Karyotype differentiation of four Cestrum species (Solanaceae) revealed by fluorescent chromosome banding and FISH

The karyotypes of four South American species of Cestrum (C. capsulare,C. corymbosum,C. laevigatum and C. megalophylum) were studied using conventional staining, C-CMA/DAPI chromosome banding and FISH with 45S and 5S rDNA probes. The karyotypes showed a chromosome number of 2n = 2x = 16, with metacentric chromosomes, except for the eighth submeta- to acrocentric pair. Several types of heterochromatin were detected, which varied in size, number, distribution and base composition. The C-CMA+ bands and 45S rDNA were located predominantly in terminal regions. The C-CMA + /DAPI + bands appeared in interstitial and terminal regions, and the C-DAPI + bands were found in all chromosome regions. The 5S rDNA sites were observed on the long arm of pair 8 in all species except C. capsulare, where they were found in the paracentromeric region of the long arm of pair 4. The differences in band patterns among the species studied here, along with data from other nine species reported in the literature, suggest that the bands are dispersed in an equilocal and non-equilocal manner and that structural rearrangements can be responsible for internal karyotype diversification. However, it is important to point out that the structural changes involving repetitive segments did not culminate in substantial changes in the general karyotype structure concerning chromosome size and morphology

Simultaneous transcriptional profiling of Leishmania major and its murine macrophage host cell reveals insights into host-pathogen interactions

Parasites of the genus Leishmania are the causative agents of leishmaniasis, a group of diseases that range in manifestations from skin lesions to fatal visceral disease. The life cycle of Leishmania parasites is split between its insect vector and its mammalian host, where it resides primarily inside of macrophages. Once intracellular, Leishmania parasites must evade or deactivate the host's innate and adaptive immune responses in order to survive and replicate. We performed transcriptome profiling using RNA-seq to simultaneously identify global changes in murine macrophage and L. major gene expression as the parasite entered and persisted within murine macrophages during the first 72 h of an infection. Differential gene expression, pathway, and gene ontology analyses enabled us to identify modulations in host and parasite responses during an infection. The most substantial and dynamic gene expression responses by both macrophage and parasite were observed during early infection. Murine genes related to both pro- and anti-inflammatory immune responses and glycolysis were substantially upregulated and genes related to lipid metabolism, biogenesis, and Fc gamma receptor-mediated phagocytosis were downregulated. Upregulated parasite genes included those aimed at mitigating the effects of an oxidative response by the host immune system while downregulated genes were related to translation, cell signaling, fatty acid biosynthesis, and flagellum structure. The gene expression patterns identified in this work yield signatures that characterize multiple developmental stages of L. major parasites and the coordinated response of Leishmania-infected macrophages in the real-time setting of a dual biological system. This comprehensive dataset offers a clearer and more sensitive picture of the interplay between host and parasite during intracellular infection, providing additional insights into how pathogens are able to evade host defenses and modulate the biological functions of the cell in order to survive in the mammalian environment.https://doi.org/10.1186/s12864-015-2237-

Na Nota de Conjuntura desse mês abrimos espaço para uma discussão cada vez mais presente no setor leiteiro nacional: como desenvolver a cadeia produtiva do leite com vistas a torna-la um importante exportador mundial de lácteos. O Brasil tem se consolidado como um importador líquido de lácteos. Todavia, o país tem condições excepcionais de produção de leite. Assim, à luz da teoria econômica quais variáveis podem ser manuseadas tendo a indústria como agente para o desenvolvimento da cadeia de lácteos brasileira? Pelo lado da demanda, a indústria pode trabalhar a questão do preço e também o hábito alimentar. Já pelo lado da oferta deve atentar-se para o custo dos fatores de produção, a tecnologia e os objetivos corporativos. Nesse contexto, um trabalho integrado entre as industrias com vistas a uma marca brasileira, como o Goodairy Brazil são de grande importância. Integração das atividades de vacinação pelo BCG intradérmico nos programas gerais de imunização das unidades sanitárias Integration of intradermic BCG vaccination in general public health immunization programmes

A imunização pelo BCG constitui um recurso operacional de grande alcance nos países em desenvolvimento, onde é elevada a incidência da tuberculose. Os conhecimentos adquiridos nos últimos cinco anos, em nosso meio, permitem elaborar programas para a aplicação do BCG, por via intradérmica, em extensão útil epidemiologicamente, sem o teste tuberculínico prévio. É analisada a experiência da integração da vacinação pelo BCG intradérmico no programa geral de imunização do Centro de Saúde "Geraldo de Paula Souza", da Faculdade de Saúde Pública da USP. As suas conclusões são favoráveis à viabilidade do método, permitindo ampla cobertura de imunização anti-tuberculose da população, desde os primeiros meses de vida.<br>Immunization by BCG is of great effect in developing countries where the incidence of tuberculosis is high. The knowledge that has been acquired in our environment in the last five years has made possible the organization of programmes for BCG intradermic immunization, to a useful epidemiological extent, without a prior tuberculin test. The practice of the integration of intradermic BCG vaccination in the general immunization programmes of the "Geraldo de Paula Souza" Health Unit of S. Paulo University School of Public Health was analysed and the conclusion was favourable as to the viability of the method, which allows a large-scale anti-tuberculosis immunization of the population right from the very first months of life

Nota de Conjuntura: Leite e Derivados - Maio de 2019.

A renda per capita está estagnada inibindo o consumo geral das famílias e, em particular, o de lácteos. Vendas menores no varejo pressionam os preços para baixo inibindo as margens e a expansão de toda a cadeia produtiva. Os preços ao produtor tiveram uma trajetória ascendente iniciada em fevereiro, mas já em abril, as cotações ao produtor ficaram praticamente estagnadas. No caso da indústria, a situação é bem mais complicada. Os laticínios não conseguem reajustar os preços, sobretudo de produtos tradicionais, comprometendo a rentabilidade de diversas empresas que atuam nesses mercados. Um outro complicador é que Argentina e Uruguai devem ter sua disponibilidade recomposta até meados do ano, gerando excedentes que podem pressionar as exportações para o Brasil. Isso cria uma grande incerteza para o segundo semestre, baseada em pelo menos quatro motivos: 1) a relação entre os preços do leite e dos insumos está favorável ao produtor e tende a estimular a oferta; 2) o baixo consumo de lácteos decorrente do fraco desempenho da economia; 3) baixa competitividade para exportar excedentes de produção; 4) preços de importação mais competitivos que os domésticos.
