    The genome-scale metabolic network analysis of Zymomonas mobilis ZM4 explains physiological features and suggests ethanol and succinic acid production strategies

    <p>Abstract</p> <p>Background</p> <p><it>Zymomonas mobilis </it>ZM4 is a Gram-negative bacterium that can efficiently produce ethanol from various carbon substrates, including glucose, fructose, and sucrose, <it>via </it>the Entner-Doudoroff pathway. However, systems metabolic engineering is required to further enhance its metabolic performance for industrial application. As an important step towards this goal, the genome-scale metabolic model of <it>Z. mobilis </it>is required to systematically analyze <it>in silico </it>the metabolic characteristics of this bacterium under a wide range of genotypic and environmental conditions.</p> <p>Results</p> <p>The genome-scale metabolic model of <it>Z. mobilis </it>ZM4, ZmoMBEL601, was reconstructed based on its annotated genes, literature, physiological and biochemical databases. The metabolic model comprises 579 metabolites and 601 metabolic reactions (571 biochemical conversion and 30 transport reactions), built upon extensive search of existing knowledge. Physiological features of <it>Z. mobilis </it>were then examined using constraints-based flux analysis in detail as follows. First, the physiological changes of <it>Z. mobilis </it>as it shifts from anaerobic to aerobic environments (i.e. aerobic shift) were investigated. Then the intensities of flux-sum, which is the cluster of either all ingoing or outgoing fluxes through a metabolite, and the maximum <it>in silico </it>yields of ethanol for <it>Z. mobilis </it>and <it>Escherichia coli </it>were compared and analyzed. Furthermore, the substrate utilization range of <it>Z. mobilis </it>was expanded to include pentose sugar metabolism by introducing metabolic pathways to allow <it>Z. mobilis </it>to utilize pentose sugars. Finally, double gene knock-out simulations were performed to design a strategy for efficiently producing succinic acid as another example of application of the genome-scale metabolic model of <it>Z. mobilis</it>.</p> <p>Conclusion</p> <p>The genome-scale metabolic model reconstructed in this study was able to successfully represent the metabolic characteristics of <it>Z. mobilis </it>under various conditions as validated by experiments and literature information. This reconstructed metabolic model will allow better understanding of <it>Z. mobilis </it>metabolism and consequently designing metabolic engineering strategies for various biotechnological applications.</p

    Temperament and Character Profiles of Sasang Typology in an Adult Clinical Sample

    The purpose of this study was to examine the biopsychological personality profiles of traditional Korean Sasang typology based on the Temperament and Character Inventory (TCI) in a Korean adult clinical sample. A total of 97 adults completed the Korean version of the TCI. The participants were classified as one of three traditional Korean Sasang types (31 So-Yang, 41 Tae-Eum, 25 So-Eum) by three specialists in Sasang typology. The seven dimensions of TCI were compared between the different Sasang types using analysis of variance (ANOVA) and profile analysis. There were no significant differences in age, gender and education across the Sasang types. The TCI profile for each of the Sasang types was significantly different (profile analysis, df = 5.038, F = 3.546, P = .004). There were significant differences in the temperament dimensions of Novelty Seeking (F = 3.43, P = .036) and Harm Avoidance (F = 5.43, P = .006) among the Sasang types. The Novelty Seeking score of the So-Yang type (31.90 ± 9.87) was higher than that of the So-Eum type (25.24 ± 9.21; P = .019) while the So-Eum type (44.64 ± 8.47) scored higher on the Harm Avoidance score compared to the So-Yang type (35.16 ± 11.50; P = .003). There were no significant differences in the temperament dimension of Reward Dependence and Persistence, and the three character dimensions of Self-Directedness, Cooperativeness and Self-Transcendence. Results demonstrated distinct temperament traits associated with traditional Korean Sasang types using an objective biopsychological personality inventory. With further study, the Sasang typology may lead to enhanced clinical safety and efficacy as part of personalized medicine with traditional medicine

    Anti-Inflammatory Effects of Bangpungtongsung-San, a Traditional Herbal Prescription

