    Higher-group structure in lattice Abelian gauge theory under instanton-sum modification

    We consider the U(1)U(1) gauge theory on a four-dimensional torus, where the instanton number is restricted to an integral multiple of pp. This theory possesses the nontrivial higher-group structure, which can be regarded as a generalization of the Green--Schwarz mechanism, between Zq\mathbb{Z}_q 11-form and Zp\mathbb{Z}_p 33-form symmetries. Here, Zq\mathbb{Z}_q is a subgroup of the center of~U(1)U(1). Following the recent study of the lattice construction of the U(1)/ZqU(1)/\mathbb{Z}_q principal bundle, we examine how such a structure is realized on the basis of lattice regularization.Comment: 16 pages, to appear in EPJ

    Optimization temperature sensitivity using the optically detected magnetic resonance spectrum of a nitrogen-vacancy center ensemble

    Temperature sensing with nitrogen vacancy (NV) centers using quantum techniques is very promising and further development is expected. Recently, the optically detected magnetic resonance (ODMR) spectrum of a high-density ensemble of the NV centers was reproduced with noise parameters [inhomogeneous magnetic field, inhomogeneous strain (electric field) distribution, and homogeneous broadening] of the NV center ensemble. In this study, we use ODMR to estimate the noise parameters of the NV centers in several diamonds. These parameters strongly depend on the spin concentration. This knowledge is then applied to theoretically predict the temperature sensitivity. Using the diffraction-limited volume of 0.1 micron^3, which is the typical limit in confocal microscopy, the optimal sensitivity is estimated to be around 0.76 mK/Hz^(1/2) with an NV center concentration of 5.0e10^17/cm^3. This sensitivity is much higher than previously reported sensitivities, demonstrating the excellent potential of temperature sensing with NV centers.Comment: 17 pages, 4 figures, 1 tabl

    Background: Recent clinical problems in primary care medicine include metabolic syndrome (Met-S) as well as other problems, such as diabetes mellitus, locomotive syndrome (Loc-S) and flailty in middle-age to older people. Regarding exercise therapy for patients with Met-S and Loc-S, aerobic exercise and also resistance (anaerobic) exercise have been considered as necessary. Thus, simple and effective resistance exercise methods should be clarified. Subjects and Methods: The subjects were 61 patients (male 36, female 25) with diabetes mellitus (66.9 ± 13.5 years old, mean ± SD). Subjects were instructed to perform a continuous squat exercises 5 minutes in morning and evening every day for 6 months, using the small exercise equipment ‘HiSquat”. The movements included 1) walking or jogging, 2) inner thigh exercise, 3) outer thigh exercise, 4) straight thigh exercise. Parameters were height, body weight, body mass index (BMI), abdominal circumference, thigh circumference and blood HbA1c value. The treatment of the subjects were not changed during the 6 months. HbA1c value was compared between 0 and 6 months. Results: The fundamental data before the study were as follows: height 159.4 ± 8.4 cm, body weight 62.5 ± 13.8 kg, BMI 24.5 ± 4.7 kg/m2, abdominal circumference 86.8 ± 10.7 cm, thigh circumference 42.9 ± 6.4 cm, HbA1c 6.8 ± 0.9%. HbA1c value at 6 months was 6.4 ± 1.0%, with a statistically significant decrease of 0.4%. As for the correlation analyses, a positive significant correlation was observed between BMI and the thigh/height ratio, and a negative significant correlation was observed between age and thigh/waist ratio. Conclusion: In this study, the HbA1c value was significantly decreased, which suggests the exercise effect of HiSquat. Our study provides fundamental data for the value of waist/thigh circumference in the exercise therapy, and also the efficacy of HiSquat; consequently it suggests the efficacy of HiSquat use for Met-S, Loc-S and flailty, which are increasing concerns in anti-aging medicine of Japan

