    Hydrogen Cyanamide Causes Reversible G2/M Cell Cycle Arrest Accompanied by Oxidation of the Nucleus and Cytosol

    Hydrogen cyanamide (HC) has been widely used in horticulture to trigger bud burst following dormancy. Its use has been banned in some countries due to human health concerns, however the search for effective safe alternatives is delayed by lack of knowledge of the mechanism of HC action. Earlier studies demonstrate that HC stimulates the production of reactive oxygen species (ROS) and alters the rate of cell division. However, the relationships between HC effects on ROS, redox (reduction/oxidation) homeostasis and cell division are unknown. This study used Arabidopsis thaliana ((L.) Heynh.) seedlings expressing the redox reporter roGFP2 to measure the oxidation states of the nuclei and cytosol in response to HC treatment. The Cytrap dual cell cycle phase marker system and flow cytometry were used to study associated changes in cell proliferation. HC (1.5 mM) reversibly inhibited root growth during a 24 h treatment. Higher concentrations were not reversible. HC did not synchronise the cell cycle, in contrast to hydroxyurea. Rather, HC caused a gradual accumulation of cells in the G2/M phase and decline of G1/S phase cells, 16 to 24 h post-treatment. This was accompanied by increased oxidation of both the nuclei and cytosol. Taken together, these findings show that HC impairs proliferation of embryonic root meristem cells in a reversible manner through restriction of G2/M transition accompanied by increased cellular oxidation

    Participatory GIS as a Tool for Stakeholder Engagement in Building Resilience to Sea Level Rise: A Demonstration Project

    This article describes a participatory geographical information system (PGIS) demonstration project used as part of the stakeholder engagement efforts undertaken by the Citizen Engagement Working Group of the Hampton Roads Sea Level Rise Preparedness and Resilience Intergovernmental Planning Pilot Project. The PGIS demonstration project was conducted in the Little Creek/Pretty Lake case study area in the Hampton Roads region of southeastern coastal Virginia. PGIS served as a deliberative and participatory mechanism to obtain local knowledge from residents about the location of valued assets within the community and locations challenged by increased flooding and sea level rise. The PGIS application, using the weTable tool, was found to be useful for soliciting and documenting local knowledge, such as by highlighting community assets and identifying community challenges. It was also found to be useful for facilitating community-wide discussion, visualizing the problem, and understanding the severity of sea level rise and flooding. The PGIS demonstration project showed how participatory mapping can directly engage residents in creating sociospatial data, build knowledge, and foster learning and deliberation in a complex issue such as resilience to flooding and sea level rise

    Developing an Institutional Arrangement for a Whole-of-Government and Whole-of-Community Approach to Regional Adaptation to Sea Level Rise: The Hampton Roads Pilot Project

    Adaptation to sea level rise (SLR) requires coordination among local, state, and federal entities and collaboration across governments, nonprofits, businesses, and residents. This coordination and collaboration are reflected in institutional arrangements associated with a whole-of-government and whole-of-community approach to regional adaptation. This study analyzes the development of an interlocal agreement (ILA), the Hampton Roads Sea Level Rise Preparedness and Resilience Intergovernmental Planning Pilot Project (the Pilot Project), as an example of such an arrangement. This study assesses how factors throughout three phases of ILA development (initiation, implementation, and execution) influence outcomes and effectiveness. Drawing upon participant observation, document analysis, survey of participants, and interviews with key informants this study identifies factors that facilitate effective regional adaptation to SLR (impetus, agreement) and factors that hamper adaptation efforts (funding, ease of delivery), and offers insight into the complexities of institutional collective action to address contentious and challenging issues such as SLR

    Weight loss in individuals with metabolic syndrome given DASH diet counseling when provided a low sodium vegetable juice: a randomized controlled trial

    <p>Abstract</p> <p>Background</p> <p>Metabolic syndrome, a constellation of metabolic risk factors for type 2 diabetes and cardiovascular disease, is one of the fastest growing disease entities in the world. Weight loss is thought to be a key to improving all aspects of metabolic syndrome. Research studies have suggested benefits from diets rich in vegetables and fruits in helping individuals reach and achieve healthy weights.</p> <p>Objective</p> <p>To evaluate the effects of a ready to serve vegetable juice as part of a calorie-appropriate Dietary Approaches to Stop Hypertension (DASH) diet in an ethnically diverse population of people with Metabolic Syndrome on weight loss and their ability to meet vegetable intake recommendations, and on their clinical characteristics of metabolic syndrome (waist circumference, triglycerides, HDL, fasting blood glucose and blood pressure).</p> <p>A secondary goal was to examine the impact of the vegetable juice on associated parameters, including leptin, vascular adhesion markers, and markers of the oxidative defense system and of oxidative stress.</p> <p>Methods</p> <p>A prospective 12 week, 3 group (0, 8, or 16 fluid ounces of low sodium vegetable juice) parallel arm randomized controlled trial. Participants were requested to limit their calorie intake to 1600 kcals for women and 1800 kcals for men and were educated on the DASH diet. A total of 81 (22 men & 59 women) participants with Metabolic Syndrome were enrolled into the study. Dietary nutrient and vegetable intake, weight, height, leptin, metabolic syndrome clinical characteristics and related markers of endothelial and cardiovascular health were measured at baseline, 6-, and 12-weeks.</p> <p>Results</p> <p>There were significant group by time interactions when aggregating both groups consuming vegetable juice (8 or 16 fluid ounces daily). Those consuming juice lost more weight, consumed more Vitamin C, potassium, and dietary vegetables than individuals who were in the group that only received diet counseling (p < 0.05).</p> <p>Conclusion</p> <p>The incorporation of vegetable juice into the daily diet can be a simple and effective way to increase the number of daily vegetable servings. Data from this study also suggest the potential of using a low sodium vegetable juice in conjunction with a calorie restricted diet to aid in weight loss in overweight individuals with metabolic syndrome.</p

    Finishing the euchromatic sequence of the human genome

    The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∼99% of the euchromatic genome and is accurate to an error rate of ∼1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead

    Assessment of the impact of changes in transit systems using intervention analysis

    Marketing control enables effective and efficient use of marketing resources. This study uses intervention analysis to assess the effectiveness of a transit system change in Chattanooga, Tennessee. The goal of the change was to improve the effectiveness of the transit system, although, "effectiveness" was not formally defined before the change. In this study, four measures of effectiveness were used: total ridership, total operating revenues, the ratio of total operating revenues to total revenue miles, and the ratio of total passenger trips to total revenue miles. Intervention analysis showed that three out of four of these measures had been improved by the transit system change.
