    Tuning ubiquinone position in biomimetic monolayer membranes

    Artificial lipid bilayers have been extensively studied as models that mimic natural membranes (biomimetic membranes). Several attempts of biomimetic membranes inserting ubiquinone (UQ) have been performed to enlighten which the position of UQ in the lipid layer is, although obtaining contradictory results. In this work, pure components (DPPC and UQ) and DPPC:UQ mixtures have been studied using surface pressure-area isotherms and Langmuir-Blodgett (LB) films of the same compounds have been transferred onto solid substrates being topographically characterized on mica using atomic force microscopy and electrochemically on indium tin oxide slides. DPPC:UQ mixtures present less solid-like physical state than pure DPPC indicating a higher-order degree for the latter. UQ influences considerably DPPC during the fluid state, but it is mainly expelled after the phase transition at ˜˜ 26 mN·m^-1 for the 5:1 ratio and at ˜˜ 21 mN·m^-1 for lower UQ content. The thermodynamic studies confirm the stability of the DPPC:UQ mixtures before that event, although presenting a non-ideal behaviour. The results indicate that UQ position can be tuned by means of the surface pressure applied to obtain LB films and the UQ initial content. The UQ positions in the biomimetic membrane are distinguished by their formal potential: UQ located in “diving” position with the UQ placed in the DPPC matrix in direct contact with the electrode surface ( -0.04±0.02 V), inserted between lipid chains without contact to the substrate ( 0.00±0.01 V) and parallel to the substrate, above the lipid chains ( 0.09±0.02 V).Peer ReviewedPostprint (author's final draft

    The NADPH-Dependent thioredoxin reductase C-2-Cys peroxiredoxin redox system modulates the activity of thioredoxin x in arabidopsis chloroplasts

    The chloroplast redox network is composed of a complex set of thioredoxins (Trxs), reduced by ferredoxin (Fdx) via a Fdx-dependent Trx reductase (FTR), and an NADPH-dependent Trx reductase with a joint Trx domain, NTRC, which efficiently reduces 2-Cys peroxiredoxins (2-Cys Prxs). Recently, it was proposed that the redox balance of 2-Cys Prxs maintains the redox state of f-type Trxs, thus allowing the proper redox regulation of Calvin-Benson cycle enzymes such as fructose 1,6-bisphosphatase (FBPase). Here, we have addressed whether the action of 2-Cys Prxs is also exerted on Trx x. To that end, an Arabidopsis thaliana quadruple mutant, ntrc-trxx-δ "2cp, which is knocked out for NTRC and Trx x, and contains severely decreased levels of 2-Cys Prxs, was generated. In contrast to ntrc-trxx, which showed a severe growth inhibition phenotype and poor photosynthetic performance, the ntrc-trxx-δ "2cp mutant showed a significant recovery of growth rate and photosynthetic efficiency, indicating that the content of 2-Cys Prxs is critical for the performance of plants lacking both NTRC and Trx x. Light-dependent reduction of FBPase was severely impaired in mutant plants lacking NTRC or NTRC plus Trx x, despite the fact that neither NTRC nor Trx x is an effective reductant of this enzyme. However, FBPase reduction was recovered in the ntrc-trxx-δ "2cp mutant. Our results show that the redox balance of 2-Cys Prxs, which is mostly dependent on NTRC, modulates the activity of Trx x in a similar way as f-type Trxs, thus suggesting that the activity of these Trxs is highly interconnected.Ministerio de Economía y Competitividad BIO2017-85195-C2-1-

    Contribution of a pumped-storage hydropower plant to reduce the scheduling costs of an isolated power system with high wind power penetration

    AbstractThe paper aims at demonstrating that the consideration of constant start-up costs and ramps of the thermal generating units for assessing the contribution of pumped-hydro energy storage to reduce the scheduling costs of hydrothermal power systems with high wind penetration, may yield unrealistic results. For this purpose, an isolated power system is used as a case study. The contribution of a pumped-storage hydropower plant to reduce the system scheduling costs is assessed in the paper by using a hydrothermal weekly unit commitment model. The model considers different start-up costs and ramps of the thermal generating units as a function of the start-up type. The effects of including pumped hydro energy storage in the system on the integration of wind energy, and on the start-ups and capacity factors of the thermal generating units are also evaluated. The results of the paper demonstrate that the consideration of constant start-up costs and ramps of the thermal generating units yields unrealistic results, and that the pumped-storage hydropower plant may help reduce the system scheduling costs by 2.5–11% and integrate wind power and may allow dispensing with some inflexible thermal generating units

    Dimensioning influence on the final critical stage of double-layer space trusses

    [Abstract] The present paper suggests the application of a non-linear model of analysis to study the final critical stage of double-Iayer space trusses. A description of the analysis techniques is proposed to take into account the resistant and geometric nonlinearity. The model developed has been applied to the study of the repercussion that the dimensioning of the members has on the stress-strain curves of the structure, as well as in the evaluation of itsbearing capability when the final limit stage is reached. In order to exemplify this, various solved cases are enclosed, corresponding to different typologies of trusses and dimensions for which different dimensioning criteria have been used. Furthermore, a new method is proposed for the dimensioning of space structures, starting from the described technique of non-linear analysis

