    In recent years, in order to adapt to the rapid development of the warehouse-storage sales industry and to solve the problems of location cost and efficiency and optimization of the methods of the new retail store, we have integrated and innovated the barycenter method and grey correlation method, and analyzed the grey correlation method with the weight obtained by the comprehensive analysis. In order to achieve the optimal cost effect, we choose the optimal solution from several alternative address schemes. It is found that using the integrated method as the reference standard for the location calculation of Warehouse Logistics Enterprises under the new retail background is helpful to improve the accuracy rate, and reduce the defects and defects caused by the independent use of the various methods, and adapt to the more practical and concrete conditions of the location selection of warehouse storage enterprises.     Cytoplasmic p21 induced by p65 prevents doxorubicin-induced cell death in pancreatic carcinoma cell line

    <p>Abstract</p> <p>Background</p> <p>Studies have shown the existence of p21 induction in a p53-dependent and -independent pathway. Our previous study indicates that DOX-induced p65 is able to bind the p21 promoter to activate its transactivation in the cells.</p> <p>Methods</p> <p>Over-expression and knock-down experiments were performed in Human Pancreatic Carcinoma (PANC1) cells. Cell cycle and cell death related proteins were assessed by Western Blotting. Cytotoxicity assay was checked by CCK-8 kit. Cell growth was analyzed by flow cytometers.</p> <p>Results</p> <p>Here we showed that over-expression of p65 decreased the cytotoxic effect of DOX on PANC1 cells, correlating with increased induction of cytoplasmic p21. We observed that pro-caspase-3 physically associated with cytoplasmic p21, which may be contribution to prevent p21 translocation into the nucleus. Our data also suggested that no clear elevation of nuclear p21 by p65 provides a survival advantage by progression cell cycle after treatment of DOX. Likewise, down-regulation of p65 expression enhanced the cytotoxic effect of DOX, due to a significant decrease of mRNA levels of anti-apoptotic genes, such as the cellular inhibitor of apoptosis-1 (c-IAP1), and the long isoform of B cell leukemia/lymphoma-2 (Bcl-2), leading to efficient induction of caspase-3 cleavage in the cells. More, we present evidence that over-expression of p53 or p53/p65 in the PANC1 cells were more sensitive to DOX treatment, correlated with activation of caspase-3 and clear elevation of nuclear p21 level. Our previous data suggested that expression of p21 increases Gefitinib-induced cell death by blocking the cell cycle at the G1 and G2 phases. The present findings here reinforced this idea by showing p21's ability of potentiality of DOX-induced cell death correlated with its inhibition of cell cycle progression after over-expression of p53 or p53/p65.</p> <p>Conclusion</p> <p>Our data suggested p65 could increase p53-mediated cell death in response to DOX in PANC1 cells. Thus, it is worth noting that in p53 null or defective tumors, targeting in down-regulation of p65 may well be useful, leading to the potentiality of chemotherapeutic drugs.</p

    Central Angiotensin II Stimulation Promotes β Amyloid Production in Sprague Dawley Rats

    BACKGROUND: Stress and various stress hormones, including catecholamines and glucocorticoids, have recently been implicated in the pathogenesis of Alzheimer's disease (AD), which represents the greatest unresolved medical challenge in neurology. Angiotensin receptor blockers have shown benefits in AD and prone-to-AD animals. However, the mechanisms responsible for their efficacy remain unknown, and no studies have directly addressed the role of central angiotensin II (Ang II), a fundamental stress hormone, in the pathogenesis of AD. The present study focused on the role of central Ang II in amyloidogenesis, the critical process in AD neuropathology, and aimed to provide direct evidence for the role of this stress hormone in the pathogenesis of AD. METHODOLOGY/PRINCIPAL FINDINGS: Increased central Ang II levels during stress response were modeled by intracerebroventricular (ICV) administration of graded doses of Ang II (6 ng/hr low dose, 60 ng/hr medium dose, and 600 ng/hr high dose, all delivered at a rate of 0.25 µl/hr) to male Sprague Dawley rats (280-310 g) via osmotic pumps. After 1 week of continuous Ang II infusion, the stimulation of Ang II type 1 receptors was accompanied by the modulation of amyloid precursor protein, α-, β-and γ-secretase, and increased β amyloid production. These effects could be completely abolished by concomitant ICV infusion of losartan, indicating that central Ang II played a causative role in these alterations. CONCLUSIONS/SIGNIFICANCE: Central Ang II is essential to the stress response, and the results of this study suggest that increased central Ang II levels play an important role in amyloidogenesis during stress, and that central Ang II-directed stress prevention and treatment might represent a novel anti-AD strategy

