    Solving second kind integral equations by Galerkin methods with continuous orthogonal wavelets

    AbstractIn this paper, We use the continuous wavelets on the interval constructed by Cohen et al. (Appl. Comput. Harm. Anal. 1 (1993) 54–81) to solve the second kind integral equations. To this end, we give the decomposition and reconstruction algorithm for these wavelets, and construct the quadrature formulae for the calculation of inner products of any functions and the scaling functions, which are required in the wavelet-Galerkin methods for integral equations. In this method, the integral kernels are represented in these wavelet bases as sparse matrices, to high precision. Thus, we present an efficient algorithm for numerical solution of second kind integral equations

    Alkylating HIV-1 Nef - a potential way of HIV intervention

    <p>Abstract</p> <p>Background</p> <p>Nef is a 27 KDa HIV-1 accessory protein. It downregulates CD4 from infected cell surface, a mechanism critical for efficient viral replication and pathogenicity. Agents that antagonize the Nef-mediated CD4 downregulation may offer a new class of drug to combat HIV infection and disease. TPCK (N-α-p-tosyl-L-phenylalanine chloromethyl ketone) and TLCK (N-α-p-tosyl-L-lysine chloromethyl ketone) are alkylation reagents that chemically modify the side chain of His or Cys residues in a protein. In search of chemicals that inhibit Nef function, we discovered that TPCK and TLCK alkylated HIV Nef.</p> <p>Methods</p> <p>Nef modification by TPCK was demonstrated on reducing SDS-PAGE. The specific cysteine residues modified were determined by site-directed mutagenesis and mass spectrometry (MS). The effect of TPCK modification on Nef-CD4 interaction was studied using fluorescence titration of a synthetic CD4 tail peptide with recombinant Nef-His protein. The conformational change of Nef-His protein upon TPCK-modification was monitored using CD spectrometry</p> <p>Results</p> <p>Incubation of Nef-transfected T cells, or recombinant Nef-His protein, with TPCK resulted in mobility shift of Nef on SDS-PAGE. Mutagenesis analysis indicated that the modification occurred at Cys55 and Cys206 in Nef. Mass spectrometry demonstrated that the modification was a covalent attachment (alkylation) of TPCK at Cys55 and Cys206. Cys55 is next to the CD4 binding motif (A<sub>56</sub>W<sub>57</sub>L<sub>58</sub>) in Nef required for Nef-mediated CD4 downregulation and for AIDS development. This implies that the addition of a bulky TPCK molecule to Nef at Cys55 would impair Nef function and reduce HIV pathogenicity. As expected, Cys55 modification reduced the strength of the interaction between Nef-His and CD4 tail peptide by 50%.</p> <p>Conclusions</p> <p>Our data suggest that this Cys55-specific alkylation mechanism may be exploited to develop a new class of anti HIV drugs.</p

    An Optimization Model Based on Electric Power Generation in Steel Industry

    Electric power is an important energy in steel industry. Electricity accounts for roughly 20% to 30% of the gross energy consumption and costs about 10% of the gross cost of energy. In this paper, under the premise of ensuring the stability of energy supply and the normal production safety, the mathematical programming method and the dynamic mathematical optimization model were used to set up the surplus gas in the optimal allocation among the buffer users and steam production dispatching for the production equipment. The application of this optimization model can effectively improve the energy efficiency and the accuracy of power generation, making full use of secondary energy and residual heat. It also can realize the rationalization of the electricity production structure optimization which can effectively reduce the flare of the gas and steam on one hand, and save energy and decrease production cost on the other

    First complete mitochondrial genome of the tribe Coccini (Hemiptera, Coccomorpha, Coccidae) and its phylogenetic implications

    Soft scale insects (Hemiptera, Coccidae) are important pests of various agricultural and horticultural crops and ornamental plants. They have negative impacts on agriculture and forestry. The tribe Coccini represents one of the most ancient evolutionary lineages of soft scale insects. However, no complete Coccini mitochondrial genome (mitogenome) is available in public databases. Here, we described the complete mitogenome of Coccus hesperidum L., 1758. The 15,566 bp mitogenome of C. hesperidum had a high A+T content (83.4%) and contained a typical set of 37 genes, with 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs) and two ribosomal RNA genes (rRNAs). Only seven tRNAs had the typical clover-leaf secondary structure and the remaining tRNAs lacked the DHU arm, TψC arm or both. Moreover, a comparative analysis of all reported scale insect mitogenomes from GenBank database was performed. The mitogenomes of scale insects showed high similarities in base composition and A+T content. Additionally, our phylogenetic analysis confirmed the monophyly of Coccomorpha and revealed that the archaeococcoids were the most basal lineage within Coccomorpha, while Ericerus pela and Didesmococcus koreanus, belonging to Coccidae, were often mixed with Aclerdidae, making Coccidae a paraphyletic group. These findings expand the mitogenome database of scale insects and provide new insights on mitogenome evolution for future studies across different insect groups

