    CoTDet: Affordance Knowledge Prompting for Task Driven Object Detection

    Task driven object detection aims to detect object instances suitable for affording a task in an image. Its challenge lies in object categories available for the task being too diverse to be limited to a closed set of object vocabulary for traditional object detection. Simply mapping categories and visual features of common objects to the task cannot address the challenge. In this paper, we propose to explore fundamental affordances rather than object categories, i.e., common attributes that enable different objects to accomplish the same task. Moreover, we propose a novel multi-level chain-of-thought prompting (MLCoT) to extract the affordance knowledge from large language models, which contains multi-level reasoning steps from task to object examples to essential visual attributes with rationales. Furthermore, to fully exploit knowledge to benefit object recognition and localization, we propose a knowledge-conditional detection framework, namely CoTDet. It conditions the detector from the knowledge to generate object queries and regress boxes. Experimental results demonstrate that our CoTDet outperforms state-of-the-art methods consistently and significantly (+15.6 box AP and +14.8 mask AP) and can generate rationales for why objects are detected to afford the task.Comment: Accepted by ICCV 202

    Active Q-switched X-Ray Regenerative Amplifier Free-Electron Laser

    Despite tremendous progress in x-ray free-electron laser (FEL) science over the last decade, future applications still demand fully coherent, stable x-rays that have not been demonstrated in existing X-ray FEL facilities. In this Letter, we describe an active Q-switched x-ray regenerative amplifier FEL scheme to produce fully coherent, high-brightness, hard x rays at a high-repetition rate. By using simple electron-beam phase space manipulation, we show this scheme is flexible in controlling the x-ray cavity quality factor Q and hence the output radiation. We report both theoretical and numerical studies on this scheme with a wide range of accelerator, x-ray cavity, and undulator parameters

    Peptide 17, an inhibitor of YAP/TEAD4 pathway, mitigates lung cancer malignancy

    Purpose: To investigate whether and how peptide 17 affects lung cancer cells.Methods: Human lung carcinoma cells, LLC and PC-9, were employed to study the therapeutic effect of peptide 17 on lung cancer. After exogenous expression of peptide 17, a co-immunoprecipitation experiment was used to examine the inhibitory effect of peptide 17. CCK8 assay was employed to assess the lung cancer cells’ viability while clone formation assays were used to assess lung cancer cell proliferation. Colony number was also determined. The stimulatory effect of peptide 17 on lung cancer cell apoptosis was assessed by fluorescence-activated cell sorting (FACS).Results: Peptide 17 efficiently disrupted the interaction between YAP and TEAD4 (p < 0.001), and decreased the expression of CTGF and Cyr61. In addition, lung cancer cell viability and proliferation significantly decreased (p < 0.001) in a time- and concentration-dependent manner. On the other hand, the proportion of apoptotic cells was significantly elevated with rising concentration of peptide 17.Conclusion: Exogenous expression of peptide 17 activates Bcl2/Bax/caspase-9 signal and isresponsible for its inhibitory effects on lung cancer cells.     Nonlinearly Shaped Pulses in Photoinjectors and Free-Electron Lasers

    Photoinjectors and Free Electron Lasers (FEL) are amongst the most advanced systems in accelerator physics and have consistently pushed the boundaries of emittance and x-ray peak power. In this paper, laser shaping at the cathode is proposed to further lower the emittance and reduce electron beam tails, which would result in brighter x-ray production. Using dispersion controlled nonlinear shaping (DCNS), laser pulses and beam dynamics were simulated in LCLS-II. The photoinjector emittance was optimized and the resulting e-beam profiles were then simulated and optimized in the linac. Finally, the expected FEL performance is estimated and compared to the current technology: Gaussian laser pulses on the cathode. The e-beams produced by DCNS pulses show a potential for 35% increase in x-ray power per pulse during SASE when compared to the standard Gaussian laser pulses

    The association between sleep and early pubertal development in Chinese children: a school population-based cross-sectional study

    BackgroundThere is an increasing tendency toward early pubertal development, and sleep might be related to pubertal onset. We aimed to investigate the association of sleep duration and bedtime with early pubertal development.MethodsThis was a cross-sectional study of 8,007 children (53.6% boys) from Qufu city, Shandong province and Zhongshan city, Guangdong province, China. Data on sleep duration and bedtime were obtained by questionnaire. Early pubertal development was the primary outcome and it was evaluated by the pediatrician according to Tanner staging. Logistic regression models were used to separately examine the association between sleep duration or bedtime and early pubertal development, controlling body mass index (BMI), dietary pattern, soft drink, feeding pattern and mother’s BMI.ResultsIn boys, short sleep duration was strongly related to early pubertal development [OR (95%CI): 4.26 (1.30, 13.94)], and this association was intensified after adjusted BMI, dietary pattern, soft drink, feeding pattern and mother’s BMI. In girls, OR (95%CI) was 1.62 (1.04, 2.51), and increased after controlling BMI. Bedtime was associated with early pubertal development on weekdays [OR (95%CI): 6.39 (1.54, 26.45) in boys and 1.93 (1.23, 3.05) in girls], but not on weekends [OR (95%CI): 2.49 (0.61, 10.21) in boys; 1.31 (0.76, 2.25) in girls].ConclusionThis study underscores the positive association between the risk of early pubertal development and insufficient sleep duration and late bedtime

    Pediatric infection with the Omicron variant increases the risks of febrile seizures among COVID-19 infected children

