    Accelerated Innovation Development of Laser Metrology for Steel Bridge Fabrication

    This report documents the use of a laser metrology technique in the production of splice connections for steel bridge girders which was successfully performed at Hirschfeld Industries steel bridge fabrication plant in Bristol, VA. Over the course of this project, the traditional method of splice connection fabrication was studied and compared alongside a newer method involving the use of laser metrology to enable the use of pre-drilled girder splice connection holes and custom fabricated splice plates. The primary benefit of this new method is that time consuming match drilling of girder splice connections is replaced with automated drilling methods. This new technique was demonstrated on girders that were fabricated for use in a new bridge being built in Dandridge, Tennessee. In addition to documenting the technique and its implementation, this report examines various aspects of the technology used, costs and benefits, possible sources of error, and potential uses and extensions of the technique in the future. Based on the findings in this report and the implementation documented, it is clear that the laser metrology technique studied can successfully be implemented in steel bridge fabrication; furthermore, this technique has great potential to provide significant time, money, and space savings in the girder fabrication process.     Incorporating lakes within the river discontinuum: longitudinal changes in ecological characteristics in streamlake networks

    Abstract: Lakes and rivers are intimately connected in an alternating series of lentic and lotic reaches in many regions. The study of lakes and their outlets in hierarchical and branching river networks has not gained the attention of stream ecologists, and little effort has been focused on synthesizing the ecology of lake-stream interactions within a drainage network. Rapid and predictable changes in the ecological characteristics of streams occur at the interface with lakes. The influence that a lake might have on a stream is dependent on its position within the stream, stream type and size, lake size and shape, and the inlet and outlet positions. Little is known about the influences of multiple lakes within stream-lake networks and how these influences are determined by network shape and pattern. Fruitful collaborations and novel insights will come from the combined efforts of limnologists, stream ecologists, and landscape ecologists. Geographic information systems and network analyses will play an important role in summarizing aquatic landscape characteristics and creating a predictive science of aquatic networks. Lakes need to be more explicitly incorporated into ecological concepts in stream ecology, and reciprocally, streams need to be incorporated into ecological concepts involving lakes for the successful management and conservation of our aquatic resources

    Spa47 is an oligomerization-activated type three secretion system (T3SS) ATPase from \u3cem\u3eShigella flexneri\u3c/em\u3e

    Gram-negative pathogens often use conserved type three secretion systems (T3SS) for virulence. The Shigella type three secretion apparatus (T3SA) penetrates the host cell membrane and provides a unidirectional conduit for injection of effectors into host cells. The protein Spa47 localizes to the base of the apparatus and is speculated to be an ATPase that provides the energy for T3SA formation and secretion. Here, we developed an expression and purification protocol, producing active Spa47 and providing the first direct evidence that Spa47 is a bona fide ATPase. Additionally, size exclusion chromatography and analytical ultracentrifugation identified multiple oligomeric species of Spa47 with the largest greater than 8 fold more active for ATP hydrolysis than the monomer. An ATPase inactive Spa47 point mutant was then engineered by targeting a conserved Lysine within the predicted Walker A motif of Spa47. Interestingly, the mutant maintained a similar oligomerization pattern as active Spa47, but was unable to restore invasion phenotype when used to complement a spa47 null S. flexneri strain. Together, these results identify Spa47 as a Shigella T3SS ATPase and suggest that its activity is linked to oligomerization, perhaps as a regulatory mechanism as seen in some related pathogens. Additionally, Spa47 catalyzed ATP hydrolysis appears to be essential for host cell invasion, providing a strong platform for additional studies dissecting its role in virulence and providing an attractive target for anti-infective agents

