    Linear B-cell epitopes of the NS3-NS4-NS5 proteins of the hepatitis C virusas modeled with synthetic peptides

    AbstractA set of 150 synthetic peptides spanning the proteins NS3-NS4-NS5 of the hepatitis C virus (HCV) was synthesizedand tested with a panel of 20 sera obtained from HCV-infected patients. Of 62 peptides prepared from the NS3 region, none exhibited strong antigenic reactivity. Rather, five peptides from this region demonstrated specific reactivity with only 5ā€“10% of anti-HCV-positive sera. Nonetheless, it is well known that the NS3 region contains strong antigenic epitopes. These epitopes appear to be modeled in a functionally active manner with recombinant proteins and cannot be mimicked properly with short synthetic peptides. This finding suggests that the major NS3 antigenic epitopes are conformationally dependent. Seven of 20 peptides prepared from the NS4 region were immunoreactive. Five peptides from this region demonstrated very strong HCV-specific antigenic reactivity. Four of the five peptides belong to the recognized immunoreactive 5-1-1 region located inside the C100-3 antigen. One peptide demonstrating immunoreactivity with approximately 90% of anti-HCV-positive sera was found outside the C100-3 region at the C-terminal part of the NS4 protein. Of 68 peptides synthesized from the NS5 protein, 30 were immunoreactive. Six of the 30 demonstrated immunoreactivity with 35ā€“50% of anti-HCV-positive sera. Thus, the NS4 and NS5 regions of the HCV polyprotein contain a large number of specific, broadly reactive, linear antigenic epitopes. The highly antigenic reactivity of the NS5 region suggests that this protein may have significant diagnostic potential

    Spatial Pattern of Transportation Carbon Emission based on Behavior Zones: Evidence from Beijing,China.

    Reducing daily transportation carbon emission is one of themain tasksfor accomplishing low carbon city. Most existing studies have evaluated transportation carbon emission from industrial structure or micro-economic viewpoint, such as the efficiency of public transportation, work and home relationship, and modalchoiceof personal trips, but studies on the general impact of personal behaviors on transportation carbon emission is inadequate. A main reason of this relies on thelack of an appropriate spatial unit for integrating peopleā€™s greatlydiversifiedbehaviors. This study proposes the concept of ā€˜behavior zoneā€™(BZ) based on homogeneous assumption of behaviors, in order to analyze peopleā€™s traffic behavior and its carbon emission effect by sub-areas. With a survey analysis of the characteristics of peopleā€™s daily trips in Beijingā€™s sample residential areas, the critical indices of BZ are identified including housing price, development intensity and population density, and access to public transportation. With these indices, Beijing is classified into five BZ types, where the modal choice behaviors of inhabitants are projected. Then the total environmental impact of daily trips is estimated based on carbon emission levels of various traffic modes. The results provide a plenty of implications for low carbon strategies such as intensity control by floor-area-ratios and household densities, and adjustment of public transportation services

    Enhanced Breast Cancer Therapy with nsPEFs and Low Concentrations of Gemcitabine

    Chemotherapy either before or after surgery is a common breast cancer treatment. Long-term, high dose treatments with chemotherapeutic drugs often result in undesirable side effects, frequent recurrences and resistances to therapy. The anti-cancer drug, gemcitabine (GEM) was used in combination with pulse power technology with nanosecond pulsed electric fields (nsPEFs) for treatment of human breast cancer cells in vitro. Two strategies include sensitizing mammary tumor cells with GEM before nsPEF treatment or sensitizing cells with nsPEFs before GEM treatment.Breast cancer cell lines MCF-7 and MDA-MB-231 were treated with 250 65 ns-duration pulses and electric fields of 15, 20 or 25 kV/cm before or after treatment with 0.38 Ī¼M GEM. Both cell lines exhibited robust synergism for loss of cell viability 24 h and 48 h after treatment; treatment with GEM before nsPEFs was the preferred order. In clonogenic assays, only MDA-MB-231 cells showed synergism; again GEM before nsPEFs was the preferred order. In apoptosis/necrosis assays with Annexin-V-FITC/propidium iodide 2 h after treatment, both cell lines exhibited apoptosis as a major cell death mechanism, but only MDA-MB-231 cells exhibited modest synergism. However, unlike viability assays, nsPEF treatment before GEM was preferred. MDA-MB-231 cells exhibited much greater levels of necrosis then in MCF-7 cells, which were very low. Synergy was robust and greater when nsPEF treatment was before GEM. Combination treatments with low GEM concentrations and modest nsPEFs provide enhanced cytotoxicity in two breast cancer cell lines. The treatment order is flexible, although long-term survival and short-term cell death analyses indicated different treatment order preferences. Based on synergism, apoptosis mechanisms for both agents were more similar in MCF-7 than in MDA-MB-231 cells. In contrast, necrosis mechanisms for the two agents were distinctly different in MDA-MB-231, but too low to reliably evaluate in MCF-7 cells. While disease mechanisms in the two cell lines are different based on the differential synergistic response to treatments, combination treatment with GEM and nsPEFs should provide an advantageous therapy for breast cancer ablation in vivo

