    The distribution of Bibionidae (Diptera) in Scotland

    We review the material of the family Bibionidae from Scotland in the Natural History Museum, London and the National Museums of Scotland, Edinburgh, as well as some material from other collections and published records from the SIRI (Scottish Insect Records Index). Eleven species of the genus Bibio Geoffroy, 1762 and four species in the genus Dilophus Meigen, 1803 have been collected in Scotland. We review the distribution and ecology of each species and present maps of records. Previous Scottish records of Bibio hortulanus (Linnaeus, 1758) and B. reticulatus Loew, 1846 are deemed to be most likely erroneous. Additionally, we have not found any specimens confirming the literature record of Bibio venosus (Meigen, 1804), but we believe this record is most likely correct since this species is distinctive and well defined with no substantial confusion about its identity. There is also a recent record of Dilophus humeralis Zetterstedt, 1850 which we have not been able to confirm. The following species have unambiguous records from Scotland: Bibio clavipes Meigen, 1818; B. ferruginatus (Linnaeus, 1758); B. johannis (Linnaeus, 1767); B. lanigerus Meigen, 1818; B. leucopterus (Meigen, 1804); B. longipes Loew, 1864; B. marci (Linnaeus, 1758); B. nigriventris Haliday, 1833; B. pomonae (Fabricius, 1775); B. varipes Meigen, 1830; Dilophus bispinosus Lundström, 1913; D. febrilis (Linnaeus, 1758); D. femoratus Meigen, 1804

    Morbidity and mortality in reptiles presented to a wildlife care facility in Central Illinois

    We examined morbidity and mortality of 200 reptiles, representing 13 different species that were presented to the University of Illinois Wildlife Medical Clinic (WMC) from 2003 to 2010. Snapping turtles (Chelydra serpentine; n = 46), box turtles (Terrapene sp.; n = 43), painted turtles (Chrysemys picta; n = 37), and red-eared slider turtles (Trachemys scripta elegans; n = 33) were the most frequently seen species. Turtles were significantly more likely to be presented to the WMC following collision with a motor vehicle (n = 73) than any other reason, including idiopathic trauma (i.e., trauma of unknown origin; n = 25) or infectious disease (n =18). The findings from this cross-sectional study suggest a potential for community education in limiting reptile traumas resulting in presentations to a wildlife hospital

    Comparative study of the commonly used virulence tests for laboratory diagnosis of ovine footrot caused by Dichelobacter nodosus in Australia.

    Footrot in sheep and goats is expressed as a spectrum of clinical entities ranging from benign, which is a self limiting interdigital dermatitis to highly virulent, in which severe under running of the horn of the hoof occurs. Interactions between the host, the virulence of the causative strain of Dichelobacter nodosus and environmental conditions determine the severity of the disease. Clinical diagnosis of virulent footrot, which a notifiable disease in some states of Australia, is not always straightforward. Therefore, the Gelatin Gel and Elastase tests for protease activity, and the intA PCR test for an inserted genetic element in D. nodosus are commonly used to support or to confirm a clinical diagnosis. A comparative study of these laboratory tests with a large number of samples collected from 12 flocks of sheep with clinically virulent footrot was conducted. Based on the elastase test, 64% of the isolates tested were classified as virulent compared to 91% on the gelatin gel test and 41% according to the intA test. The agreement between the elastase and the gelatin gel test was low (kappa =0.12) as were the agreements between other tests. Only about 21% of the isolates were virulent in all 3 tests. Therefore these tests on their own may not provide standard and reliable results and are likely to remain as supplementary tests for clinical diagnosis of the disease.     Cost-effective recruitment strategies that attract underrepresented minority undergraduates who persist to STEM doctorates

    The paucity of underrepresented minorities (URMs) earning science, technology, engineering, and mathematics (STEM) degrees remains an issue in revitalizing the U.S. biomedical workforce. Due to reductions in federal funding, maintaining the integrity of programs that focus on URM retention and recruitment is crucial. We present data on the mechanisms used to recruit URM students to our program (e.g., email, events, referrals, website), which individually were equally effective in attracting applicants to the program. Recruitment mechanisms were grouped and further classified relative to their cost to implement as lower and higher cost. Our results indicate that lower cost mechanisms, statistically, were as effective as higher cost mechanisms in recruiting students who persisted to PhD programs. Using a binary logistic regression model to predict PhD matriculation, higher cost mechanisms were not significant predictors of PhD matriculation. Collectively, these data demonstrate for the first time that lower cost mechanisms can be as successful in recruiting URM students to summer programs who pursue PhDs in STEM fields. </jats:p

    Estimation of sensitivity and flock-sensitivity of pooled faecal culture for Mycobacterium avium subsp. paratuberculosis in sheep

