    Strategic priorities for agricultural development in Eastern and Central Africa:

    "Agricultural development strategies delineate priorities for actions to enhance agricultural and overall development. They are usually put forward by individual countries based on assessments of national needs. Seldom are attempts made to identify strategic priorities for agricultural development that cut across national boundaries. This gap is perhaps not surprising—organizations mandated to develop and implement regional agricultural development programs are rare. Although the gap may be understandable, it is also troubling. This report helps to fill that gap for eastern and central Africa (ECA), focusing on Burundi, Democratic Republic of Congo (DRC), Eritrea, Ethiopia, Kenya, Madagascar, Rwanda, Sudan, Tanzania, and Uganda. Recent trends and the current performance of agriculture in these countries expose a region progressively less able to meet the needs of its burgeoning population...     A four-step concept for fault detection and fault diagnosis for solar thermal systems has been developed in the framework of the ongoing research activities at Kassel University. This concept is able to combine different general approaches for fault detection and fault diagnosis in one structure.     A 5‑lipoxygenase-specific sequence motif impedes enzyme activity and confers dependence on a partner protein

    © 2018 Elsevier B.V. Leukotrienes (LT) are lipid mediators of the inflammatory response that play key roles in diseases such as asthma and atherosclerosis. The precursor leukotriene A 4 (LTA 4 ) is synthesized from arachidonic acid (AA) by 5‑lipoxygenase (5-LOX), a membrane-associated enzyme, with the help of 5‑lipoxygenase-activating protein (FLAP), a nuclear transmembrane protein. In lipoxygenases the main chain carboxylate of the C-terminus is a ligand for the non-heme iron and thus part of the catalytic center. We investigated the role of a lysine-rich sequence (KKK 653–655 ) 20 amino acids upstream of the C-terminus unique to 5-LOX that might displace the main-chain carboxylate in the iron coordination sphere. A 5-LOX mutant in which KKK 653–655 is replaced by ENL was transfected into HEK293 cells in the absence and presence of FLAP. This mutant gave ~20-fold higher 5-LOX product levels in stimulated HEK cells relative to the wild-type 5-LOX. Co-expression of the enzymes with FLAP led to an equalization of 5-LOX products detected, with wild-type 5-LOX product levels increased and those from the mutant enzyme decreased. These data suggest that the KKK motif limits 5-LOX activity and that this attenuated activity must be compensated by the presence of FLAP as a partner protein for effective LT biosynthesis

    Effectiveness and safety of tocilizumab in patients with systemic sclerosis: a propensity score matched controlled observational study of the EUSTAR cohort

    OBJECTIVES: Tocilizumab showed trends for improving skin fibrosis and prevented progression of lung fibrosis in systemic sclerosis (SSc) in randomised controlled clinical trials. We aimed to assess safety and effectiveness of tocilizumab in a real-life setting using the European Scleroderma Trial and Research (EUSTAR) database. METHODS: Patients with SSc fulfilling the American College of Rheumatology (ACR)/EULAR 2013 classification criteria, with baseline and follow-up visits at 12±3 months, receiving tocilizumab or standard of care as the control group, were selected. Propensity score matching was applied. Primary endpoints were the modified Rodnan skin score (mRSS) and FVC at 12±3 months compared between the groups. Secondary endpoints were the percentage of progressive/regressive patients for skin and lung at 12±3 months. RESULTS: Ninety-three patients with SSc treated with tocilizumab and 3180 patients with SSc with standard of care fulfilled the inclusion criteria. Comparison between groups did not show significant differences, but favoured tocilizumab across all predefined primary and secondary endpoints: mRSS was lower in the tocilizumab group (difference -1.0, 95% CI -3.7 to 1.8, p=0.48). Similarly, FVC % predicted was higher in the tocilizumab group (difference 1.5 (-6.1 to 9.1), p=0.70). The percentage of progressive/regressive patients favoured tocilizumab over controls. These results were robust regarding the sensitivity analyses. Safety analysis confirmed previously reported adverse event profiles. CONCLUSION: Although this large, observational, controlled, real-life EUSTAR study did not show significant effectiveness of tocilizumab on skin and lung fibrosis, the consistency of direction of all predefined endpoints generates hypothesis for potential effectiveness in a broader SSc population

    Introduction of a guideline for measurements of greenhouse gas fluxes from soils using non-steady-state chambers

    Method Soils represent a major global source and sink of greenhouse gases (GHGs). Many studies of GHG fluxes between soil, plant and atmosphere rely on chamber measurements. Different chamber techniques have been developed over the last decades, each characterised by different requirements and limitations. In this manuscript, we focus on the non-steady-state technique which is widely used for manual measurements but also in automatic systems. Although the measurement method appears very simple, experience gained over the years shows that there are many details which have to be taken into account to obtain reliable measurement results. Aim This manuscript aims to share lessons learnt and pass on experiences in order to assist the reader with possible questions or unexpected challenges, ranging from the planning of the design of studies and chambers to the practical handling of the chambers and the quality assurance of the gas and data analysis. This concise introduction refers to a more extensive Best Practice Guideline initiated by the Working Group Soil Gases (AG Bodengase) of the German Soil Science Society (Deutsche Bodenkundliche Gesellschaft). The intention was to collect and aggregate the expertise of different working groups in the research field. As a compendium, this Best Practice Guideline is intended to help both beginners and experts to meet the practical and theoretical challenges of measuring soil gas fluxes with non-steady-state chamber systems and to improve the quality of the individual flux measurements and thus entire GHG studies by reducing sources of uncertainty and error

    SolNet:PhD-scholarships and courses on solar heating

    AbstractSolNet, founded in 2006, is the first coordinated International PhD education program on Solar Thermal Engineering. The SolNet network is coordinated by the Institute of Thermal Engineering at Kassel University, Germany. The network offers PhD courses on solar heating and cooling, conference-accompanying Master courses, placements of internships, and PhD scholarship projects. A new scholarship project, “SHINE”, will be launched in autumn 2013 in the frame work of the Marie Curie program of the European Union (Initial Training Network, ITN). 13 PhD-scholarships on solar district heating, solar heat for industrial processes, as well as sorption stores and materials will be offered, starting in December 2013. Additionally, the project comprises a training program with five PhD courses and several workshops on solar thermal engineering that will be open also for other PhD students working in the field. The research projects will be hosted by six different universities and five companies from all over Europe

    Differential sensing with arrays of de novo designed peptide assemblies

    Differential sensing attempts to mimic the mammalian senses of smell and taste to identify analytes and complex mixtures. In place of hundreds of complex, membrane-bound G-protein coupled receptors, differential sensors employ arrays of small molecules. Here we show that arrays of computationally designed de novo peptides provide alternative synthetic receptors for differential sensing. We use self-assembling α-helical barrels (αHBs) with central channels that can be altered predictably to vary their sizes, shapes and chemistries. The channels accommodate environment-sensitive dyes that fluoresce upon binding. Challenging arrays of dye-loaded barrels with analytes causes differential fluorophore displacement. The resulting fluorimetric fingerprints are used to train machine-learning models that relate the patterns to the analytes. We show that this system discriminates between a range of biomolecules, drink, and diagnostically relevant biological samples. As αHBs are robust and chemically diverse, the system has potential to sense many analytes in various settings