    BackgroundThe Omicron variant of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is less likely to cause severe disease in children than the other variants but has become an increasing cause of febrile seizures (FS) among children. In this case-control study, we aimed to examine the risk factors associated with FS in children infected with the COVID-19 Omicron variant and related treatment modalities.MethodsThis retrospective case-control study includes 113 subjects infected with the COVID-19 Omicron variant, grouped into 45 cases (those with FS) and 68 controls (those without FS). Data on clinical features, laboratory parameters, and treatment modalities were collected and analyzed.ResultsApproximately 5.74% of COVID-19 infected children developed COVID-19-associated FS. Children with COVID-19 and high body temperatures [RR 1.474; (95% CI: 1.196–1.818), p < 0.001], previous history of FS [RR 1.421; (95% CI: 1.088–1.855), p = 0.010], high procalcitonin levels [RR 1.140; (95% CI: 1.043–1.246), p = 0.048] and high neutrophil counts [RR 1.015; (95% CI: 1.000–1.029), p = 0.048] were more likely to experience FS than the controls. In contrast, children with COVID-19 and low eosinophil counts, low hemoglobin levels, and cough had a lower risk of developing FS [RR 0.494; (95% CI: 0.311–0.783), p = 0.003], [RR 0.979; (95% CI: 0.959–0.999), p = 0.044]; and [RR 0.473 (95% CI 0.252–0.890), p = 0.020]; respectively. Children with FS received more anti-flu medications than those without.ConclusionA significant increase in FS was observed in children with Omicron SARS-CoV-2 infection. A higher body temperature, a history of FS, a higher procalcitonin level, and a high neutrophil count were all associated with an increased risk of FS in children with COVID-19.     ObjectiveTo investigate the effect of social support on stress, and to clarify the effect and mechanism of Online Emotional Support Accompany Group (OESAG).MethodsThe group members who signed up for the public welfare project “Psychological Rehabilitation Group Psychological Service under the COVID-19 Pandemic” were divided into the treatment group, the control group, and the blank group with 37 members each. The treatment group received OESAG intervention, the control group received online time management group intervention, and the blank group was the waiting group. The three groups of subjects were synchronously tested before and after the intervention group.ResultsAfter the OESAG intervention, compared with the control group and the blank group, the treatment group showed that perceived social support was improved, and loneliness and stress were decreased.ConclusionImproving social support can effectively reduce stress. OESAG can effectively improve social support and so too decrease stress. This study could help in designing effective psychological intervention measures to reduce the degree of stress symptoms and enhance both personal and social levels of coping with stressful events

    LncRNA BASP1-AS1 interacts with YBX1 to regulate Notch transcription and drives the malignancy of melanoma

    Melanoma is a fatal skin malignant tumor with a poor prognosis. We found that long noncoding RNA BASP1-AS1 is essential for the development and prognosis of melanoma. The methylation, RNA sequencing, copy number variation, mutation data, and sample follow-up information of melanoma from The Cancer Genome Atlas (TCGA) were analyzed using weighted gene co-expression network analysis and 366 samples common to the three omics were selected for multigroup clustering analysis. A four-gene prognostic model (BASP1-AS1, LOC100506098, ARHGAP27P1, and LINC01532) was constructed in the TCGA cohort and validated using the GSE65904 series. The expression of BASP1-AS1 was upregulated in melanoma tissues and various melanoma cell lines. Functionally, the ectopic expression of BASP1-AS1 promoted cell proliferation, migration, and invasion in both A375 and SK-MEL-2 cells. Mechanically, BASP1-AS1 interacted with YBX1 and recruited it to the promoter of NOTCH3, initiating its transcription process. The activation of the Notch signaling then resulted in the transcription of multiple oncogenes, including c-MYC, PCNA, and CDK4, which contributed to melanoma progression. Thus, BASP1-AS1 could act as a potential biomarker for cutaneous malignant melanoma

    CD23 expression in mantle cell lymphoma is associated with CD200 expression, leukemic non-nodal form, and a better prognosis

    Mantle cell lymphoma (MCL) is usually CD23 negative, a feature helpful in distinguishing MCL from chronic lymphocytic leukemia/small lymphocytic lymphoma. However, a subset of MCL cases can be CD23+. Limited data are available regarding the clinicopathological features and prognosis of patients with CD23+ MCL. In this study, we reviewed 798 cases of MCL and identified 103 (13%) that were CD23+ by flow cytometry, all of which were positive for cyclin D1 and/or associated with CCND1/IGH. In all cases of CD23+ MCL, CD23 expression was dim partial or dim, unlike moderate to bright CD23 expression observed in chronic lymphocytic leukemia/small lymphocytic lymphoma. The clinicopathological features and outcome of patients with CD23+ MCL were compared with 240 patients with typical MCL negative for CD23. Patients with CD23+ MCL more often had an elevated leukocyte count (33% versus 18%, P = .009), bone marrow involvement (89% versus 78%, P = .02), stage 4 disease (87% versus 77%, P = .03), and a leukemic presentation (42% versus 11%, P = .0001). CD23+ MCL was also more often positive for CD200 (17% versus. 4.6%, P = .0005) and less commonly positive for SOX11 (55% versus. 74%, P = .027). All other clinicopathological features were similar. With similar treatment regimens and observation times, patients with CD23+ MCL had a significant better overall survival (P = .02) and progression-free survival (P = .029). In conclusion, CD23 expression was observed in 13% of MCL cases and is associated with a better prognosis in patients with MCL. CD23 is associated with leukocytosis, a leukemic presentation, bone marrow involvement, CD200 expression, and a lower frequency of SOX11 positivity
