The glycoprotein IIb/IIIa antagonist c7E3 inhibits platelet aggregation in the presence of heparin-associated antibodies
AbstractPurpose: Heparin-associated antibodies (HAAb), in the presence of heparin, can cause platelet activation and aggregation. The purpose of this study was to assess whether a platelet glycoprotein (GP) IIb/IIIa receptor antagonist, c7E3, would inhibit platelet aggregation in the presence of HAAb. If aggregation is inhibited by c7E3, enzyme-linked immunosorbent assays (ELISA) would be done to determine whether c7E3 interfered with the binding of heparin and the HAAb.Methods: HAAb-positive plasmas from 21 patients (determined by platelet aggregation assays) were studied. Normal donor platelet-rich plasmas (PRP) were incubated (1 minute) with either saline solution or 3 μg/ml of c7E3. Platelet-poor plasma from patients with HAAb and one of three sources of heparin (25 μl, 10 U/ml; porcine heparin, bovine heparin, and low molecular weight heparin [enoxaparin]) were added to the PRP mixture. Aggregation was determined using a platelet aggregometer by measuring time to aggregation, the slope of the aggregation curve, and the percent change in optical density.Results: Platelet aggregation occured in 100%, 100%, and 95% of the saline solution incubations exposed to porcine heparin, bovine heparin, and enoxaparin, respectively. Incubation with c7E3 caused 100% inhibition of platelet aggregation in plasma exposed to porcine heparin, bovine heparin, and enoxaparin. The optical density curves obtained from the ELISA, which were dependent on the binding of HAAb to heparin, were not significantly different when c7E3 was compared to buffer alone.Conclusions: The GP IIb/IIIa receptor antagonist, c7E3, inhibits HAAb-induced platelet aggregation via a mechanism that does not appear to interfere with the binding between heparin and HAAb. Clinical trials are warranted to assess whether GP IIb/IIIa antagonists may allow patients with HAAb to safely receive heparin. (J Vasc Surg 1997;25:124-30.
Abnormal neurofilament inclusions and segregations in dorsal root ganglia of a Charcot-Marie-Tooth type 2E mouse model
Charcot-Marie-Tooth (CMT) disease or hereditary motor and sensory neuropathy is the most prevalent inherited peripheral neuropathy and is associated with over 90 causative genes. Mutations in neurofilament light polypeptide gene, NEFL cause CMT2E, an axonal form of CMT that results in abnormal structures and/or functions of peripheral axons in spinal cord motor neurons and dorsal root ganglion neurons. We have previously generated and characterized a knock-in mouse model of CMT2E with the N98S mutation in Nefl that presented with multiple inclusions in spinal cord neurons. In this report, we conduct immunofluorescence studies of cultured dorsal root ganglia (DRG) from NeflN98S/+ mice, and show that inclusions found in DRG neurites can occur in embryonic stages. Ultrastructural analyses reveal that the inclusions are disordered neurofilaments packed in high density, segregated from other organelles. Immunochemical studies show decreased NFL protein levels in DRG, cerebellum and spinal cord in NeflN98S/+ mice, and total NFL protein pool is shifted toward the triton-insoluble fraction. Our findings reveal the nature of the inclusions in NeflN98S/+ mice, provide useful information to understand mechanisms of CMT2E disease, and identify DRG from NeflN98S/+ mice as a useful cell line model for therapeutic discoveries

Perceptions and Experiences of Perinatal Mental Disorders in Rural, Predominantly Ethnic Minority Communities in Northern Vietnam
Background: Preliminary research has suggested that perinatal mental disorders (PMDs), including post-partum depression, are prevalent in Vietnam. However the extent to which these disorders are recognized at the community level remains largely undocumented in the literature. PMDs have also never been investigated within Vietnam’s significant ethnic minority populations, who are known to bear a greater burden of maternal and infant health challenges than the ethnic majority. Objective: To investigate knowledge and perceptions of PMDs and their treatments at the community level in a rural, predominantly ethnic minority region of northern Vietnam
From Murder to Imprisonment: Mapping the Flow of Homicide Cases-A Systematic Review
This systematic review examined the evidence on factors influencing the flow of homicide, from suspicious death to imprisonment. Bibliographic databases and thesis portals were searched. The total number of hits was 15,986, of which 15,830 were irrelevant, 35 did not include a quantitative sample, 26 did not focus on homicide, 18 did not present flow data, and for seven there was no full text available. The remaining 70 papers were analyzed. With the exception of one, no study presented a complete longitudinal flow. Results indicated that both legal and extralegal characteristics influence the likelihood of cases to drop out. Aside from a first mapping of homicide case flows, future research should explore false positives and false negatives, to come to a first understanding of funnel selectivity in homicide cases.Security and Global Affair
The illegal use of hand grenades in the Netherlands: 2008-2021
Despite the fact that the issue of illegal hand grenades in the Netherlands received ample public attention, a number of questionssurrounding this phenomenon remain: For what kind of crimes are hand grenades used as weapons? What do we know about the victims and perpetrators of these incidents? Where do the hand grenades come from? Whilst anecdotal reports by the media have addressed some of these questions, there is still a vast scientific lacuna regarding this phenomenon. Therefore, in 2019, we launched the Dutch Hand Grenade Monitor, a database to track all incidents of illegal use of hand grenades in the Netherlands. Combining information from news articles, public trial documents and data fromthe Dutch National Police, we gathered information on the characteristics of these incidents, victims, perpetrators and possible connections to other crimes. Clinical findings and survival in cats naturally infected with feline immunodeficiency virus (FIV)
Background: The clinical course and outcome of natural feline immunodeficiency virus (FIV) infection are variable and incompletely understood. Assigning clinical relevance to FIV infection in individual cats represents a considerable clinical challenge. Objective: To compare signalment, hematologic and biochemical data, major clinical problem and survival between client-owned, FIV-infected and uninfected domestic cats. Animals: Client-owned, domestic cats tested for FIV (n=520). Methods: Retrospective, case control study. Logistic regression analyses were conducted to identify risk factors for FIV infection and to compare hematologic and biochemical data between cases and controls, after adjusting for potential confounders. Survival times were compared using Kaplan-Meier curves. Results: The prevalence of FIV infection was 14.6%. Mixed breed, male sex and older age were risk factors for FIV infection. Hematologic abnormalities, biochemical abnormalities, or both were common in both FIV-infected and uninfected cats. Lymphoid malignancies were slightly more common in FIVinfected than uninfected cats. Survival of FIV-infected cats was not significantly different from that of uninfected cats. Conclusions and clinical importance: Multiple hematologic and biochemical abnormalities are common in old, sick cats regardless of their FIV status. Their presence should not be assumed to indicate clinical progression of FIV infection. A negative effect of FIV on survival was not apparent in this study.