    Analysis of Two Polyhydroxyalkanoate Synthases in Bradyrhizobium japonicum USDA 110

    Bradyrhizobium japonicum USDA 110 has five polyhydroxyalkanoate (PHA) synthases (PhaC) annotated in its genome: bll4360 (phaC1), bll6073 (phaC2), blr3732 (phaC3), blr2885 (phaC4), and bll4548 (phaC5). All these proteins possess the catalytic triad and conserved amino acid residues of polyester synthases and are distributed into four different PhaC classes. We obtained mutants in each of these paralogs and analyzed phaC gene expression and PHA production in liquid cultures. Despite the genetic redundancy, only phaC1 and phaC2 were expressed at significant rates, while PHA accumulation in stationary-phase cultures was impaired only in the phaC1 mutant. Meanwhile, the phaC2 mutant produced more PHA than the wild type under this condition, and surprisingly, the phaC3 transcript increased in the phaC2 background. A double mutant, the phaC2 phaC3 mutant, consistently accumulated less PHA than the phaC2 mutant. PHA accumulation in nodule bacteroids followed a pattern similar to that seen in liquid cultures, being prevented in the phaC1 mutant and increased in the phaC2 mutant in relation to the level in the wild type. Therefore, we used these mutants, together with a phaC1 phaC2 double mutant, to study the B. japonicum PHA requirements for survival, competition for nodulation, and plant growth promotion. All mutants, as well as the wild type, survived for 60 days in a carbon-free medium, regardless of their initial PHA contents. When competing for nodulation against the wild type in a 1:1 proportion, the phaC1 and phaC1 phaC2 mutants occupied only 13 to 15% of the nodules, while the phaC2 mutant occupied 81%, suggesting that the PHA polymer is required for successful competitiveness.     Simheuristics extend metaheuristics by adding a simulation layer that allows the optimization component to deal efficiently with scenarios under uncertainty. This presentation reviews both initial as well as recent applications of simheuristics, mainly in the area of logistics and transportation. We also discuss a novel agent-based simheuristic (ABSH) approach that combines simheuristic and multi-agent systems to efficiently solve stochastic combinatorial optimization problems.     A proposed reaction mechanism for rice NADPH thioredoxin reductase C, an enzyme with protein disulfide reductase activity

    AbstractNADPH thioredoxin reductase C (NTRC) is an interesting NTR with a thioredoxin (Trx) domain at the C-terminus, able to conjugate both activities for 2-Cys peroxiredoxin (Prx) reduction. NTRC is dimeric in the presence of NADPH and interacted with dimeric 2-Cys Prx through the Trx module by a mixed disulfide between Cys377 of NTRC and Cys61 of the 2-Cys Prx. NTRC variants of both NTR and Trx active sites were inactive, but 1:1 mixtures of both variants allowed partial recovery of activity suggesting inter-subunit transfer of electrons during catalysis.     A variable neighborhood search simheuristic for project portfolio selection under uncertainty

    With limited nancial resources, decision-makers in rms and governments face the task of selecting the best portfolio of projects to invest in. As the pool of project proposals increases and more realistic constraints are considered, the problem becomes NP-hard. Thus, metaheuristics have been employed for solving large instances of the project portfolio selection problem (PPSP). However, most of the existing works do not account for uncertainty. This paper contributes to close this gap by analyzing a stochastic version of the PPSP: the goal is to maximize the expected net present value of the inversion, while considering random cash ows and discount rates in future periods, as well as a rich set of constraints including the maximum risk allowed. To solve this stochastic PPSP, a simulation-optimization algorithm is introduced. Our approach integrates a variable neighborhood search metaheuristic with Monte Carlo simulation. A series of computational experiments contribute to validate our approach and illustrate how the solutions vary as the level of uncertainty increases

    Mechanical properties of hot mix asphalt made with recycled aggregates from reclaimed construction and demolition debris

    [Resumen:] En este trabajo se dosificaron mezclas bituminosas fabricadas con áridos reciclados de residuos de construcción y demolición. Se realizaron una serie de ensayos que permitieron caracterizar el comportamiento mecánico de estas mezclas. Los parámetros mecánicos de las mezclas con áridos reciclados son similares a los de las mezclas fabricadas únicamente con áridos naturales de cantera. Sin embargo, las mezclas bituminosas con áridos reciclados tuvieron un mal comportamiento frente a la disminución de la resistencia por pérdida de cohesión por la acción del agua. No obstante, mediante la selección de materiales de mejor calidad y corrección de este mal comportamiento, existen posibilidades de utilizar áridos reciclados en mezclas bituminosas. Para ello será necesario ampliar la primera aproximación realizada en este artículo.[Abstract:] The mix design for asphalt mixtures containing recycled aggregates from construction and demolition debris was evaluated. The tests conducted to characterize the mechanical behaviour of these mixtures showed that the mechanical properties of mixtures with recycled and virgin aggregate are similar. The asphalt mixtures containing recycled aggregate proved to have lower resistance to water action. Nonetheless, recycled aggregate was found to be potentially usable in asphalt mixtures if higher quality materials are selected and such low resistance is corrected. This will call for expanding upon the preliminary work described in the present article
