    Understanding and Application of Systems Thinking to Diversity Challenges at Higher Education Institutions

    This research argues that establishing and sustaining leadership diversity within Higher Education Institutions (HEIs) is a complex-systems challenge and that those responsible for leading efforts to establish and sustain diversity, referred to as DEI leaders, should adopt a mindset of systems thinking and apply problem-solving methods and tools informed by systems thinking. Semi-structured interviews with DEI Leaders at HEIs were conducted regarding background and perceptions of their work, and how they thought about and made choices concerning diversity challenges. Interview questions were designed to gain an understanding of the degree to which these leaders think about DEI and if they approach these problems from a systems perspective and use system-informed methodologies and tools. Results indicated that DEI leaders acknowledged diversity to be a challenging problem but there was little understanding that this kind of problem was either complex or systemic. Furthermore, DEI leaders relied on conventional linear formulations and problem solving because they did not know how to think in systems or use methods and tools derived from this mode of thinking. Implications suggest that those responsible for leading efforts to establish and sustain diversity within HEIs rely on methodologies and tools which are insufficient to solve complex DEI problems. Recommendations are made for education to help DEI leaders to adopt a mindset of systems thinking and apply problem solving methods and tools informed by this mode of cognition

    The WTO in Buenos Aires: the outcome and its significance for the future of the multilateral trading system

    The conclusion of the World Trade Organization’s (WTO) Buenos Aires ministerial conference (10-13 December 2017) was immediately celebrated and derided in equal measure. For its supporters, Buenos Aires opened the way toward negotiations in e-commerce, investment facilitation for development, and measures designed to help micro, small and medium sized enterprises (MSMEs). For its detractors, the meeting underscored the gridlock that continues to blight the WTO’s negotiating function and underlined the organisation’s declining credibility as a mechanism for governing global trade. In this paper we provide one of the first full length critical evaluations of the Buenos Aires conference and its outcome. In so doing, we offer answers to three questions. What accounts for such dramatically different assessments of the meeting’s outcome? How should the outcome be interpreted? What is its significance for the future of the WTO and the multilateral trading system? We argue that the meeting’s outcome was indeed significant. It has consolidated the process of reconfiguring the WTO’s negotiating function; and it enables members to tackle more effectively a range of pressing economic and social issues as well as to navigate blockers and blockages in the negotiations.     Specific structure and unique function define the hemicentin

    Hemicentin has come a long way from when it was first identified in C. elegans as him-4 (High incidence of males). The protein is now a recognized player in maintaining the architectural integrity of vertebrate tissues and organs. Highly conserved hemicentin sequences across species indicate this gene’s ancient evolutionary roots and functional importance. In mouse, hemicentin is liberally distributed on the cell surface of many cell types, including epithelial cells, endothelial cells of the eye, lung, and uterus, and trophectodermal cells of blastocyst. Recent discoveries have uncovered yet another vital purpose of hemicentin 1. The protein also serves a unique function in mitotic cytokinesis, during which this extracellular matrix protein plays a key role in cleavage furrow maturation. Though understanding of hemicentin function has improved through new discoveries, much about this protein remains mysterious

    Tumour cell CD99 regulates transendothelial migration via CDC42 and actin remodelling

    Metastasis requires tumour cells to cross endothelial cell (EC) barriers using pathways similar to those used by leucocytes during inflammation. Cell surface CD99 is expressed by healthy leucocytes and ECs, and participates in inflammatory transendothelial migration (TEM). Tumour cells also express CD99, and we have analysed its role in tumour progression and cancer cell TEM. Tumour cell CD99 was required for adhesion to ECs but inhibited invasion of the endothelial barrier and migratory activity. Furthermore, CD99 depletion in tumour cells caused redistribution of the actin cytoskeleton and increased activity of the Rho GTPase CDC42, known for its role in actin remodelling and cell migration. In a xenograft model of breast cancer, tumour cell CD99 expression inhibited metastatic progression, and patient samples showed reduced expression of the CD99 gene in brain metastases compared to matched primary breast tumours. We conclude that CD99 negatively regulates CDC42 and cell migration. However, CD99 has both pro- and anti-tumour activity, and our data suggest that this results in part from its functional linkage to CDC42 and the diverse signalling pathways downstream of this Rho GTPase.     High Angular Resolution and Lightweight X-Ray Optics for Astronomical Missions

    X-ray optics with both high angular resolution and lightweight is essential for further progress in x-ray astronomy. High angular resolution is important in avoiding source confusion and reducing background to enable the observation of the most distant objects of the early Universe. It is also important in enabling the use of gratings to achieve high spectral resolution to study, among other things, the myriad plasmas that exist in planetary, stellar, galactic environments, as well as interplanetary, inter-stellar, and inter-galactic media. Lightweight is important for further increase in effective photon collection area, because x-ray observations must take place on space platforms and the amount of mass that can be launched into space has always been very limited and is expected to continue to be very limited. This paper describes an x-ray optics development program and reports on its status that meets these two requirements. The objective of this program is to enable Explorer type missions in the near term and to enable flagship missions in the long term

    Pro- and anti-tumour activities of CD146/MCAM in breast cancer result from its heterogeneous expression and association with epithelial to mesenchymal transition

    CD146, also known as melanoma cell adhesion molecule (MCAM), is expressed in numerous cancers and has been implicated in the regulation of metastasis. We show that CD146 negatively regulates transendothelial migration (TEM) in breast cancer. This inhibitory activity is reflected by a reduction in MCAM gene expression and increased promoter methylation in tumour tissue compared to normal breast tissue. However, increased CD146/MCAM expression is associated with poor prognosis in breast cancer, a characteristic that is difficult to reconcile with inhibition of TEM by CD146 and its epigenetic silencing. Single cell transcriptome data revealed MCAM expression in multiple cell types, including the malignant cells, tumour vasculature and normal epithelium. MCAM expressing malignant cells were in the minority and expression was associated with epithelial to mesenchymal transition (EMT). Furthermore, gene expression signatures defining invasiveness and a stem cell-like phenotype were most strongly associated with mesenchymal-like tumour cells with low levels of MCAM mRNA, likely to represent a hybrid epithelial/mesenchymal (E/M) state. Our results show that high levels of MCAM gene expression are associated with poor prognosis in breast cancer because they reflect tumour vascularisation and high levels of EMT. We suggest that high levels of mesenchymal-like malignant cells reflect large populations of hybrid E/M cells and that low CD146 expression on these hybrid cells is permissive for TEM, aiding metastasis