    Bangpungtongsung-san (BPTS), a traditional oriental herbal prescription, is widely used for expelling wind, draining heat, and providing general improvement to the immune system. In this study, we investigated the effects of BPTS on induction of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), proinflammatory cytokines, nuclear factor-kappa B (NF-κB), and mitogen-activated protein kinases (MAPKs) in lipopolysaccharide- (LPS- ) stimulated Raw 264.7 cells, and on paw edema in rats. At concentrations of 0.5, 0.75, and 1 mg/mL, treatment with BPTS inhibited levels of expression of LPS-induced NF-κB and MAPKs (ERK, JNK, and p38) as well as production of proinflammatory mediators, such as nitric oxide (NO), prostaglandin E2 (PGE2), tumor necrosis factor-α (TNF-α), and interleukin-6 (IL-6) by LPS. These results suggest that BPTS may exert anti-inflammatory effects via reduction of proinflammatory mediators, including NO, PGE2, TNF-α, and IL-6 through suppression of the signaling pathways of NF-κB and MAPKs in LPS-induced macrophages. In addition, using the carrageenan-induced paw edema assay, an antiedema effect of BPTS was observed in rats. These findings may provide scientific evidence validating the use of BPTS in treatment of patients with heat syndrome in Korean oriental medicine

    PCR for Diagnosis of Male Trichomonas vaginalis Infection with Chronic Prostatitis and Urethritis

    The aim of this study was to assess the usefulness of PCR for diagnosis of Trichomonas vaginalis infection among male patients with chronic recurrent prostatitis and urethritis. Between June 2001 and December 2003, a total of 33 patients visited the Department of Urology, Hanyang University Guri Hospital and were examined for T. vaginalis infection by PCR and culture in TYM medium. For the PCR, we used primers based on a repetitive sequence cloned from T. vaginalis (TV-E650). Voided bladder urine (VB1 and VB3) was sampled from 33 men with symptoms of lower urinary tract infection (urethral charge, residual urine sensation, and frequency). Culture failed to detect any T. vaginalis infection whereas PCR identified 7 cases of trichomoniasis (21.2%). Five of the 7 cases had been diagnosed with prostatitis and 2 with urethritis. PCR for the 5 prostatitis cases yielded a positive 330 bp band from bothVB1 and VB3, whereas positive results were only obtained from VB1 for the 2 urethritis patients. We showed that the PCR method could detect T. vaginalis when there was only 1 T. vaginalis cell per PCR mixture. Our results strongly support the usefulness of PCR on urine samples for detecting T. vaginalis in chronic prostatitis and urethritis patients

    The Antioxidant Activity and Their Major Antioxidant Compounds from Acanthopanax senticosus and A. koreanum

    The antioxidant activity and chlorogenic acid and caffeic acid contents were investigated from different parts of Acanthopanax senticosus and A. koreanum. Antioxidant activity was assessed by various in vitro assays such as DPPH, ABTS, FRAP, reducing power assays and ORAC, and the chlorogenic acid and caffeic acid were validated by HPLC chromatography. Among the various extracts, the fruit extracts of A. senticosus and A. koreanum exhibited strongest antioxidant activities including ABTS, FRAP, reducing power and ORAC, however, strongest DPPH radical scavenging activity was observed from the leaf extract of A. senticosus. In addition, the antioxidant activities of various extracts were correlated with total phenolic and proanthocyanidin contents. The major phenolic contents from various parts of these plants observed that leaf extract of A. senticosus expressed higher levels of chlorogenic acid (14.86 mg/dry weigh g) and caffeic acid (3.09 mg/dry weigh g) than other parts. Therefore, these results suggest that the leaf of A. senticosus may be an excellent natural source for functional foods and pharmaceutical agents, and the validated method was useful for the quality control of A. senticosus

    A standardized formula for aesthetic mandibular reconstruction using an osteocutaneous fibular free flap

    Ameloblastoma is the most common benign odontogenic tumor of the jaw, and expansional growth of a huge untreated ameloblastoma can result in disturbances in facial aesthetics and function, such as difficulty with mouth opening, swallowing, chewing, breathing, neurologic deficits, and pathologic fractures. Radical wide resection with safety margins and subsequent reconstruction is generally recommended. A fibular free flap (FFF) is commonlyused to reconstruct the mandible in order to adequately restore both aesthetic appearance and function. The aim of this brief clinical report is to present a case of huge ameloblastoma after wide resection with free safety margins, and describe the immediate one-step mandibular reconstruction using a vascularized composite FFF. The sterolithographic(rapid prototype, RP) model, a wax pattern of the resected mandible, and a surgical fibular stent made from the wax pattern were constructed preoperatively.     Changes in oncogenic protein levels in peri-implant oral malignancy: a case report

