    Isobutanol production at elevated temperatures in thermophilic Geobacillus thermoglucosidasius

    The potential advantages of biological production of chemicals or fuels from biomass at high temperatures include reduced enzyme loading for cellulose degradation, decreased chance of contamination, and lower product separation cost. In general, high temperature production of compounds that are not native to the thermophilic hosts is limited by enzyme stability and the lack of suitable expression systems. Further complications can arise when the pathway includes a volatile intermediate. Here we report the engineering of Geobacillus thermoglucosidasius to produce isobutanol at 50 °C. We prospected various enzymes in the isobutanol synthesis pathway and characterized their thermostabilities. We also constructed an expression system based on the lactate dehydrogenase promoter from Geobacillus thermodenitrificans. With the best enzyme combination and the expression system, 3.3 g/l of isobutanol was produced from glucose and 0.6 g/l of isobutanol from cellobiose in G. thermoglucosidasius within 48 h at 50 °C. This is the first demonstration of isobutanol production in recombinant bacteria at an elevated temperature

    -The process of European integration has brought attention to the relation of different national narratives on modern European history. Especially for the construction of a European identity, it seems crucial that states come into a dialogue about their interpretation of the past and find common ground for remembrance and reconciliation. One aspect of the current debate centres around the memory of postwar forced migration. This discussion is connected to a complex, morally ambiguous and very sensitive part of European history. Even seventy years after the end of the Second World War, forced migration is still interpreted in many different ways. Especially in Poland and Germany, memory of postwar forced migration has often given cause for heated debates. It is a problematic issue, because for both countries, it touches upon painful individual memories and is further complicated by the fact that the population movements can only be understood with reference to previous wartime experiences. Since Poland and Germany have been highly involved in the European debate on forced migration, they can serve as informative case studies to investigate, how European integration impact upon the reconstruction of national or transnational historical narratives. Is it possible to discern the emergence of a European historical consciousness? The general aspiration for undertaking this research is to integrate the study of national memory into a wider European framework. The aim is to explore how different arenas of memory construction influence each other and shape historical narratives. This will be done by looking at the German and Polish public memory discourses and by exploring the process of institutionalising the memory of forced migration in museum settings. In this context, it will consider two national memory projects: The Foundation Flight, Expulsion, Reconciliation in Berlin and the Museum of the Second World War in Gdansk