    Clinical Study CEUS Helps to Rerate Small Breast Tumors of BI-RADS Category 3 and Category 4

    . Purpose. The primary aim of this study was to explore if classification, whether using the BI-RADS categories based on CEUS or conventional ultrasound, was conducive to the identification of benign and malignant category 3 or 4 small breast lesions. Material and Methods. We evaluated 30 malignant and 77 benign small breast lesions using CEUS. The range of enhancement, type of enhancement strength, intensity of enhancement, and enhancement patterns were independent factors included to assess the BI-RADS categories. Results. Of the nonenhanced breast lesions, 97.8% (44/45) were malignant, while, of the hyperplasic nodules, 96.8% (30/31) showed no enhancement in our study. Category changes of the lesions were made according to the features determined using CEUS. The results showed that these features could improve diagnostic sensitivity (from 70.0 to 80.0, 80.0, 90.0, and 90.0%), reduce the negative likelihood ratio (from 0.33 to 0.22, 0.25, 0.11, and 0.12), and improve the NPV (from 88.8 to 92.2, 91.2, 96.2, and 95.5%). However, this was not conducive to improve diagnostic specificity or the PPV. Conclusion. The vast majority of nonenhanced small breast lesions were malignant and most of the hyperplasic nodules showed no contrast enhancement. As a reference, CEUS was helpful in identifying BI-RADS category 3 or 4 small breast lesions

    Strain Analysis of GaN HEMTs on (111) Silicon with Two Transitional AlxGa1-xN Layers

    We have designed and then grown a simple structure for high electron mobility transistors (HEMTs) on silicon, where as usual two transitional layers of AlxGa1−xN (x = 0.35, x = 0.17) have been used in order to engineer the induced strain as a result of the large lattice mismatch and large thermal expansion coefficient difference between GaN and silicon. Detailed x-ray reciprocal space mapping (RSM) measurements have been taken in order to study the strain, along with cross-section scanning electron microscope (SEM) images and x-ray diffraction (XRD) curve measurements. It has been found that it is critical to achieve a crack-free GaN HEMT epi-wafer with high crystal quality by obtaining a high quality AlN buffer, and then tuning the proper thickness and aluminium composition of the two transitional AlxGa1−xN layers. Finally, HEMTs with high performance that are fabricated on the epi-wafer have been demonstrated to confirm the success of our strain engineering and above analysis

    Practice on the Watershed Hydrological Experimental System Reconciling Deterministic and Stochastic Subjects Based on the System Complexity: 1. Theoretical Study

    This is the first of a two-part series on the watershed hydrological experimental system (WHES). Since the foundational stage and developmental stage of hydrological basin study with a duration of more than ca. one century, facing with the changing environment and, the declined risk of field study while the catchment hydrology is trapped in a theoretical impasse, a third phase of renovation on hydrological experiments seems ready to come out inevitably. Learned from Chinese decades’ experiences on the field basin study for the question of what is wrong with the status quo, our exploratory idea is reported in this part. From the viewpoint of general system theory based on the paralleled concepts of the ancient Chinese and the Western, it is considered that the adequate method should face the characters of the complex dynamic system instead of previous static, linear system. From the viewpoint of another philosophical paralleled concept of the Middle Way, it should also face the operation and organizing of the mesoscopic systems for the organized complexity. Then, a framework of WHES is suggested with its organization based on the strategy of constrain complexity and add complexity and on the strategy of manipulation including the artificial-natural and controlled-natural objects. Such a trial framework, the Chuzhou WHES, is reported including the suggested critical zone experimental block (CZEB) instead of the experimental basin (EB) in the last decades

    This is the second of a two-part series on the watershed hydrological experimental system (WHES) aimed at practice and test of it at Chuzhou Hydrology Laboratory. It constitutes both natural and artificial entities of different scales, within which two typical main subjects are reviewed here. First is a natural watershed Nandadish, which is subjected to be a Critical Zone Experimental Block, under manipulation strategy of constrain complexity to compare with the pure natural watersheds, it is the controlled-natural as we termed. Second is an artificial catchment Hydrohill, under the strategy of add complexity to compare with the simple artificial lysimeters, it is the artificial-natural as we termed. The constructions and instrumentations of these experimental catchments are reviewed, especially their renovation version during recent years after a long abandonment. Some results get during the operation of Chuzhou WHES are outlined here as well