    Background Oral squamous cell carcinoma (OSCC) constitutes a group of tumors that exhibit heterogeneous biology, histopathology, and clinical behaviors. Case presentation A 73-year-old male had a whitish leukoplakia-like lesion around inflamed peri-implant area (#42, #43, and #44), and this lesion had transformed to OSCC within 3 years. He underwent mass resection, selective neck dissection, and reconstructive surgery. To detect any carcinogenesis progression, we examined the removed tumor tissue as well as the patients preoperative and postoperative sera to identify causative oncogenic proteins using immunoprecipitation high-performance liquid chromatography (IP-HPLC). Conclusions The protein expression levels of p53, E-cadherin, β-catenin, MMP-10, HER2, NRAS, Met, HER2, and ERb were significantly lower in the serum collected on postoperative day 10 than in the preoperative serum, and if these proteins are consistently not elevated in the serum 3 months after surgery compared with the preoperative serum, these proteins can be potential oncogenic proteins. However, we also found that the serum extracted 3 months after the operation had elevated levels of oncogenic proteins compared with that of the preoperative and 10-day postoperative serum indicating the possibility of tumor recurrence. At postoperative follow-up period, ipsilateral neck metastasis and second primary lesion were found and additional surgery was performed to the patient. IP-HPLC using the patients serum shows the possibility of oncogenic protein detection. However, follow-up IP-HPLC data is needed to find out patient-specific prognostic factors

    A-Site Cation Engineering for Efficient Blue-Emissive Perovskite Light-Emitting Diodes

    Metal halide perovskites have been investigated for the next-generation light-emitting materials because of their advantages such as high photoluminescence quantum yield (PLQY), excellent color purity, and facile color tunability. Recently, red- and green-emissive perovskite light-emitting diodes (PeLEDs) have shown an external quantum efficiency (EQE) of over 20%, whereas there is still room for improvement for blue emissive PeLEDs. By controlling the halide compositions of chloride (Cl-) and bromide (Br-), the bandgap of perovskites can be easily tuned for blue emission. However, halide segregation easily occurrs in the mixed-halide perovskite under light irradiation and LED operation because of poor phase stability. Here, we explore the effect of A-site cation engineering on the phase stability of the mixed-halide perovskites and find that a judicious selection of low dipole moment A cation (formamidinium or cesium) suppresses the halide segregation. This enables efficient bandgap tuning and electroluminescence stability for sky blue emissive PeLEDs over the current density of 15 mA/cm(2)

    High-Spatial Resolution Monitoring of Phycocyanin and Chlorophyll-a Using Airborne Hyperspectral Imagery

    Hyperspectral imagery (HSI) provides substantial information on optical features of water bodies that is usually applicable to water quality monitoring. However, it generates considerable uncertainties in assessments of spatial and temporal variation in water quality. Thus, this study explored the influence of different optical methods on the spatial distribution and concentration of phycocyanin (PC), chlorophyll-a (Chl-a), and total suspended solids (TSSs) and evaluated the dependence of algal distribution on flow velocity. Four ground-based and airborne monitoring campaigns were conducted to measure water surface reflectance. The actual concentrations of PC, Chl-a, and TSSs were also determined, while four bio-optical algorithms were calibrated to estimate the PC and Chl-a concentrations. Artificial neural network atmospheric correction achieved Nash-Sutcliffe Efficiency (NSE) values of 0.80 and 0.76 for the training and validation steps, respectively. Moderate resolution atmospheric transmission 6 (MODTRAN 6) showed an NSE value &gt;0.8; whereas, atmospheric and topographic correction 4 (ATCOR 4) yielded a negative NSE value. The MODTRAN 6 correction led to the highest R-2 values and lowest root mean square error values for all algorithms in terms of PC and Chl-a. The PC:Chl-a distribution generated using HSI proved to be negatively dependent on flow velocity (p-value = 0.003) and successfully indicated cyanobacteria risk regions in the study area
