    Stochastic Inflation and Replica Field Theory

    We adopt methods from statistical field theory to stochastic inflation. For the example of a free test field in de Sitter and power-law inflation, the power spectrum of long-wavelength fluctuations is computed. We study its dependence on the shape of the filter that separates long from short wavelength modes. While for filters with infinite support the phenomenon of dimensional reductions is found on large super-horizon scales, filters with compact support return a scale-invariant power spectrum in the infra-red. Features of the power spectrum, induced by the filter, decay within a few e-foldings. Thus the late-time power spectrum is independent of the filter details.Comment: 15 pages, 14 figure

    The IcsA (intracellular spread gene A) autotransporter from Shigella flexneri is a key virulence factor. We identified a stable fragment comprising residues 591 to 758, which corresponds to the autochaperone region of the IcsA passenger domain. We showed that thermal unfolding of the autochaperone region is reversible and determined its crystal structure at 2.0-Å resolution

    Crystal structure of the Escherichia coli RNA degradosome component enolase.

    The crystal structure of Escherichia coli enolase (EC 4.2.1.11, phosphopyruvate hydratase), which is a component of the RNA degradosome, has been determined at 2.5 Å. There are four molecules in the asymmetric unit of the C2 cell, and in one of the molecules, flexible loops close onto the active site. This closure mimics the conformation of the substrate-bound intermediate. A comparison of the structure of the E. coli enolase with the eukaryotic enolase structures available (lobster and yeast) indicates a high degree of conservation of the hydrophobic core and the subunit interface of this homodimeric enzyme. The dimer interface is enriched in charged residues compared with other protein homodimers, which may explain our observations from analytical ultracentrifugation that dimerisation is affected by ionic strength. The putative role of enolase in the RNA degradosome is discussed; although it was not possible to ascribe a specific role to it, a structural role is possible.http://dx.doi.org

    In mitotic cells, establishment of sister chromatid cohesion requires acetylation of the cohesin subunit SMC3 (acSMC3) by ESCO1 and/or ESCO2. Meiotic cohesin plays additional but poorly understood roles in the formation of chromosome axial elements (AEs) and synaptonemal complexes. Here, we show that levels of ESCO2, acSMC3, and the pro-cohesion factor sororin increase on meiotic chromosomes as homologs synapse. These proteins are less abundant on the largely unsynapsed sex chromosomes, whose sister chromatid cohesion appears weaker throughout the meiotic prophase. Using three distinct conditional Esco2 knockout mouse strains, we demonstrate that ESCO2 is essential for male gametogenesis. Partial depletion of ESCO2 in prophase I spermatocytes delays chromosome synapsis and further weakens cohesion along sex chromosomes, which show extensive separation of AEs into single chromatids. Unsynapsed regions of autosomes are associated with the sex chromatin and also display split AEs. This study provides the first evidence for a specific role of ESCO2 in mammalian meiosis, identifies a particular ESCO2 dependence of sex chromosome cohesion and suggests support of autosomal synapsis by acSMC3-stabilized cohesion

    Low Power Measurements on a Finger Drift Tube Linac

    The efficiency of RFQs decreases at particle energies higher than a few MeV/u and thus typically DTL structures are used in this energy region. However, the rf field in the gap always has a defocusing influence on the beam. In order to compensate this effect, fingers with quadrupole symmetry were added to the drift tubes, the focusing fingers do not need an additional power source or feed through. Beam dynamics have been studied with the code RFQSIM. Detailed analysis of the field distribution was done and the geometry of the finger array has been optimized with respect to beam dynamics. A prototype cavity with finger drift tubes was built and low power measurements were done. In this contribution, the results of the rf simulation with Microwave Studio are compared to bead perturbation measurements and the focusing effect on the beam is discussed

    Topological Modes in Dual Lattice Models

    Lattice gauge theory with gauge group ZPZ_{P} is reconsidered in four dimensions on a simplicial complex KK. One finds that the dual theory, formulated on the dual block complex K^\hat{K}, contains topological modes which are in correspondence with the cohomology group H2(K^,ZP)H^{2}(\hat{K},Z_{P}), in addition to the usual dynamical link variables. This is a general phenomenon in all models with single plaquette based actions; the action of the dual theory becomes twisted with a field representing the above cohomology class. A similar observation is made about the dual version of the three dimensional Ising model. The importance of distinct topological sectors is confirmed numerically in the two dimensional Ising model where they are parameterized by H1(K^,Z2)H^{1}(\hat{K},Z_{2}).Comment: 10 pages, DIAS 94-3

