    Hepatocytes as in vitro test system to investigate metabolite patterns of pesticides in farmed rainbow trout and common carp: Comparison between in vivo and in vitro and across species.

    In vitro tools using isolated primary fish hepatocytes have been proposed as a useful model to study the hepatic metabolism of xenobiotics in fish. In order to evaluate the potential of in vitro fish hepatocyte assays to provide information on in vivo metabolite patterns of pesticides in farmed fish, the present study addressed the following questions: Are in vitro and in vivo metabolite patterns comparable? Are species specific differences of metabolite patterns in vivo reflected in vitro? Are metabolite patterns obtained from cryopreserved hepatocytes comparable to those from freshly isolated cells? Rainbow trout and common carp were dosed orally with feed containing the pesticide methoxychlor (MXC) for 14days. In parallel, in vitro incubations using suspensions of freshly isolated or cryopreserved primary hepatocytes obtained from both species were performed. In vivo and in vitro samples were analyzed by thin-layer chromatography with authentic standards supported by HPLC-MS. Comparable metabolite patterns from a qualitative perspective were observed in liver in vivo and in hepatocyte suspensions in vitro. Species specific differences of MXC metabolite patterns observed between rainbow trout and common carp in vivo were well reflected by experiments with hepatocytes in vitro. Finally, cryopreserved hepatocytes produced comparable metabolite patterns to freshly isolated cells. The results of this study indicate that the in vitro hepatocyte assay could be used to identify metabolite patterns of pesticides in farmed fish and could thus serve as a valuable tool to support in vivo studies as required for pesticides approval according to the EU regulation 1107

    Neodymium-140 DOTA-LM3:Evaluation of an <i>In Vivo</i> Generator for PET with a Non-Internalizing Vector

    140Nd (t1/2 = 3.4 days), owing to its short-lived positron emitting daughter 140Pr (t1/2 = 3.4 min), has promise as an in vivo generator for positron emission tomography (PET). However, the electron capture decay of 140Nd is chemically disruptive to macrocycle-based radiolabeling, meaning that an in vivo redistribution of the daughter 140Pr is expected before positron emission. The purpose of this study was to determine how the delayed positron from the de-labeled 140Pr affects preclinical imaging with 140Nd. To explore the effect, 140Nd was produced at CERN-ISOLDE, reacted with the somatostatin analogue, DOTA-LM3 (1,4,7,10- tetraazacyclododecane, 1,4,7- tri acetic acid, 10- acetamide N - p-Cl-Phecyclo(d-Cys-Tyr-d-4-amino-Phe(carbamoyl)-Lys-Thr-Cys)d-Tyr-NH2) and injected into H727 xenograft bearing mice. Comparative pre- and post-mortem PET imaging at 16 h postinjection was used to quantify the in vivo redistribution of 140Pr following 140Nd decay. The somatostatin receptor-positive pancreas exhibited the highest tissue accumulation of 140Nd-DOTA-LM3 (13% ID/g at 16 h) coupled with the largest observed redistribution rate, where 56 ± 7% (n = 4, mean ± SD) of the in situ produced 140Pr washed out of the pancreas before decay. Contrastingly, the liver, spleen, and lungs acted as strong sink organs for free 140Pr3+. Based upon these results, we conclude that 140Nd imaging with a non-internalizing vector convolutes the biodistribution of the tracer with the accumulation pattern of free 140Pr. This redistribution phenomenon may show promise as a probe of the cellular interaction with the vector, such as in determining tissue dependent internalization behavior

    Novel Insights into the Bovine Polled Phenotype and Horn Ontogenesis in Bovidae

    Despite massive research efforts, the molecular etiology of bovine polledness and the developmental pathways involved in horn ontogenesis are still poorly understood. In a recent article, we provided evidence for the existence of at least two different alleles at the Polled locus and identified candidate mutations for each of them. None of these mutations was located in known coding or regulatory regions, thus adding to the complexity of understanding the molecular basis of polledness. We confirm previous results here and exhaustively identify the causative mutation for the Celtic allele (PC) and four candidate mutations for the Friesian allele (PF). We describe a previously unreported eyelash-and-eyelid phenotype associated with regular polledness, and present unique histological and gene expression data on bovine horn bud differentiation in fetuses affected by three different horn defect syndromes, as well as in wild-type controls. We propose the ectopic expression of a lincRNA in PC/p horn buds as a probable cause of horn bud agenesis. In addition, we provide evidence for an involvement of OLIG2, FOXL2 and RXFP2 in horn bud differentiation, and draw a first link between bovine, ovine and caprine Polled loci. Our results represent a first and important step in understanding the genetic pathways and key process involved in horn bud differentiation in Bovidae

