    Beyond Agenda 2030: Future-Oriented Mechanisms in Localising the Sustainable Development Goals (SDGs)

    Given the complex nature of the UN’s Sustainable Development Goals (SDGs), there are increasing calls for new inclusive and bottom-up governance mechanisms in building a relationship between governments and their citizens, in particular, the youth, to localise the 2030 Agenda. But such successful bottom-up multi-stakeholder engagement tools have yet to emerge in practice. Hence, of specific interest in this study is exploring bottom-up approaches useful for localising the SDGs and harnessing real transformative change to leave no one behind by 2030. Using a case study from the UK, we present a novel integrated mechanism to achieve this. An integrated Social Innovation (SI) and Scenarios Thinking (ST) mechanism remains a valuable bottom-up tool capable of empowering citizens, including the youth and decision-makers in delivering coherent SDGs plans, policies, and programmes. The study reveals that although the SDGs are distinct, they are also interconnected. A scenario development workshop with youth with no prior knowledge of the SDGs showed a common thread of policy measures for different SDG future images. Standard policy measures amongst different SDGs call for an equitable society at all levels; that all energy sources be from clean and renewable sources; investment in low-carbon technologies and research; and financial support for promoting sustainable transportation and consumption measures. This study highlights that we need to change how we think and talk about SDGs and recommends socially innovative steps to embrace cross-sectoral and nexus thinking as the backdrop of the citizen science concept. We conclude that the SDGs should not become a performative exercise or failed social experiment. And any practical localisation from the UN’s Member States across the northern and southern hemispheres will require robust measures addressing future-oriented systemic thinking, inclusivity and good governance, together with standards for community resilience and sustainability

    Examining graphemic and lexical anglicisms in Twi for academic purposes in textbooks written in Twi

    This paper examines graphemic and lexical borrowings in Twi for Academic Purposes (TAP). Textbooks written in Asante Twi by some renowned scholars in the language constituted the dataset for this study. The textbooks were read and all instances of anglicisms or English features borrowed into Twi were collected. The borrowed-features were identified by drawing on our native speaker and scholarly competencies. The dataset was analyzed by drawing on Clyne’s (1977) Borrowing Typology and Haugen’s (1950) Borrowability Scale. Three key findings emerged from the analysis. First, the study yielded that in TAP two letters (<v>,  and <j>) are borrowed towards empowering Asante Twi to enable it to account for words that contain these letters. Second, at the lexical level, it was found that the borrowed words were either integrated or adapted into Asante Twi linguistic environment. The final point was that all the lexical items realized were nominals affirming the primacy of noun on borrowability scales.     Peripheral insulin resistance rather than beta cell dysfunction accounts for geographical differences in impaired fasting blood glucose among sub-Saharan African individuals: findings from the RODAM study.

    AIMS/HYPOTHESIS: The aim of this study was to assess the extent to which insulin resistance and beta cell dysfunction account for differences in impaired fasting blood glucose (IFBG) levels in sub-Saharan African individuals living in different locations in Europe and Africa. We also aimed to identify determinants associated with insulin resistance and beta cell dysfunction among this population. METHODS: Data from the cross-sectional multicentre Research on Obesity and Diabetes among African Migrants (RODAM) study were analysed. Participants included Ghanaian individuals without diabetes, aged 18-96 years old, who were residing in Amsterdam (n = 1337), Berlin (n = 502), London (n = 961), urban Ghana (n = 1309) and rural Ghana (n = 970). Glucose and insulin were measured in fasting venous blood samples. Anthropometrics were assessed during a physical examination. Questionnaires were used to assess demographics, physical activity, smoking status, alcohol consumption and energy intake. Insulin resistance and beta cell function were determined using homeostatic modelling (HOMA-IR and HOMA-B, respectively). Logistic regression analysis was used to study the contribution of HOMA-IR and inverse HOMA-B (beta cell dysfunction) to geographical differences in IFBG (fasting glucose 5.6-6.9 mmol/l). Multivariate linear regression analysis was used to identify determinants associated with HOMA-IR and inverse HOMA-B. RESULTS: IFBG was more common in individuals residing in urban Ghana (OR 1.41 [95% CI 1.08, 1.84]), Amsterdam (OR 3.44 [95% CI 2.69, 4.39]) and London (OR 1.58 [95% CI 1.20 2.08), but similar in individuals living in Berlin (OR 1.00 [95% CI 0.70, 1.45]), compared with those in rural Ghana (reference population). The attributable risk of IFBG per 1 SD increase in HOMA-IR was 69.3% and in inverse HOMA-B was 11.1%. After adjustment for HOMA-IR, the odds for IFBG reduced to 0.96 (95% CI 0.72, 1.27), 2.52 (95%CI 1.94, 3.26) and 1.02 (95% CI 0.78, 1.38) for individuals in Urban Ghana, Amsterdam and London compared with rural Ghana, respectively. In contrast, adjustment for inverse HOMA-B had very minor impact on the ORs of IFBG. In multivariate analyses, BMI (β = 0.17 [95% CI 0.11, 0.24]) and waist circumference (β = 0.29 [95%CI 0.22, 0.36]) were most strongly associated with higher HOMA-IR, whereas inverse HOMA-B was most strongly associated with age (β = 0.20 [95% CI 0.16, 0.23]) and excess alcohol consumption (β = 0.25 [95% CI 0.07, 0.43]). CONCLUSIONS/INTERPRETATION: Our findings suggest that insulin resistance, rather than beta cell dysfunction, is more important in accounting for the geographical differences in IFBG among sub-Saharan African individuals. We also show that BMI and waist circumference are important factors in insulin resistance in this population