    The differential associations of positive and negative symptoms with suicidality

    BACKGROUND: Suicide is one of the leading causes of death in people with schizophrenia. Identifying risk factors for suicide in schizophrenia is therefore an important clinical and research priority. METHOD: A cross-sectional secondary analysis was conducted on the DNA Polymorphisms in Mental Illness Study (DPIM) data. Suicidality data was extracted, and the number of positive and negative symptoms were established for a total of 1494 participants. Logistic and negative binomial regression analyses were conducted to assess for associations between positive or negative symptoms and suicidal ideation, attempt, or number of attempts, whilst adjusting for potential confounders. RESULTS: Negative symptoms were associated with a reduction in the risk of suicidal ideation (odds ratio [OR]: 0.83; 95 % CI: 0.75-0.91) and suicide attempt (OR: 0.79; 95 % CI: 0.71-0.88) after adjusting for age and sex. Positive symptoms were associated with an increased risk of suicidal ideation (OR: 1.06; 95 % CI: 1.03-1.09), suicide attempt (OR: 1.04; 95 % CI: 1.00-1.07) and number of suicide attempts (incidence rate ratio [IRR]: 1.05; 95 % CI: 1.01-1.08). Further adjusting for depressive symptoms slightly increased the magnitude of associations with negative symptoms but attenuated associations between positive symptoms and suicidality to the null. CONCLUSIONS: Negative symptoms are associated with a reduced risk of suicidality, whilst positive symptoms are associated with an increased risk of suicidality. Depressive symptoms may confound or mediate these associations

    Ecological Characteristics of Streams in the Barrenlands near Lac de Gras, N.W.T., Canada

    We examined spatiotemporal variation in the physical, chemical, and biological characteristics of pristine streams that represent a range of conditions near Lac de Gras in the Barrenlands region of the Northwest Territories, Canada. Principal component analysis organized streams into four groups on the basis of seven physical characteristics. Despite broad differences among groups in physical characteristics, variation in chemical and biological characteristics was generally not large, with only pH and coarse particulate organic matter differing among the four groups. Nevertheless, several chemical and biological variables were correlated with physical characteristics, particularly measures of stream size (bankfull width and depth, drainage area, and stream discharge). Annual variability in climate affected stream temperature and discharge and influenced several biotic characteristics, particularly the growth of young-of-the-year arctic grayling (Thymallus arcticus). Barrenland streams share basic characteristics of Alaskan tundra streams, as a result of similar climatic regimes. Key differences between the two areas, however, appear related to the lake-outlet nature of the Barrenland streams, which may contribute to higher growth of young-of-the-year arctic grayling than would be expected from regional climate.On a étudié la variation spatio-temporelle dans les caractéristiques physiques, chimiques et biologiques de cours d'eau vierges qui témoignent d'une gamme de conditions près du lac de Gras dans la région des Barrenlands, dans les Territoires du Nord-Ouest, au Canada. L'analyse des composantes principales a divisé les cours d'eau en quatre groupes, d'après sept caractéristiques physiques. En dépit de différences marquées dans ces dernières entre les groupes, la variation dans les attributs chimiques et biologiques n'était généralement pas très grande, avec seulement le pH et les grosses particules organiques qui montraient des différences entre les quatre groupes. Plusieurs variables chimiques et biologiques étaient néanmoins corrélées avec les attributs physiques, en particulier les mesures de la taille des cours d'eau (largeur et profondeur du débordement, aire de drainage et débit). La variabilité annuelle du climat avait une incidence sur la température et le débit des cours d'eau et elle influençait plusieurs caractéristiques biotiques, en particulier la croissance des jeunes de l'année chez l'ombre arctique. Les cours d'eau des Barrenlands ont en commun les caractéristiques fondamentales des cours d'eau de la toundra alaskienne, vu qu'ils sont soumis à des régimes climatiques similaires. Les différences majeures entre les deux régions semblent cependant liées au fait que la décharge des cours d'eau des Barrenlands se fait dans un lac, ce qui pourrait contribuer à une croissance des jeunes de l'année chez l'ombre arctique plus forte que le climat régional ne le laisserait croire

    Unified Total Synthesis of the Brevianamide Alkaloids Enabled by Chemical Investigations into their Biosynthesis

