Clinical characteristics and prognosis of sudden sensorineural hearing loss in single-sided deafness patients
BackgroundSudden sensorineural hearing loss (SSNHL) in patients with single-sided deafness (SSD) is rare. The prognosis of the sole serviceable hearing ear is very important for these patients. However, the clinical characteristics and prognosis of SSNHL in SSD patients are not well-documented.ObjectiveThis study aimed to investigate the clinical features and treatment outcomes of SSNHL in SSD patients.MethodsClinical data of 36 SSD patients and 116 non-SSD patients with unilateral SSNHL from January 2013 to December 2022 were retrospectively investigated. The clinical characteristics of the SSD patients were analyzed. All SSD patients were treated with intratympanic steroids plus intravenous steroids. Pure-tone average (PTA) and word recognition score (WRS) before and after treatment were recorded. The hearing recovery of SSNHL in SSD patients in comparison with non-SSD patients was explored. Auditory outcomes in SSD patients with different etiologies were also compared.ResultsInitial hearing threshold showed no significant differences between the SSD group and the non-SSD group (66.41 ± 24.64 dB HL vs. 69.21 ± 31.48 dB HL, p = 0.625). The SSD group had a higher post-treatment hearing threshold (median (interquartile range, IQR) 53.13(36.56) dB HL) than the non-SSD group (median 32.50(47.5) dB HL, p < 0.01). Hearing gains (median 8.75(13.00) dB) and the rate of significant recovery (13.89%) were lower in the SSD group than in the non-SSD group (median 23.75(34.69) dB, 45.69%). The etiology of SSD was classified as SSNHL, special types of infection, chronic otitis media, and unknown causes. SSNHL accounted for the maximum proportion (38.9%) of causes of SSD in the SSD group. Hearing gains were lower in the SSNHL-SSD group than in other causes of the SSD group. A binary logistic regression analysis demonstrated that SSD serves as an indicator of unfavorable hearing recovery outcomes (OR = 5.264, p < 0.01).ConclusionThe prognosis of SSNHL in SSD patients is unsatisfactory. SSNHL accounts for the maximum proportion of causes of SSD in this group of patients. For SSD patients caused by SSNHL, less hearing improvement after treatment was expected when SSNHL occurred in the contralateral ear in comparison with SSD patients with other causes

A Rare Functional Noncoding Variant at the GWAS-Implicated MIR137/MIR2682 Locus Might Confer Risk to Schizophrenia and Bipolar Disorder
Schizophrenia (SZ) genome-wide association studies (GWASs) have identified common risk variants in >100 susceptibility loci; however, the contribution of rare variants at these loci remains largely unexplored. One of the strongly associated loci spans MIR137 (miR137) and MIR2682 (miR2682), two microRNA genes important for neuronal function. We sequenced ∼6.9 kb MIR137/MIR2682 and upstream regulatory sequences in 2,610 SZ cases and 2,611 controls of European ancestry. We identified 133 rare variants with minor allele frequency (MAF) <0.5%. The rare variant burden in promoters and enhancers, but not insulators, was associated with SZ (p = 0.021 for MAF < 0.5%, p = 0.003 for MAF < 0.1%). A rare enhancer SNP, 1:g.98515539A>T, presented exclusively in 11 SZ cases (nominal p = 4.8 × 10−4). We further identified its risk allele T in 2 of 2,434 additional SZ cases, 11 of 4,339 bipolar (BP) cases, and 3 of 3,572 SZ/BP study controls and 1,688 population controls; yielding combined p values of 0.0007, 0.0013, and 0.0001 for SZ, BP, and SZ/BP, respectively. The risk allele T of 1:g.98515539A>T reduced enhancer activity of its flanking sequence by >50% in human neuroblastoma cells, predicting lower expression of MIR137/MIR2682. Both empirical and computational analyses showed weaker transcription factor (YY1) binding by the risk allele. Chromatin conformation capture (3C) assay further indicated that 1:g.98515539A>T influenced MIR137/MIR2682, but not the nearby DPYD or LOC729987. Our results suggest that rare noncoding risk variants are associated with SZ and BP at MIR137/MIR2682 locus, with risk alleles decreasing MIR137/MIR2682 expression
Harmonizing Aerosol Carbon Measurements between Two Conventional Thermal/Optical Analysis Methods
Although total carbon (TC) can be consistently quantified by various aerosol carbon measurement methods, the demarcation of TC into organic carbon (OC) and elemental carbon (EC) has long been inconsistent. The NIOSH and IMPROVE protocols are most widely used for thermal/optical analysis (TOA), but current knowledge rests in the description that the NIOSH protocol usually gives lower EC values than does the IMPROVE protocol. This study seeks to explore the possibility of quantitatively linking the difference between the two TOA protocols. Residential coal-burning samples that had been collected and analyzed following the NIOSH protocol in previous studies were directly reanalyzed following the IMPROVE protocol for this study. A comparison of each pair of NIOSH and IMPROVE EC values reveals the dynamic relation between the two protocols, which can be expressed as a regression equation, y = (1-x)/(1 + 4.86x(2)) (R-2 = 0.96), where the independent x is the EC/TC ratio (R-EC/TC) for the IMPROVE protocol, and the dependent y is the difference between IMPROVE and NIOSH R-EC/TC relative to IMPROVE R-EC/TC. Silicon alleviates cadmium toxicity in Avicennia marina (Forsk) Vierh seedlings in relation to root anatomy and radial oxygen loss
National Important Scientific Research Program of China [2013CB956504]; National Natural Science Foundation of China [30970527, 31170471, 31370516]The effects of Si on growth, the anatomy of the roots, radial oxygen loss (ROL) and Fe/Mn plaque on the root surface were investigated in Avicennia marina (Forsk.) Vierh. seedlings under Cd stress. Si prompted the growth of seedlings and reduced the Cd concentration in the root, stem and leaf of A. marina. Si prompted the development of the apoplastic barrier in the roots, which may be related to the reduction of Cd uptake. The higher amount of ROL and Mn plaque on the root surface due to Si were also related to the promotion of Cd tolerance in A. marina seedlings. Therefore, it is concluded that the alteration of the anatomy of the roots, the increase of ROL and Mn plaque of A. marina seedlings play an important role in alleviation of Cd toxicity due to Si.