However, ethnic disparities in takotsubo syndrome (TTS) remain elusive. This study assessed differences in clinical characteristics between Japanese and European TTS patients and determined the impact of ethnicity on in-hospital outcomes. Methods: TTS patients in Japan were enrolled from 10 hospitals and TTS patients in Europe were enrolled from 32 hospitals participating in the International Takotsubo Registry. Clinical characteristics and in-hospital outcomes were compared between Japanese and European patients. Results: A total of 503 Japanese and 1670 European patients were included. Japanese patients were older (72.6 ± 11.4 years vs. 68.0 ± 12.0 years; p < 0.001) and more likely to be male (18.5 vs. 8.4%; p < 0.001) than European TTS patients. Physical triggering factors were more common (45.5 vs. 32.0%; p < 0.001), and emotional triggers less common (17.5 vs. 31.5%; p < 0.001), in Japanese patients than in European patients. Japanese patients were more likely to experience cardiogenic shock during the acute phase (15.5 vs. 9.0%; p < 0.001) and had a higher in-hospital mortality (8.2 vs. 3.2%; p < 0.001). However, ethnicity itself did not appear to have an impact on in-hospital mortality. Machine learning approach revealed that the presence of physical stressors was the most important prognostic factor in both Japanese and European TTS patients. Conclusion: Differences in clinical characteristics and in-hospital outcomes between Japanese and European TTS patients exist. Ethnicity does not impact the outcome in TTS patients. The worse in-hospital outcome in Japanese patients, is mainly driven by the higher prevalence of physical triggers.Open Access funding provided by Universität Zürich.     DOCK2 is involved in the host genetics and biology of severe COVID-19

Here we conducted a genome-wide association study (GWAS) involving 2, 393 cases of COVID-19 in a cohort of Japanese individuals collected during the initial waves of the pandemic, with 3, 289 unaffected controls. We identified a variant on chromosome 5 at 5q35 (rs60200309-A), close to the dedicator of cytokinesis 2 gene (DOCK2), which was associated with severe COVID-19 in patients less than 65 years of age. This risk allele was prevalent in East Asian individuals but rare in Europeans, highlighting the value of genome-wide association studies in non-European populations. RNA-sequencing analysis of 473 bulk peripheral blood samples identified decreased expression of DOCK2 associated with the risk allele in these younger patients. DOCK2 expression was suppressed in patients with severe cases of COVID-19. Single-cell RNA-sequencing analysis (n = 61 individuals) identified cell-type-specific downregulation of DOCK2 and a COVID-19-specific decreasing effect of the risk allele on DOCK2 expression in non-classical monocytes. Immunohistochemistry of lung specimens from patients with severe COVID-19 pneumonia showed suppressed DOCK2 expression. Moreover, inhibition of DOCK2 function with CPYPP increased the severity of pneumonia in a Syrian hamster model of SARS-CoV-2 infection, characterized by weight loss, lung oedema, enhanced viral loads, impaired macrophage recruitment and dysregulated type I interferon responses. We conclude that DOCK2 has an important role in the host immune response to SARS-CoV-2 infection and the development of severe COVID-19, and could be further explored as a potential biomarker and/or therapeutic target

In particular, studies of gene expression dynamics and regulation landscape in COVID-19 infected individuals are limited. Here, we report on a thorough analysis of whole blood RNA-seq data from 465 genotyped samples from the Japan COVID-19 Task Force, including 359 severe and 106 non-severe COVID-19 cases. We discover 1169 putative causal expression quantitative trait loci (eQTLs) including 34 possible colocalizations with biobank fine-mapping results of hematopoietic traits in a Japanese population, 1549 putative causal splice QTLs (sQTLs; e.g. two independent sQTLs at TOR1AIP1), as well as biologically interpretable trans-eQTL examples (e.g., REST and STING1), all fine-mapped at single variant resolution. We perform differential gene expression analysis to elucidate 198 genes with increased expression in severe COVID-19 cases and enriched for innate immune-related functions. Finally, we evaluate the limited but non-zero effect of COVID-19 phenotype on eQTL discovery, and highlight the presence of COVID-19 severity-interaction eQTLs (ieQTLs; e.g., CLEC4C and MYBL2). Our study provides a comprehensive catalog of whole blood regulatory variants in Japanese, as well as a reference for transcriptional landscapes in response to COVID-19 infection

    Atomic level observation of octahedral distortions at the perovskite oxide heterointerface.

    For perovskite oxides, ABO3, slight octahedral distortions have close links to functional properties. While perovskite oxide heterostructures offer a good platform for controlling functionalities, atomistic understanding of octahedral distortion at the interface has been a challenge as it requires precise measurements of the oxygen atomic positions. Here we demonstrate an approach to clarify distortions at an atomic level using annular bright-field imaging in aberration-corrected scanning transmission electron microscopy, which provides precise mappings of cation and oxygen atomic positions from distortion-minimized images. This technique revealed significant distortions of RuO6 and ScO6 octahedra at the heterointerface between a SrRuO3 film and a GdScO3 substrate. We also found that structural mismatch was relieved within only four unit cells near the interface by shifting the oxygen atomic positions to accommodate octahedral tilt angle mismatch. The present results underscore the critical role of the oxygen atom in the octahedral connectivity at the perovskite oxide heterointerface