    The bicyclo[2.2.2]diazaoctane alkaloids are a vast group of natural products which have been the focus of attention from the scientific community for several decades. This interest stems from their broad range of biological activities, their diverse biosynthetic origins, and their topologically complex structures, which combined make them enticing targets for chemical synthesis. In this article, full details of our synthetic studies into the chemical feasibility of a proposed network of biosynthetic pathways towards the brevianamide family of bicyclo[2.2.2]diazaoctane alkaloids are disclosed. Insights into issues of reactivity and selectivity in the biosynthesis of these structures have aided the development of a unified biomimetic synthetic strategy, which has resulted in the total synthesis of all known bicyclo[2.2.2]diazaoctane brevianamides and the anticipation of an as-yet-undiscovered congener

    Adding a Brane to the Brane-Anti-Brane Action in BSFT

    We attempt to generalize the effective action for the D-brane-anti-D-brane system obtained from boundary superstring field theory (BSFT) by adding an extra D-brane to it to obtain a covariantized action for 2 D-branes and 1 anti-D-brane. We discuss the approximations made to obtain the effective action in closed form. Among other properties, this effective action admits solitonic solutions of codimension 2 (vortices) when one of the D-brane is far separated from the brane-anti-brane pair.Comment: 23 pages, 2 figures, minor revision

    DEVELOPMENT OF RECOMBINANT PROTEIN BASED CHEMICAL CONJUGATE MALARIA VACCINES TARGETING THE PRE-ERYTHROCYTIC STAGE, TRANSMISSION BLOCKING, OR BOTH

    The development of a Plasmodium falciparum malaria vaccine is critical for future control and elimination programs. Recombinant protein based chemical conjugate vaccines, covering different parasite stages, are being developed due to complexity of the parasite and sub-optimal immunogenicity of recombinant malaria proteins in humans, respectively. Chemical conjugation of recombinant malaria proteins to carrier proteins improves their immunogenicity in animal studies. A transmission blocking vaccine comprised of the ookinete protein Pfs25 chemically conjugated to Pseudomonas aeruginosa ExoProtein A (EPA) is currently being developed for pilot scale cGMP production. Bulk lots of Pfs25 and EPA have already been produced and released following cGMP. Appropriate analytical assays are being evaluated for both in-process and bulk release of the Pfs25-EPA conjugate vaccine. One critical assay already evaluated for determining the average mass is analytical size exclusion HPLC coupled with multi-angle light scattering. Phase 1 human clinical trials are planned for 2011. Another biological target of interest is the circumsporozoite protein (CSP), the most abundant and immunogenic protein on the surface of the sporozoite. A recombinant nearly full-length CSP has been produced in the methylotrophic yeast Pichia pastoris containing a bioengineered free thiol for chemical conjugation to various carrier proteins, including a chemically conjugated form of Pfs25. This recombinant protein based chemical conjugation platform, in combination with better adjuvant selection, will improve the potential for developing an efficacious malaria vaccine

    Canonical explorations of 'Tel' environments for computer programming

    This paper applies a novel technique of canonical gradient analysis, pioneered in ecological sciences, with the aim of exploring student performance and behaviours (such as communication and collaboration) while undertaking formative and summative tasks in technology enhanced learning (TEL) environments for computer programming. The research emphasis is, therefore, on revealing complex patterns, trends, tacit communications and technology interactions associated with a particular type of learning environment, rather than the testing of discrete hypotheses. The study is based on observations of first year programming modules in BSc Computing and closely related joint-honours with software engineering, web and game development courses. This research extends earlier work, and evaluates the suitability of canonical approaches for exploring complex dimensional gradients represented by multivariate and technology-enhanced learning environments. The advancements represented here are: (1) an extended context, beyond the use of the ‘Ceebot’ learning platform, to include learning-achievement following advanced instruction using an industrystandard integrated development environment, or IDE, for engineering software; and (2) longitudinal comparison of consistency of findings across cohort years. Direct findings (from analyses based on code tests, module assessment and questionnaire surveys) reveal overall engagement with and high acceptance of collaborative working and of the TEL environments used, but an inconsistent relationship between deeply learned programming skills and module performance. The paper also discusses research findings in the contexts of established and emerging teaching practices for computer programming, as well as government policies and commercial requirements for improved capacity in computer-science related industries
