    Modeling and numerical analysis of beam matrix plasma display system

    This research investigates a new display device - Beam Matrix Plasma Display Panel (BM PDP). The scan of a PDP in such a system is accomplished through two electrical-beam guns instead of semiconductor switches. The BM-PDP eliminates the expensive semiconductor switches in the current plasma display device systems. Its drive circuit has only three parts: electron beam guns, resistors and capacitors. During the operation of BM PDP, first a switch cell is turned on by a selection gun (X gun). Then another electron gun (Y gun) emits electrons onto column electrodes and capacitors. When the voltage over the corresponding luminous cell reaches its breakdown point, gas discharges and generates light. Drive circuit design and analysis for BM-PDP is an important research topic. This work derives the formulae describing the operation of the drive circuit. With these formulae all the cases in which the drive circuit may work are discussed theoretically and numerically. Two equations are also given to determine the time of cell breakdown in these cases. The results of numerical simulation show that the current of an electron beam gun can be employed to carry the signal of image, the capacitance of a display cell is not sensitive to the initial current of gas discharge. The later property can be used to reduce the difficulties of manufacturing. The process of gas discharge in a display cell is also discussed and a multi-particle physical model is given to simulate the plasma cell

    Algorithms for Computational Genetics Epidemiology

    The most intriguing problems in genetics epidemiology are to predict genetic disease susceptibility and to associate single nucleotide polymorphisms (SNPs) with diseases. In such these studies, it is necessary to resolve the ambiguities in genetic data. The primary obstacle for ambiguity resolution is that the physical methods for separating two haplotypes from an individual genotype (phasing) are too expensive. Although computational haplotype inference is a well-explored problem, high error rates continue to deteriorate association accuracy. Secondly, it is essential to use a small subset of informative SNPs (tag SNPs) accurately representing the rest of the SNPs (tagging). Tagging can achieve budget savings by genotyping only a limited number of SNPs and computationally inferring all other SNPs. Recent successes in high throughput genotyping technologies drastically increase the length of available SNP sequences. This elevates importance of informative SNP selection for compaction of huge genetic data in order to make feasible fine genotype analysis. Finally, even if complete and accurate data is available, it is unclear if common statistical methods can determine the susceptibility of complex diseases. The dissertation explores above computational problems with a variety of methods, including linear algebra, graph theory, linear programming, and greedy methods. The contributions include (1)significant speed-up of popular phasing tools without compromising their quality, (2)stat-of-the-art tagging tools applied to disease association, and (3)graph-based method for disease tagging and predicting disease susceptibility

    Promising molecular mechanisms responsible for gemcitabine resistance in cancer

    Gemcitabine is the first-line treatment for pancreatic ductual adenocarcinoma (PDAC) as well as acts against a wide range of other solid tumors. Patients usually have a good initial response to gemcitabine-based chemotherapy but would eventually develop resistance. To improve survival and prognosis of cancer patients, better understanding of the mechanisms responsible for gemcitabine resistance and discovery of new therapeutic strategies are in great need. Amounting evidence indicate that the developmental pathways, such as Hedgehog (Hh), Wnt and Notch, become reactivated in gemcitabine-resistant cancer cells. Thus, the strategies for targeting these pathways may sensitize cancer cells to gemcitabine treatment. In this review, we will summarize recent development in this area of research and discuss strategies to overcome gemcitabine resistance. Given the cross-talk between these three developmental signaling pathways, designing clinical trials using a cocktail of inhibitory agents targeting all these pathways may be more effective. Ultimately, our hope is that targeting these developmental pathways may be an effective way to improve the gemcitabine treatment outcome in cancer patients

Through pains-taking analyses of genomic profiles in cancer patients, a number of targetable gene alterations have been discovered, with some leading to novel therapies, such as activating mutations of EGFR, BRAF and ALK gene fusions. As a result, clinical management of cancer through targeted therapy has finally become a reality for a subset of cancers, such as lung adenocarcinomas and melanomas. In this review, we summarize how gene mutation discovery leads to new treatment strategies using non-small cell lung cancer (NSCLC) as an example. We also discuss possible future implications of cancer genome analyses

    A YBCO RF-SQUID magnetometer and its applications

    An applicable RF-superconducting quantum interference detector (SQUID) magnetometer was made using a bulk sintered yttrium barium copper oxide (YBCO). The temperature range of the magnetometer is 77 to 300 K and the field range 0 to 0.1T. At 77 K, the equivalent flux noise of the SQUID is 5 x 10 to minus 4 power theta sub o/square root of Hz at the frequency range of 20 to 200 Hz. The experiments show that the SQUID noise at low-frequency end is mainly from 1/f noise. A coil test shows that the magnetic moment sensitivity delta m is 10 to the minus 6th power emu. The RF-SQUID is shielded in a YBCO cylinder with a shielding ability B sub in/B sub ex of about 10 to the minus 6th power when external dc magnetic field is about a few Oe. The magnetometer is successfully used in characterizing superconducting thin films

    The Hedgehog pathway: role in cell differentiation, polarity and proliferation.

    Hedgehog (Hh) is first described as a genetic mutation that has "spiked" phenotype in the cuticles of Drosophila in later 1970s. Since then, Hh signaling has been implicated in regulation of differentiation, proliferation, tissue polarity, stem cell population and carcinogenesis. The first link of Hh signaling to cancer was established through discovery of genetic mutations of Hh receptor gene PTCH1 being responsible for Gorlin syndrome in 1996. It was later shown that Hh signaling is associated with many types of cancer, including skin, leukemia, lung, brain and gastrointestinal cancers. Another important milestone for the Hh research field is the FDA approval for the clinical use of Hh inhibitor Erivedge/Vismodegib for treatment of locally advanced and metastatic basal cell carcinomas. However, recent clinical trials of Hh signaling inhibitors in pancreatic, colon and ovarian cancer all failed, indicating a real need for further understanding of Hh signaling in cancer.