    Functional Analysis of the Cathepsin-Like Cysteine Protease Genes in Adult Brugia malayi Using RNA Interference

    Filarial nematodes are an important group of human pathogens, causing lymphatic filariasis and onchocerciasis, and infecting around 150 million people throughout the tropics with more than 1.5 billion at risk of infection. Control of filariasis currently relies on mass drug administration (MDA) programs using drugs which principally target the microfilarial life-cycle stage. These control programs are facing major challenges, including the absence of a drug with macrofilaricidal or permanent sterilizing activity, and the possibility of the development of drug-resistance against the drugs available. Cysteine proteases are essential enzymes which play important roles in a wide range of cellular processes, and the cathepsin-like cysteine proteases have been identified as potential targets for drug or vaccine development in many parasites. Here we have studied the function of several of the cathepsin-like enzymes in the filarial nematode, B. malayi, and demonstrate that these cysteine proteases are involved in the development of embryos, show similar functions to their counterparts in C. elegans, and therefore, provide an important target for future drug development targeted to eliminate filariasis

    The evolving SARS-CoV-2 epidemic in Africa: Insights from rapidly expanding genomic surveillance

    INTRODUCTION Investment in Africa over the past year with regard to severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) sequencing has led to a massive increase in the number of sequences, which, to date, exceeds 100,000 sequences generated to track the pandemic on the continent. These sequences have profoundly affected how public health officials in Africa have navigated the COVID-19 pandemic. RATIONALE We demonstrate how the first 100,000 SARS-CoV-2 sequences from Africa have helped monitor the epidemic on the continent, how genomic surveillance expanded over the course of the pandemic, and how we adapted our sequencing methods to deal with an evolving virus. Finally, we also examine how viral lineages have spread across the continent in a phylogeographic framework to gain insights into the underlying temporal and spatial transmission dynamics for several variants of concern (VOCs). RESULTS Our results indicate that the number of countries in Africa that can sequence the virus within their own borders is growing and that this is coupled with a shorter turnaround time from the time of sampling to sequence submission. Ongoing evolution necessitated the continual updating of primer sets, and, as a result, eight primer sets were designed in tandem with viral evolution and used to ensure effective sequencing of the virus. The pandemic unfolded through multiple waves of infection that were each driven by distinct genetic lineages, with B.1-like ancestral strains associated with the first pandemic wave of infections in 2020. Successive waves on the continent were fueled by different VOCs, with Alpha and Beta cocirculating in distinct spatial patterns during the second wave and Delta and Omicron affecting the whole continent during the third and fourth waves, respectively. Phylogeographic reconstruction points toward distinct differences in viral importation and exportation patterns associated with the Alpha, Beta, Delta, and Omicron variants and subvariants, when considering both Africa versus the rest of the world and viral dissemination within the continent. Our epidemiological and phylogenetic inferences therefore underscore the heterogeneous nature of the pandemic on the continent and highlight key insights and challenges, for instance, recognizing the limitations of low testing proportions. We also highlight the early warning capacity that genomic surveillance in Africa has had for the rest of the world with the detection of new lineages and variants, the most recent being the characterization of various Omicron subvariants. CONCLUSION Sustained investment for diagnostics and genomic surveillance in Africa is needed as the virus continues to evolve. This is important not only to help combat SARS-CoV-2 on the continent but also because it can be used as a platform to help address the many emerging and reemerging infectious disease threats in Africa. In particular, capacity building for local sequencing within countries or within the continent should be prioritized because this is generally associated with shorter turnaround times, providing the most benefit to local public health authorities tasked with pandemic response and mitigation and allowing for the fastest reaction to localized outbreaks. These investments are crucial for pandemic preparedness and response and will serve the health of the continent well into the 21st century

    The characterisation of polymer-solute and solvent-solute interactions

    Solvent-solute interactions have been characterised by investigating the effect a solvent or polymer has on the wavelength at maximum absorption of a solvatochromic dye. This allowed the solvent or polymer to be characterised in terms of hydrogen bond basicity, β1, hydrogen bond acidity, α1 and dipolarity π1. Inverse gas chromatography (IGC) has also been used to characterise the polymer-solute interactions; specific retention volumes Vg, for a set of solutes (or probes) were obtained on various polymeric phases, and the logVg values regressed against the solute descriptors in a multiple linear solvation equation, logVg = c + rR2 + sπH2 + aσαH2 + bσβH2 + 1 logL16 The solute descriptors denoted by subscript 2 are R2 the excess molar refraction, π2 the solute dipolarity/ polarisability; σβH2 and σαH2 are the solute hydrogen bond basicity and acidity respectively. L16 the partition coefficient of the solute on hexadecane at 298K and it is a measure of dispersion interaction. The constants r, s, a, b and 1 are the characteristic properties of the polymer complimentary to the solute descriptors. Solvatochromic parameters have been determined for pure solvents and polymers including polystyrene (PS), polyvinylmethylether (PVME), a blend of PS and PVME, polyethylene glycol, polyester polyols and polyether polyols. IGC has been used to characterize PS, PVME, a blend of PS and PVME, polyacrylic acid (PAA), polyethylene oxide (PEO), a complex formed from PEO and PAA, and polyethyleneglycol. Literature data on retention volumes have also been analysed by the application of the linear solvation equation