    Detecting and removing visual distractors for video aesthetic enhancement

    Personal videos often contain visual distractors, which are objects that are accidentally captured that can distract viewers from focusing on the main subjects. We propose a method to automatically detect and localize these distractors through learning from a manually labeled dataset. To achieve spatially and temporally coherent detection, we propose extracting features at the Temporal-Superpixel (TSP) level using a traditional SVM-based learning framework. We also experiment with end-to-end learning using Convolutional Neural Networks (CNNs), which achieves slightly higher performance than other methods. The classification result is further refined in a post-processing step based on graph-cut optimization. Experimental results show that our method achieves an accuracy of 81% and a recall of 86%. We demonstrate several ways of removing the detected distractors to improve the video quality, including video hole filling; video frame replacement; and camera path re-planning. The user study results show that our method can significantly improve the aesthetic quality of videos

    A publicly verifiable quantum signature scheme based on asymmetric quantum cryptography

    In 2018, Shi et al. \u27s showed that Kaushik et al.\u27s quantum signature scheme is defective. It suffers from the forgery attack. They further proposed an improvement, trying to avoid the attack. However, after examining we found their improved quantum signature is deniable, because the verifier can impersonate the signer to sign a message. After that, when a dispute occurs, he can argue that the signature was not signed by him. It was from the signer. To overcome the drawback, in this paper, we raise an improvement to make it publicly verifiable and hence more suitable to be applied in real life. After cryptanalysis, we confirm that our improvement not only resist the forgery attack but also is undeniable

    Polycystic ovary syndrome in patients with epilepsy: A study in 102 Chinese women

    AbstractPurposeThe incidence of polycystic ovary syndrome (PCOS) increases in women with epilepsy (WWE), which appears to vary with ethnicity. This study was conducted to determine the incidence and risk factors of PCOS in Chinese WWE.MethodsThe study was carried out in 102 of 139 Chinese WWE at reproductive ages, with 32 receiving valproic acid (VPA), 40 receiving other antiepileptic drugs (AEDs), and 30 without AEDs therapy. PCOS was defined as having 2 or more of the following components: polycystic ovaries, hyperandrogenism, and amenorrhoea or oligomenorrhoea (a/oligomenorrhoea).ResultsOne or more isolated components of PCOS were found in 56 (54.9%) patients, with 29 (28.4%) having polycystic ovaries, 20 (19.6%) with a/oligomenorrhea, 7 (6.9%) with hyperandrogenism, and 13 (12.7%) with defined PCOS. Their average age at the start of seizure was 13.8Ā±6.5years, younger than that of patients without these disorders (16.9Ā±8.6years, p<0.05). VPA therapy increased the incidence of PCOS (11/32, 34.4%), in addition to increased blood levels of testosterone and luteinizing hormone (LH) as well as LH to FSH (follicle-stimulating hormone) ratio. No significant relationship was found between the incidence of PCOS and the type, duration, or frequency of seizures in these WWE.ConclusionThere is an increased incidence of PCOS in Chinese WWE at reproductive ages, by more than 2 times of that in the general population. Risk factors include seizures starting at a young age and VPA therapy