    Comparison of lignocellulose composition in four major species of Miscanthus

    Miscanthus is a perennial grass rich in lignocellulose that has attracted interest as a non-food crop for renewable bioenergy with major environmental and economic benefits for China. The lignocellulose composition of whole stems of four major species of Miscanthus was assessed. The average values of total moisture content (TMC) (61.90%) and hemicelluloses (34.86%) were the highest while cellulose (32.71%) and acid detergent lignin (ADL) (8.90%) were the lowest in Miscanthus floridulus. On the contrary, the contents of cellulose (42.11%) and ADL (13.64%) were the highest and total ash (TA) (2.89%) was the lowest in Miscanthus lutarioriparius. The Shannon–Weaver diversity indices of components for the four species showed that hemicellulose content (H’= 2.00±0.11) was the most variable trait followed by cellulose (H’= 1.84±0.07), then ADL (H’= 1.84±0.07). The variational range of each component was relatively higher in Miscanthus sacchariflorus. In M. lutarioriparius, the diversity indices of each component were moderate. The diversity of cellulose was the highest and hemicellulose, ADL, TA and TMC were low in Miscanthus sinensis. By correlation analysis, neutral detergent fiber (NDF) significantly and positively correlated with ADF, cellulose and ADL at P&lt;0.01 as well as the relationship of cellulose and ADL in the four species. Hemicellulose showed significant (P&lt;0.01) but negative correlation with cellulose and ADL in M. floridulus, M. lutarioriparius and M. sacchariflorus. By principal component analysis (PCA), the components ADF and cellulose were the PC1 that were considered the foremost for the evaluation and selection of resource in the four species. The conclusions show that lignocellulose composition contents of Miscanthus culms were different. M. floridulus was more fit to ethanol fermentation. Though the components contents in M. sinensis and M. sacchariflorus were moderate, the range of choice was large. It provided a possible means to screen the appropriate materials according to different utilization. M. lutarioriparius had more superiorities relatively. So the four species of Miscanthus were appropriate for extension as excellent herbaceous energy plants, though, reasonable species choice should be employed according to the conversion approach and the growth characteristics, productivity levels and biomass quality characteristics of these tall grasses.Keywords: Miscanthus, bioenergy, lignocellulose compositions, detergent fiber, diversity analysis, PC

    Enhanced baseline activity in the left ventromedial putamen predicts individual treatment response in drug-naive, first-episode schizophrenia: Results from two independent study samples

    BACKGROUND: Antipsychotic medications are the common treatment for schizophrenia. However, reliable biomarkers that can predict individual treatment response are still lacking. The present study aimed to examine whether baseline putamen activity can predict individual treatment response in schizophrenia. METHODS: Two independent samples of patients with drug-naive, first-episode schizophrenia (32 patients in sample 1 and 44 in sample 2) and matched healthy controls underwent resting-state functional magnetic resonance imaging (fMRI) at baseline. Patients were treated with olanzapine for 8 weeks; symptom severity was assessed using the Positive and Negative Syndrome Scale (PANSS) at baseline and week 8. Fractional amplitude of low frequency fluctuation (fALFF) and pattern classification techniques were used to analyze the data. FINDINGS: Univariate analysis shows an elevated pre-treatment fALFF in the left ventromedial putamen in both patient samples compared to healthy controls (p\u27s \u3c 0.001). The support vector regression (SVR) analysis suggests a positive relationship between baseline pre-treatment fALFF in the left ventromedial putamen and improvement in positive symptom at week 8 in each patient group using a cross-validated method (r=0.452, p=.002; r=0.511, p=.003, respectively). INTERPRETATION: Our study suggests that elevated pre-treatment mean fALFF in the left ventromedial putamen may predict individual therapeutic response to olanzapine treatment in drug-naive, first-episode patients with schizophrenia. Future studies are needed to confirm whether this finding is generalizable to patients with schizophrenia treated with other antipsychotic medications.
