sri Lanka. CONCLUSION: Further multidisciplinary studies are needed to establish the species status of all chromosomal forms in the Subpictus complex. This study emphasizes the difficulties in using morphological characters for species identification in An. subpictus s.l. in Sri Lanka and demonstrates the utility of an allele specific identification method that can be used to characterize the differential bio-ecological traits of species A and B in Sri Lanka

    Molecular evidence for the presence of malaria vector species a of the Anopheles annularis complex in Sri Lanka

    <p>Abstract</p> <p>Background</p> <p><it>Anopheles annularis s.l</it>. is a wide spread malaria vector in South and Southeast Asia, including Sri Lanka. The taxon <it>An. annularis </it>is a complex of two sibling species viz. A and B, that are differentiated by chromosome banding patterns and ribosomal gene sequences in India. Only species A is reported to be a malaria vector in India while the occurrence of sibling species in Sri Lanka has not been documented previously.</p> <p>Findings</p> <p>Anopheline larvae were collected at a site in the Jaffna district, which lies within the dry zone of Sri Lanka, and reared in the laboratory. Emerged adults were identified using standard keys. DNA sequences of the D3 domain of 28S ribosomal DNA (rDNA) and the internal transcribed spacer-2 (ITS-2) of the morphologically identified <it>An. annularis </it>were determined. BLASTn searches against corresponding <it>An. annularis </it>sequences in GenBank and construction of phylogenetic trees from D3 and ITS-2 rDNA sequences showed that the Sri Lankan specimens, and <it>An. annularis s.l</it>. specimens from several Southeast Asian countries were closely related to species A of the Indian <it>An. annularis </it>complex.</p> <p>Conclusions</p> <p>The results show the presence of the malaria vector <it>An. annularis </it>species A in Sri Lanka and Southeast Asia. Because <it>An. annularis </it>vectors have been long associated with malaria transmission in irrigated agricultural areas in the Sri Lankan dry zone, continued monitoring of <it>An. annularis </it>populations, and their sibling species status, in these areas need to be integral to malaria control and eradication efforts in the island.</p

    Phonological Game-based Module to Enhance Tamil School Linus Program

    This study evaluates the relationship between phonological game based module to enhance reading skills among Tamil school Linus Program students. When one is considered a normal reader, he or she does not encounter much difficulty in reading and understanding texts. There are, however, children who have problems with phoneme identification and thus do not read well and they also have difficulties in comprehending texts. These learners should be identified and be assisted with the game module so that they can proceed with their learning lives. This study was guided by the following objectives: to identify phonological game based scores among the participants; to identify the significances of the phonological game based module in classroom practices. A sample of respondents from standard one from two selected schools in Perak state was used in this study. Phonological awareness skills of these respondents were tested to find out the nature of their phonological awareness. The results indicated that the phonological game based module has a significant role in enhancing the participants' phonological awareness and reading skills

    Comparing Malaysian and Scottish Firms on Practices for Strategic Capability Management

    The study of practices for Strategic Capacity Management at five Malaysian companies and four Scottish companies shows that the Malaysian manufacturing managers acted more reactive due to pressures by sales and processing orders, whereas the Scottish managers were implementing a manufacturing strategy more `independently'. Problems with suppliers, albeit sometimes caused by outsourcing, feature high on the list of challenges in both samples. Alignment of organisational structures and investment in technologies are seen by all as key to aligning the manufacturing strategy with the competitive strategy, though actual investments tend to be happening more in Scottish companies
