    Birth, life and death of a cyclonic eddy in the Southern Ocean

    The ACC is a climatically relevant frontal structure of global importance that regularly develops instabilities which grow into meanders that eventually evolve into long-lived cyclonic eddies. These eddies exhibit sustain primary productivity that can last several months fuelled by local resupply of nutrients. During April-May 2015 we conducted an intensive field experiment in the Southern Ocean (SMILES) where we sampled and tracked an ACC meander as it developed into an eddy and later vanished some 90 days later. The meander and later eddy physical characteristics were observed with a combination of high resolution hydrography, ADCP and turbulence observations in addition to surface and depth resolved biogeochemical observations of nutrients and phytoplankton.     Reversibility of Apixaban Anticoagulation with a Four-Factor Prothrombin Complex Concentrate in Healthy Volunteers.

    It was hypothesized that the four-factor prothrombin complex concentrate (4F-PCC) Kcentra 25 unit/kg would reverse impairment of thrombin generation in healthy volunteers dosed with apixaban to steady state. In this randomized, two-period crossover, assessor-blinded trial, 12 healthy subjects received 5 mg apixaban every 12 h. Three h after the fifth dose, four-factor prothrombin complex concentrate (4F-PCC) 25 unit/kg or saline were infused. Serial blood samples were assessed for thrombin generation using PPP-reagent and PPP-reagent low, anti-Xa, PT, and PTT assays. Geometric mean ratio was calculated at 30 min postinfusion, and at 24, 48, and 72 h. Peak thrombin generation was 76% higher at 30 min postinfusion with 4F-PCC (p = 0.025). The difference declined to 24% at 24 h and resolved by 48 h. Other thrombin generation parameters were also partially normalized. There was no difference between 4F-PCC and saline in anti-Xa assessment at 30 min or later time points

    Fundamental trade-offs between information flow in single cells and cellular populations

    Signal transduction networks allow eukaryotic cells to make decisions based on information about intracellular state and the environment. Biochemical noise significantly diminishes the fidelity of signaling: networks examined to date seem to transmit less than 1 bit of information. It is unclear how networks that control critical cell-fate decisions (e.g., cell division and apoptosis) can function with such low levels of information transfer. Here, we use theory, experiments, and numerical analysis to demonstrate an inherent trade-off between the information transferred in individual cells and the information available to control population-level responses. Noise in receptor-mediated apoptosis reduces information transfer to approximately 1 bit at the single-cell level but allows 3–4 bits of information to be transmitted at the population level. For processes such as eukaryotic chemotaxis, in which single cells are the functional unit, we find high levels of information transmission at a single-cell level. Thus, low levels of information transfer are unlikely to represent a physical limit. Instead, we propose that signaling networks exploit noise at the single-cell level to increase population-level information transfer, allowing extracellular ligands, whose levels are also subject to noise, to incrementally regulate phenotypic changes. This is particularly critical for discrete changes in fate (e.g., life vs. death) for which the key variable is the fraction of cells engaged. Our findings provide a framework for rationalizing the high levels of noise in metazoan signaling networks and have implications for the development of drugs that target these networks in the treatment of cancer and other diseases

    Evaluating the Chinese Revised Controlling Behaviors Scale (C-CBS-R)

    The present study evaluated the utility of the Chinese version of the Revised Controlling Behaviors Scale (C-CBS-R) as a measure of controlling behaviors in violent Chinese intimate relationships. Using a mixed-methods approach, in-depth, individual interviews were conducted with 200 Chinese women survivors to elicit qualitative data about their personal experiences of control in intimate relationships. The use of controlling behaviors was also assessed using the C-CBS-R. Interview accounts suggested that the experiences of 91 of the women were consistent with the description of coercive control according to Dutton and Goodman’s (2005) conceptualization of coercion. Using the split-half validation procedure, a receiver operating characteristics (ROC) curve analysis was conducted with the first half of the sample. The area under the curve (AUC) for using the C-CBS-R to identify high control was .99, and the cutoff score of 1.145 maximized both sensitivity and specificity. Applying the cutoff score to the second half gave a sensitivity of 96% and a specificity of 95%. Overall, the C-CBS-R has demonstrated utility as a measure of controlling behaviors with a cutoff score for distinguishing high from low levels of control in violent Chinese intimate relationships

    Accurate measurement of 5-methylcytosine and 5-hydroxymethylcytosine in human cerebellum DNA by oxidative bisulfite on an array (OxBS-array).

    The Infinium 450K Methylation array is an established tool for measuring methylation. However, the bisulfite (BS) reaction commonly used with the 450K array cannot distinguish between 5-methylcytosine (5mC) and 5-hydroxymethylcytosine (5hmC). The oxidative-bisulfite assay disambiguates 5mC and 5hmC. We describe the use of oxBS in conjunction with the 450K array (oxBS-array) to analyse 5hmC/5mC in cerebellum DNA. The "methylation" level derived by the BS reaction is the combined level of 5mC and 5hmC at a given base, while the oxBS reaction gives the level of 5mC alone. The level of 5hmC is derived by subtracting the oxBS level from the BS level. Here we present an analysis method that distinguishes genuine positive levels of 5hmC at levels as low as 3%. We performed four replicates of the same sample of cerebellum and found a high level of reproducibility (average r for BS = 98.3, and average r for oxBS = 96.8). In total, 114,734 probes showed a significant positive measurement for 5hmC. The range at which we were able to distinguish 5hmC occupancy was between 3% and 42%. In order to investigate the effects of multiple replicates on 5hmC detection we also simulated fewer replicates and found that decreasing the number of replicates to two reduced the number of positive probes identified by > 50%. We validated our results using qPCR in conjunction with glucosylation of 5hmC sites followed by MspI digestion and we found good concordance with the array estimates (r = 0.94). This experiment provides a map of 5hmC in the cerebellum and a robust dataset for use as a standard in future 5hmC analyses. We also provide a novel method for validating the presence of 5hmC at low levels, and highlight some of the pitfalls associated with measuring 5hmC and 5mC.S.     The effects of magnetic field on the d-density wave order in the cuprates

    We consider the effects of a perpendicular magnetic field on the d-density wave order and conclude that if the pseudogap phase in the cuprates is due to this order, then it is highly insensitive to the magnetic field in the underdoped regime, while its sensitivity increases as the gap vanishes in the overdoped regime. This appears to be consistent with the available experiments and can be tested further in neutron scattering experiments.     Advances in Thick GEM-like gaseous electron multipliers. Part I: atmospheric pressure operation

    Thick GEM-like (THGEM) gaseous electron multipliers are made of standard printed-circuit board perforated with sub-millimeter diameter holes, etched at their rims. Effective gas multiplication factors of 100000 and 10000000 and fast pulses in the few nanosecond rise-time scale were reached in single- and cascaded double-THGEM elements, in atmospheric-pressure standard gas mixtures with single photoelectrons. High single-electron detection efficiency is obtained in photon detectors combining THGEMs and semitransparent UV-sensitive CsI photocathodes or reflective ones deposited on the top THGEM face; the latter benefits of a reduced sensitivity to ionizing background radiation. Stable operation was recorded with photoelectron fluxes exceeding MHz/mm2.
