    Assignment of the NV0 575 nm zero-phonon line in diamond to a 2E-2A2 transition

    The time-averaged emission spectrum of single nitrogen-vacancy defects in diamond gives zero-phonon lines of both the negative charge state at 637 nm (1.945 eV) and the neutral charge state at 575 nm (2.156 eV). This occurs through photo-conversion between the two charge states. Due to strain in the diamond the zero-phonon lines are split and it is found that the splitting and polarization of the two zero-phonon lines are the same. From this observation and consideration of the electronic structure of the nitrogen-vacancy center it is concluded that the excited state of the neutral center has A2 orbital symmetry. The assignment of the 575 nm transition to a 2E - 2A2 transition has not been established previously.Comment: 5 pages, 5 figure

    PrimPol is required for replicative tolerance of G quadruplexes in vertebrate cells

    G quadruplexes (G4s) can present potent blocks to DNA replication. Accurate and timely replication of G4s in vertebrates requires multiple specialized DNA helicases and polymerases to prevent genetic and epigenetic instability. Here we report that PrimPol, a recently described primase-polymerase (PrimPol), plays a crucial role in the bypass of leading strand G4 structures. While PrimPol is unable to directly replicate G4s, it can bind and reprime downstream of these structures. Disruption of either the catalytic activity or zinc-finger of PrimPol results in extreme G4-dependent epigenetic instability at the BU-1 locus in avian DT40 cells, indicative of extensive uncoupling of the replicative helicase and polymerase. Together, these observations implicate PrimPol in promoting restart of DNA synthesis downstream of, but closely coupled to, G4 replication impediments

    Unified Framework for Correlations in Terms of Local Quantum Observables

    We provide a unified framework for nonsignalling quantum and classical multipartite correlations, allowing all to be written as the trace of some local (quantum) measurements multiplied by an operator. The properties of this operator define the corresponding set of correlations.We then show that if the theory is such that all local quantum measurements are possible, one obtains the correlations corresponding to the extension of Gleason's Theorem to multipartite systems. Such correlations coincide with the quantum ones for one and two parties, but we prove the existence of a gap for three or more parties.Comment: 4 pages, final versio

    Nanomechanical squeezing with detection via a microwave cavity

    We study a parametrically driven nanomechanical resonator capacitively coupled to a microwave cavity. If the nanoresonator can be cooled to near its quantum ground state then quantum squeezing of a quadrature of the nanoresonator motion becomes feasible. We consider the adiabatic limit in which the cavity mode is slaved to the nanoresonator mode. By driving the cavity on its red-detuned sideband, the squeezing can be coupled into the microwave field at the cavity resonance. The red-detuned sideband drive is also compatible with the goal of ground state cooling. Squeezing of the output microwave field may be inferred using a technique similar to that used to infer squeezing of the field produced by a Josephson parametric amplifier, and subsequently, squeezing of the nanoresonator motion may be inferred. We have calculated the output field microwave squeezing spectra and related this to squeezing of the nanoresonator motion, both at zero and finite temperature. Driving the cavity on the blue-detuned sideband, and on both the blue and red sidebands, have also been considered within the same formalism

    Galaxy protocluster candidates around z ~ 2.4 radio galaxies

    We study the environments of 6 radio galaxies at 2.2 < z < 2.6 using wide-field near-infrared images. We use colour cuts to identify galaxies in this redshift range, and find that three of the radio galaxies are surrounded by significant surface overdensities of such galaxies. The excess galaxies that comprise these overdensities are strongly clustered, suggesting they are physically associated. The colour distribution of the galaxies responsible for the overdensity are consistent with those of galaxies that lie within a narrow redshift range at z ~ 2.4. Thus the excess galaxies are consistent with being companions of the radio galaxies. The overdensities have estimated masses in excess of 10^14 solar masses, and are dense enough to collapse into virizalised structures by the present day: these structures may evolve into groups or clusters of galaxies. A flux-limited sample of protocluster galaxies with K < 20.6 mag is derived by statistically subtracting the fore- and background galaxies. The colour distribution of the protocluster galaxies is bimodal, consisting of a dominant blue sequence, comprising 77 +/- 10% of the galaxies, and a poorly populated red sequence. The blue protocluster galaxies have similar colours to local star-forming irregular galaxies (U -V ~ 0.6), suggesting most protocluster galaxies are still forming stars at the observed epoch. The blue colours and lack of a dominant protocluster red sequence implies that these cluster galaxies form the bulk of their stars at z < 3.Comment: Accepted for publication in MNRA

    Altered expression of caspases-4 and -5 during inflammatory bowel disease and colorectal cancer : diagnostic and therapeutic potential

    Caspases are a group of proteolytic enzymes involved in the co-ordination of cellular processes, including cellular homeostasis, inflammation and apoptosis. Altered activity of caspases, particularly caspase-1, has been implicated in the development of intestinal diseases, such as inflammatory bowel disease (IBD) and colorectal cancer (CRC). However, the involvement of two related inflammatory caspase members, caspases-4 and -5, during intestinal homeostasis and disease has not yet been established. This study demonstrates that caspases-4 and -5 are involved in IBD-associated intestinal inflammation. Furthermore, we found a clear correlation between stromal caspase-4 and -5 expression levels, inflammation and disease activity in ulcerative colitis patients. Deregulated intestinal inflammation in IBD patients is associated with an increased risk of developing CRC. We found robust expression of caspases-4 and -5 within intestinal epithelial cells, exclusively within neoplastic tissue, of colorectal tumours. An examination of adjacent normal, inflamed and tumour tissue from patients with colitis-associated CRC confirmed that stromal expression of caspases-4 and -5 is increased in inflamed and dysplastic tissue, while epithelial expression is restricted to neoplastic tissue. In addition to identifying caspases-4 and -5 as potential targets for limiting intestinal inflammation, this study has identified epithelial-expressed caspases-4 and -5 as biomarkers with diagnostic and therapeutic potential in CRC

    The Pan-STARRS1 Photometric System

    The Pan-STARRS1 survey is collecting multi-epoch, multi-color observations of the sky north of declination -30 deg to unprecedented depths. These data are being photometrically and astrometrically calibrated and will serve as a reference for many other purposes. In this paper we present our determination of the Pan-STARRS photometric system: gp1, rp1, ip1, zp1, yp1, and wp1. The Pan-STARRS photometric system is fundamentally based on the HST Calspec spectrophotometric observations, which in turn are fundamentally based on models of white dwarf atmospheres. We define the Pan-STARRS magnitude system, and describe in detail our measurement of the system passbands, including both the instrumental sensitivity and atmospheric transmission functions. Byproducts, including transformations to other photometric systems, galactic extinction, and stellar locus are also provided.