    The drivers of biogeochemistry in beach ecosystems: A cross-shore transect from the dunes to the low water line

    This study addresses key processes in high-energy beach systems using an interdisciplinary approach. We assess spatial variations in subsurface pore water residence times, salinity, organic matter (OM) availability, and redox conditions and their effects on nutrient cycles as well as on microbial community patterns and microphytobenthos growth. At the study site on Spiekeroog Island, southern North Sea, beach hydrology is characterized by the classical zonation with an upper saline plume (USP), a saltwater wedge, and a freshwater discharge tube in between. Sediment and pore water samples were taken along a cross-shore transect from the dunes to the low water line reaching sediment depths down to 5 m below sediment surface. Spatial variations in pore water residence time, salinity, and organic matter availability lead to steep redox and nutrient gradients. Vertical and horizontal differences in the microbial community indicate the influence of these gradients and salinity on the community structure. Modeled seawater flux through the USP and freshwater flux through the tube are on average 2.8 and 0.75 m3 per day and meter of shoreline, respectively. Furthermore, ridge sediments at the lower beach discharge seawater at rates of 0.5 and 1.0 m3 per day and meter of shoreline towards the runnel and seaside, respectively. Applying seawater and freshwater fluxes and representative nutrient concentrations for the discharge zones, nutrient fluxes to adjacent nearshore waters are 117 mmol NH4+, 55 mmol PO43 − and 575 mmol Si(OH)4 per day and meter of shoreline. We propose that this nutrient efflux triggers growth of microphytobenthos on sediment surfaces of the discharge zone. A first comparison of nutrient discharge rates of the beach site with a nearby sandy backbarrier tidal flat margin indicates that the beach system might be of less importance in supplying recycled nutrients to nearshore waters than the backbarrier tidal flat area

    Mensch/Maschine-Systeme in der Luftfahrt und im bodengebundenen Verkehr – Konvergenz von Konzepten und Werkzeugen

    Das Design von Mensch/Maschine-Systemen ist zentraler Forschungsgegenstand in unterschiedlichen Transportdomänen, wie zum Beispiel in der Luft- und Raumfahrt, im bodengebundenen Verkehr auf Straße und Schiene sowie in der Schifffahrt. Grundlegende Erkenntnisse zu Human Factors prägen die unterschiedlichen Bereiche in Teilen durchaus gleichförmig – z.B. sind Mode Confusion, Controllability und Shared Autonomy zentrale Aspekte, die in allen Transportdomänen eine hohe Bedeutung besitzen. Andererseits existieren sehr spezielle Anforderungen, die bei der Gestaltung von konkreten Mensch/Maschine-Systemen sehr stark spezialisiert gehandhabt werden müssen. Beispielsweise sind die Nutzer in den Bereichen Luft-/Raumfahrt hochgradig trainiert und ihre Systemumgebung ist zudem stark normiert. Im Bereich des Straßenverkehrs sind solche Randbedingungen so nicht zu beobachten – insbesondere ist die Diversität innerhalb der Nutzergruppe sehr viel höher und die Systemumwelt ist sehr viel heterogener. Hieraus resultieren jeweils andere Schwerpunkte bei der Systemgestaltung. Dieser Vortrag reflektiert die unterschiedlichen Erfahrungen zur Gestaltung von Mensch/Maschine-Systemen in der Luftfahrt und im Straßenverkehr und erörtert konvergente domänenübergreifende Konzepte im Zusammenhang mit der Systemgestaltung und der Gestaltung von Entwicklungswerkzeugen für solche Systeme
