Molecular Modelling of Oligomeric States of DmOR83b, an Olfactory Receptor in D. Melanogaster

    Get PDF
After the discovery of the complete repertoire of D. melanogaster Olfactory Receptors (ORs), candidate ORs have been identified from at least 12 insect species from four orders (Coleoptera, Lepidoptera, Diptera, and Hymenoptera), including species of economic or medical importance. Although all ORs share the same G-protein coupled receptor structure with seven transmembrane domains, they share poor sequence identity within and between species, and have been identified mainly through genomic data analyses. To date, D. melanogaster remains the only insect species where ORs have been extensively studied, from expression pattern establishment to functional investigations. These studies have confirmed several observations made in vertebrates: one OR type is selectively expressed in a subtype of olfactory receptor neurons, and one olfactory neuron expresses only one type of OR. The olfactory mechanism, further, appears to be conserved between insects and vertebrates. Understanding the function of insect ORs will greatly contribute to the understanding of insect chemical communication mechanisms, particularly with agricultural pests and disease vectors, and could result in future strategies to reduce their negative effects. In this study, we propose molecular models for insect olfactory receptor co-receptor OR83b and its possible functional oligomeric states. The functional similarity of OR83b to GPCRs and ion channels has been exploited for understanding the structure of OR83b. We could observe that C-terminal region (TM4-7) of OR83b is involved in homodimer amd heterodimer formation (with OR22a) which suggests why C-terminus of insect ORs are highly conserved across different species. We also propose two possible ion channel pathways in OR83b: one formed by TM4-5 region with intracellular pore-forming domain and the other formed by TM5-6 with extracellular pore forming domain using analysis of the electrostatics distribution of the pore forming domain

Through many research and discoveries it has been widely accepted that aspect-level sentiment classification is achieved effectively by using Long Short-Term Memory (LSTM) network combined with attention mechanism and memory module. As existing approaches widely depend on the modeling of semantic relatedness of an aspect, at the same time we ignore their syntactic dependencies which are already a part of that sentence. This will result in undesirably an aspect on textual words that are descriptive of other aspects. So, in this paper, to offer syntax free contexts as well as they should be aspect specific, so we propose a proximity-weighted convolution network. To be more precise, we have one way of determining proximity weight which is dependency proximity. The construction of the model includes bidirectional LSTM architecture along with a proximity-weighted convolution neural network

Analytical chemistry plays an increasingly important role in the pharmaceutical industry where, aside from QA, it is used in discovery of new drug candidates and in clinical applications where understanding the interactions between the drug and the patient are critical. Analytical chemistry is defined as the science and art of determining the composition of material in terms of elements or compounds contained in it1 . Analytical chemistry is divided into two branches quantitative and qualitative.. A qualitative method is the information about the identity of atomic or molecular species or functional groups in the sample. A quantitative method provides numerical information as to the relative amount of one or more of these components. In the present study, new RP-HPLC method for simultaneous estimation of Hydrochlorothiazide and Quinapril in bulk and pharmaceutical dosage form was developed. The developed method was validation for various parameters such as accuracy, precision, ruggedness, linearity, robustness, system suitability, specificity, limit of detection and limit of quantitation as per ICH guidelines. The developed method was simple, precise and cost effective for the simultaneous estimation of Hydrochlorothiazide and Quinapril for routine quality control analysis of these either in bulk and pharmaceutical formulation. The developed method was superior when compared to the reported method with less retention time and composition of the mobile phase with good separation

AIM OF THE STUDY: The aim of this invitro study is to compare the fracture resistance of endodontically treated Maxillary First Premolar restored with Lithium disilicate Onlays, Lithium disilicate full crowns and Porcelain fused metal full crowns. MATERIALS AND METHODS: 48 freshly extracted Maxillary First Premolar samples with straight roots were used for this study. The samples were divided into four groups- Group 1: Control (n=12), Group 2: Lithium disilicate Onlays (n=12), Group 3: Lithium disilicate full crowns (n=12) and Group 4: Porcelain fused metal full crowns (n=12). All the samples except control group were subjected to root canal treatment and specimens were prepared according to the groups to receive Lithium disilicate Onlays, full crowns and Porcelain fused metal full crowns. The samples were subjected to thermocycling and fracture resistance for all the samples were tested with Universal testing machine and values recorded in Newtons. RESULTS: There was a statistically significant difference when Porcelain fused metal group (1588.41±593.74 N) was compared with Lithium disilicate full crowns (1030.83 ±513.49 N) and control group (1070.91± 623.34 N). Similarly, a statistically significant difference was found when Lithium disilicate Onlay (1542.50 ±283.22 N) was compared with Lithium disilicate full crown (1030.83 ±513.49 N). (P value 0.05). CONCLUSION: From this study we conclude that Lithium disilicate Onlays are a suitable alternative to the Porcelain fused metal crown for restoration of endodontically treated tooth. However further in vivo studies are needed for determining the fracture resistance of Onlays with different design and luting agents
