    Standard electrophoresis methods were used in the qualitative and quantitative protein analyses of cerebrospinal fluid (CSF). Disc electrophoresis was carried out for detection of oligoclonal IgG bands in cerebrospinal fluid on polyacrylamide gel. Pairs of CSF and serum were taken from 30 patients, mainly with multiple sclerosis and other central nervous system dysfunctions, polyradiculoneuritis, known as Guillain-Barre syndrome, encephalitis, paraproteinemia, and analyzed. ImageMaster 1D Elite and GelPro specialized software packages were used for fast accurate image and gel analysis. The results obtained from different hierarchic cluster analysis methods were compared. In some cases, despite substantial similarities between electropherograms, different cluster methods produced different dendrograms. Therefore, the cluster analysis should be used cautiously. It offers only additional diagnostic information on the inflammatory conditions of the central nervous system.Standardne metode elektroforeze upotrebljene su u kvalitativnoj i kvantitativnoj analizi proteina cerebrospinalne tekućine (CSF). Detekcija oligoklonalnih IgG proteina u cerebrospinalnoj tekućini provedena je disk elektroforezom na poliakrilamidnom gelu. Analizirani su uzorci CSF i seruma od 30 pacijenata s multiplom sklerozom i drugim oboljenjima središnjeg živčanog sustava kao što su poliradikuloneuritis, poznat kao Guillain-Barre sindrom, encefalitis i paraproteinemia. ImageMaster 1D Elite i GelPro specijalizirani kompjutorski programi upotrebljeni su za brzu analizu slike i gela. Usporedbom rezultata dobivenih iz različitih hijerarhijskih klaster analiza utvđeno je da različite klaster metode ne daju iste rezultate. Usprkos sličnostima elektroferograma različite klaster metode u nekim slučajevima daju različite dendrograme pa je potreban oprez u interpretaciji rezultata. Klaster analiza daje samo dodatne dijagnostičke informacije o upalnom stanju središnjeg živčanog sustava

    Peripheral glutamate and TNF-α levels in patients with intracerebral hemorrhage: Their prognostic values and interactions toward the formation of the edemal volume

    Objective We aimed to evaluate the prognostic values, contribution and interactions of the peripheral blood plasma glutamate and tumor-necrosis factor-α (TNF-α) levels toward the formation of the perifocal edema in patients with intracerebral hemorrhage (ICH). Methods Fifty patients with ICH and fifty healthy controls were included in the study. The peripheral markers were detected by high-sensitivity ELISA. Results A highly significant differences in plasma glutamate and TNF-α levels with good separation of their values was detected between patients and healthy controls. The two variables correlated with the severity of the symptoms and the initial volume of the ICH at admission. Both peripheral glutamate and TNF-α levels at admission were estimated as significant predictors for the formation of the perifocal edema five days after ICH; nevertheless, it was shown that they independently contribute to the development of the edema, without effects of interaction and regardless the localization of the ICH. Conclusions Our results support the idea for the significance of glutamate and TNF-α as peripheral markers for excitotoxicity and inflammation in ICH patients. The developed multiple regression model for prediction of the development of the edema could be beneficial in decision making between conservative treatment and surgical intervention in the clinical practice

    HPLC investigation of the degradation of some artificial azo food colorants in the presence of ascorbic acid

    The influence of the concentration of ascorbic acid on some azo food colorant (Brilliant Black BN, Brilliant Blue FCF, Chinoline Yellow, Indigotine, Patent Blue V and Tartazine) degradation in the solution has been studied. Rate constant (k), time of colorant half-life (t1/2), as well as the type of the kinetic reaction have been determined. The colorants and the ascorbic acid have been determined by high performance liquid chromatography (HPLC) with diode array detector (DAD) and UV-VIS spectrometry. It was found that the higher concentration of ascorbic acid leads to the higher rate constant and lower colorants half-life. The values of the rate constants are 2.50–4.25 times higher when the colorant degradation was run in the presence of 500 mg/L of ascorbic acid than in the case of the presence of 100 mg/L ascorbic acid for Brilliant Blue FCF, Chinoline Yellow, Patent Blue V and Tartazine. From the other side, the degradation of Brilliant Black BN and Indigotine is very fast with a total degradation for 6–9 days. Therefore, the rate constants for those two food colorants are much higher than for the other investigated colorants. According to the linearity of the kinetics curves (ln(c/co) v.s. time) it appeared to be first order kinetics for the interval up to 4 days.