    Design Studies of an Electrostatic Storage Ring

    Electrostatic storage rings combine a number of very interesting characteristics that make them an attractive tool in the low energy range. In contrast to magnetic rings, all of the fields in an electrostatic storage ring are completely mass independent. At the same particle energy and charge state, ions from light protons to heavy biomolecules can in principal be stored with identical field setups. A small ring for ions of energies up to 50 keV is planned to be built up at Goethe University in Frankfurt. Different designs have been calculated and the results are presented in this contribution. Furthermore, prototypes of the necessary optical elements have been manufactured and are described as well

    Utility of plasma neurofilament light and total tau for clinical trials in Alzheimer's disease

    INTRODUCTION: Several blood‐based biomarkers are associated with neuronal injury, but their utility in interventional clinical trials is unclear. This study retrospectively evaluated the utility of plasma neurofilament light (NfL) and total tau (t‐tau) in an 18‐month trial in mild Alzheimer's disease (AD). METHODS: Correlation and conditional independence analyses and Gaussian graphical models were used to investigate cross‐sectional and longitudinal relations between NfL, t‐tau, and clinical scales. RESULTS: NfL had a stronger association than t‐tau with clinical scales; t‐tau did not hold additional information to that given by NfL (P > 0.05 at all time points). NfL held independent information about shorter‐term (3‐ to 6‐month) progression beyond patient age and clinical scores. However, no meaningful gain in power was found when adjusting a longitudinal analysis of cognitive scores for baseline NfL. DISCUSSION: Plasma NfL is superior to t‐tau in mild AD. The ability of NfL to detect changes before clinical manifestations makes it a promising biomarker of drug response in trials of disease‐modifying drugs

    Testing nanomaterial toxicity in unicellular eukaryotic algae and fish cell lines.

    Nanoecotoxicology as a sub-discipline of ecotoxicology aims to identify and predict effects elicited on ecosystems by nano-sized materials (NM). Two key groups of model organisms in this context are algae and fi sh. In this chapter, we present considerations for testing NM with respect to their impact on unicellular algae and cell lines derived from various organs of fi sh. Based on currently available literature on NM effects in unicellular algae and fi sh cell lines, and our own experience, we provide guidance on test design, including principle test considerations, materials, NM presentation to cells, exposure, bioavailability, and effect assessment. Assessment needs to be based on a meaningful choice of exposure scenario(s) related to the research question. As a fi rst step, one needs to address whether effects of NMs are to be investigated under environmentally relevant or probable conditions, which may include processes such as agglomeration, or whether NM effects from mono-dispersed particles are of interest, which may require special steps to ensure stable NM suspension. Moreover, whether effects on cells are to be studied in the short- or long-term is important with regard to experimental design. Preparation of NM suspensions, which can be done in aqueous media different from the exposure medium, is addressed with regard to energy input, sterility (as required for algae and fi sh cell exposure) and particle purity. Specifi ed for the two model systems, algae and fi sh cell lines, availability and choice of culture media are presented and discussed with regard to impact on NM behavior. Light, temperature, and agitation, which are variables during exposure, are discussed. We further provide guidance on the characterization of the NM in the chosen aqueous exposure media regarding size, zeta potential and electrophoretic mobility. The state of NM in exposure media is decisive for their bioavailability and therefore for potential particle effects. Therefore, we present ways of deriving a mass balance and quantitative/qualitative information on the uptake and distribution of NM in cells. As NM have a high surface-to-volume ratio and possess specifi c physical-chemical properties, which make them prone to interfere with various compounds and certain types of toxicity tests, potential interferences and appropriate controls are introduced. Furthermore, different types of dose metrics, which is still a strongly debated issue in nanotoxicology, are highlighted. We also consider laboratory safety regarding NM handling and disposal