    Pooled faecal culture (PFC) is a widely used test in ovine Johne’s disease (OJD) control programmes in Australia but information about its characteristics is limited. We conducted this study to estimate sensitivity and flock-sensitivity of PFC in sheep with different OJD histopathological lesions in simulated flocks with a range of infection prevalence levels. Initially, a known quantity of faeces from sheep with pauci- or multi-bacillary lesions was pooled with uncontaminated faeces from confirmed non-infected sheep and cultured using PFC technique. PFC sensitivity, calculated as a proportion of the pools of a particular size that tested positive, was determined to be 90% in sheep with the multibacillary form of the disease but varied with pool size in sheep with the paucibacillary form of OJD. Subsequently, probabilistic models were developed to estimate overall pool-sensitivity achieved in a flock (Sek) and flock-sensitivity of PFC (FSe) in various simulated scenarios. In flocks with a given ratio of multi- to pauci-bacillary sheep and with low to moderate infection prevalence level, Sek decreased with increase in pool size, but increased with pool size in flocks with ≥ 10 % prevalence. FSe, in contrast, increased with pool size in all the tested scenarios. Both Sek and FSe increased with infection prevalence, ratio of multi- to pauci-bacillary sheep and the number of pools sourced from flocks. Sensitivity analyses indicated that the estimates and trends were robust to moderate changes in input parameters. The results suggest that the current testing of seven pools of size 50 is adequate for most scenarios, however, for very low prevalence flocks, a gain in FSe can be made by increasing the number of pools tested and a higher Sek can be achieved by reducing pool sizes.     TATA-Binding Protein Recognition and Bending of a Consensus Promoter Are Protein Species Dependent

    The structure and behavior of full-length human TBP binding the adenovirus major late promoter (AdMLP) have been characterized using biophysical methods. The human protein induces a 97° bend in DNAAdMLP. The high-resolution functional data provide a quantitative energetic and kinetic description of the partial reaction sequence as native human TBP binds rapidly to a consensus promoter with high affinity. The reaction proceeds with successive formation of three bound species, all having strongly bent DNA, with the concurrence of binding and bending demonstrated by both fluorescence and anisotropy stopped flow. These results establish the protein species dependence of the TBP−DNAAdMLP structure and recognition mechanism. Additionally, the strong correlation between the DNA bend angle and transcription efficiency demonstrated previously for yeast TBP is shown to extend to human TBP. The heterologous NH2-terminal domains are the apparent source of the species-specific differences. Together with previous studies the present work establishes that TBPwt−DNATATA function and structure depend both on the TATA box sequence and on the TBP species

    Self-avoiding walks crossing a square

    We study a restricted class of self-avoiding walks (SAW) which start at the origin (0, 0), end at (L,L)(L, L), and are entirely contained in the square [0,L]×[0,L][0, L] \times [0, L] on the square lattice Z2{\mathbb Z}^2. The number of distinct walks is known to grow as λL2+o(L2)\lambda^{L^2+o(L^2)}. We estimate λ=1.744550±0.000005\lambda = 1.744550 \pm 0.000005 as well as obtaining strict upper and lower bounds, 1.628<λ<1.782.1.628 < \lambda < 1.782. We give exact results for the number of SAW of length 2L+2K2L + 2K for K=0,1,2K = 0, 1, 2 and asymptotic results for K=o(L1/3)K = o(L^{1/3}). We also consider the model in which a weight or {\em fugacity} xx is associated with each step of the walk. This gives rise to a canonical model of a phase transition. For x<1/μx < 1/\mu the average length of a SAW grows as LL, while for x>1/μx > 1/\mu it grows as L2L^2. Here μ\mu is the growth constant of unconstrained SAW in Z2{\mathbb Z}^2. For x=1/μx = 1/\mu we provide numerical evidence, but no proof, that the average walk length grows as L4/3L^{4/3}. We also consider Hamiltonian walks under the same restriction. They are known to grow as τL2+o(L2)\tau^{L^2+o(L^2)} on the same L×LL \times L lattice.     On the Dominance of Trivial Knots among SAPs on a Cubic Lattice

    The knotting probability is defined by the probability with which an NN-step self-avoiding polygon (SAP) with a fixed type of knot appears in the configuration space. We evaluate these probabilities for some knot types on a simple cubic lattice. For the trivial knot, we find that the knotting probability decays much slower for the SAP on the cubic lattice than for continuum models of the SAP as a function of NN. In particular the characteristic length of the trivial knot that corresponds to a `half-life' of the knotting probability is estimated to be 2.5×1052.5 \times 10^5 on the cubic lattice.Comment: LaTeX2e, 21 pages, 8 figur
