    Inhibition of HLA-DM Mediated MHC Class II Peptide Loading by HLA-DO Promotes Self Tolerance

    Major histocompatibility class II (MHCII) molecules are loaded with peptides derived from foreign and self-proteins within the endosomes and lysosomes of antigen presenting cells (APCs). This process is mediated by interaction of MHCII with the conserved, nonpolymorphic MHCII-like molecule HLA-DM (DM). DM activity is directly opposed by HLA-DO (DO), another conserved, non-polymorphic MHCII like molecule. DO is an MHCII substrate mimic. Binding of DO to DM prevents MHCII from binding to DM, thereby inhibiting peptide loading. Inhibition of DM function enables low stability MHC complexes to survive and populate the surface of APCS. As a consequence, DO promotes the display of a broader pool of low abundance self-peptides. Broadening the peptide repertoire theoretically reduces the likelihood of inadvertently acquiring a density of self-ligands that is sufficient to activate self-reactive T cells. One function of DO, therefore, is to promote T cell tolerance by shaping the visible image of self. Recent data also shows that DO influences the adaptive immune response by controlling B cell entry into the germinal center reaction. This review explores the data supporting these concepts

    Researchers employ triangulation to increase the validity of inference in qualitative and quantitative research. Leuffen, Shikano, and Walter have presented guidance as to which strategies to use when triangulating data sources. In this article, I explore how their findings can be translated for practical research purposes. I offer an illustrative application concentrating on the political power of traditional political authorities in Uganda and Tanzania. I analyze the status quo of political power and the preferred political power of traditional leaders. To triangulate, I use three sources: (1) constitutional-legal texts, (2) the Afrobarometer survey, and (3) in-depth interviews. I shed light on possible problems and analytical strategies for triangulation in practice, with a specific focus on convergence and divergence of sources

    Talk the talk, walk the walk: Defining Critical Race Theory in research

    Over the last decade there has been a noticeable growth in published works citing Critical Race Theory (CRT). This has led to a growth in interest in the UK of practical research projects utilising CRT as their framework. It is clear that research on 'race' is an emerging topic of study. What is less visible is a debate on how CRT is positioned in relation to methodic practice, substantive theory and epistemological underpinnings. The efficacy of categories of data gathering tools, both traditional and non-traditional is a discussion point here to explore the complexities underpinning decisions to advocate a CRT framework. Notwithstanding intersectional issues, a CRT methodology is recognisable by how philosophical, political and ethical questions are established and maintained in relation to racialised problematics. This paper examines these tensions in establishing CRT methodologies and explores some of the essential criteria for researchers to consider in utilising a CRT framework. © 2012 Copyright Taylor and Francis Group, LLC

    HACIENDO [AUTO] ETNOGRAFIA POLITICAMENTE

    Building on earlier arguments, I propose a civic, publicly responsable [auto] ethnography that addresses the central issues of self, race, gender, class, society, and democracy.  I begin with pedagogies of hope and the sociological and ethnographic imaginations.  I turn then to the ethnographer and cultural studies, reviewing several models of critical ethnography. I next examine critical performance pedagogy, politics, and critical race theory, and conclude with a brief discussion of the practices of a performative cultural politics.Basado en argumentos previos, propongo una [auto] etnografía civil, públicamente responsable, que aborde las temáticas centrales de self, raza, género, clase, sociedad y democracia. Comienzo con la pedagogía de la esperanza y la imaginación sociológica y etnográfica.  Paso entonces al etnógrafo y los estudios culturales, revisando varios modelos de etnografía crítica.     The Reframing of Methodology: Revisiting a PhD Study

    The paper draws on a PhD study to explore some methodological dilemmas associated with the execution of qualitative research when framed within positivist study design. The PhD was linked to an externally funded research project which evaluated the implementation of a custody-based intervention in the secure estate. While the PhD was conceived as a qualitative study, informed by interpretivist methodology and associated epistemology, the wider funded study was informed by positivist tradition and used a quantitative method. This led to dilemmas of both practical and methodological nature. The author revisits her study's methodological position to review issues raised by the research design and suggests an alternative proposal informed methodologically by critical realism which may better serve the study's interests. In doing so, the paper suggests how revisiting previous research may assist us in gaining methodological understanding and allow us to reframe our future endeavours to more useful end
