Observations of surface deformation are essential for understanding a wide range of geophysical problems, including earthquakes, volcanoes, landslides, and glaciers. Current geodetic technologies, such as global positioning system (GPS), interferometric synthetic aperture radar (InSAR), borehole and laser strainmeters, are costly and limited in their temporal or spatial resolutions. Here we present a new type of strainmeters based on the coaxial cable Bragg grating (CCBG) sensing technology that provides cost-effective strain measurements. Two CCBGs are introduced into the geodetic strainmeter: one serves as a sensor to measure the strain applied on it, and the other acts as a reference to detect environmental noises. By integrating the sensor and reference signals in a mixer, the environmental noises are minimized and a lower mixed frequency is obtained. The lower mixed frequency allows for measurements to be taken with a portable spectrum analyzer, rather than an expensive spectrum analyzer or a vector network analyzer (VNA). Analysis of laboratory experiments shows that the strain can be measured by the CCBG sensor, and the portable spectrum analyzer can make measurements with the accuracy similar to the expensive spectrum analyzer, whose relative error to the spectrum analyzer R3272 is less than ±0.4%. The outputs of the geodetic strainmeter show a linear relationship with the strains that the CCBG sensor experienced. The measured sensitivity of the geodetic strainmeter is about −0.082 kHz/με; it can cover a large dynamic measuring range up to 2%, and its nonlinear errors can be less than 5.3%

Ni1-xCoxFe2O4 (x = 0, 0.2, 0.4, 0.6, 0.8) ceramics were prepared by chemical co-precipitation method and the effect of Co ion doping on the microstructure, magnetic and dielectric properties has been investigated. The results show that the synthesized ceramics display only spinel phase of Ni1-xCoxFe2O4, without other apparent impurities found. The lattice of Ni1-xCoxFe2O4 crystal structure was distorted as a result of the incorporation of Co ion, and the lattice parameters increase with the increase of Co ion content. The grain size decreases slightly with increasing the content of Co ion, indicating a change of particle size and morphology at higher doping content. The results of impedance analysis shows that the sample doped with 80 at.% Co possesses the maximal dielectric constant, while the pure NiFe2O4 sample shows the minimal value when the frequency is below 0.1 MHz. The M-H loops of these ceramics exhibit highly magnetic nature and the saturation magnetization. The remnant magnetization increases linearly with the increase of Co-concentration in nickel ferrite while the coercive field (Hc) shows non-monotonic variation with Co content. The minimal and maximal values of Hc can be obtained when the Co concentrations are 40 and 80 at.%, respectively. The highest value of the saturation magnetization is 63 emu/g obtained with 80 at.% Co doping while the lowest value is ∼31 emu/g for the pure NiFe2O4 ceramics. The abnormal magnetic behaviour is due to the A-B super exchange interaction when magnetic Co2+ ions are added

Neurological pain (NP) is always accompanied by symptoms of depression, which seriously affects physical and mental health. In this study, we identified the common hub genes (Co-hub genes) and related immune cells of NP and major depressive disorder (MDD) to determine whether they have common pathological and molecular mechanisms. NP and MDD expression data was downloaded from the Gene Expression Omnibus (GEO) database. Common differentially expressed genes (Co-DEGs) for NP and MDD were extracted and the hub genes and hub nodes were mined. Co-DEGs, hub genes, and hub nodes were analyzed for Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment. Finally, the hub nodes, and genes were analyzed to obtain Co-hub genes. We plotted Receiver operating characteristic (ROC) curves to evaluate the diagnostic impact of the Co-hub genes on MDD and NP. We also identified the immune-infiltrating cell component by ssGSEA and analyzed the relationship. For the GO and KEGG enrichment analyses, 93 Co-DEGs were associated with biological processes (BP), such as fibrinolysis, cell composition (CC), such as tertiary granules, and pathways, such as complement, and coagulation cascades. A differential gene expression analysis revealed significant differences between the Co-hub genes ANGPT2, MMP9, PLAU, and TIMP2. There was some accuracy in the diagnosis of NP based on the expression of ANGPT2 and MMP9. Analysis of differences in the immune cell components indicated an abundance of activated dendritic cells, effector memory CD8+ T cells, memory B cells, and regulatory T cells in both groups, which were statistically significant. In summary, we identified 6 Co-hub genes and 4 immune cell types related to NP and MDD. Further studies are needed to determine the role of these genes and immune cells as potential diagnostic markers or therapeutic targets in NP and MDD

Crucial Roles of 5-HT and 5-HT2 Receptor in Diabetes-Related Lipid Accumulation and Pro-Inflammatory Cytokine Generation in Hepatocytes

Background/Aims: Previously, we confirmed that liver-synthesized 5-HT rather than non-liver 5-HT, acting on the 5-HT2 receptor (5-HT2R), modulates lipid-induced excessive lipid synthesis (ELS). Here, we further revealed the effects of the hepatocellular 5-HT system in diabetes-related disorders. Methods: Studies were conducted in male ICR mice, human HepG2 cells, and primary mouse hepatocytes (PMHs) under gene or chemical inhibition of the 5-HT system, key lipid metabolism, and inflammation-related factors. Protein and messenger RNA expression and levels of the factors were determined via western blotting, reverse transcription PCR, and quantitative assay kits, respectively. Hepatic steatosis with inflammation and fibrosis, intracellular lipid droplet accumulation (LDA), and reactive oxygen species (ROS) location were determined via hematoxylin and eosin, Masson’s trichrome, Oil red O, and fluorescent-specific staining, respectively. Results: Palmitic acid induced the activation of the 5-HT system: the activation of 5-HT2R, primarily 5-HT2AR, in addition to upregulating monoamine oxidase A (MAO-A) expression and 5-HT synthesis, by activating the G protein/ phospholipase C pathway modulated PKCε activation, resulting in ELS with LDA; the activation of NF-κB, which mediates the generation of pro-inflammatory cytokines, was primarily due to ROS generation in the mitochondria induced by MAO-A–catalyzed 5-HT degradation, and secondarily due to the activation of PKCε. These effects of the 5-HT system were also detected in palmitic acid- or high glucose-treated PMHs and regulated multiple inflammatory signaling pathways. In diabetic mice, co-treatment with antagonists of both 5-HT synthesis and 5-HT2R significantly abolished hepatic steatosis, inflammation, and fibrosis as well as hyperglycemia and dyslipidemia. Conclusion: Activation of the hepatocellular 5-HT system plays a crucial role in inducing diabetes-related hepatic dysfunction and is a potential therapeutic target

HIV-Infected Former Plasma Donors in Rural Central China: From Infection to Survival Outcomes, 1985–2008

BACKGROUND: The HIV epidemic among former plasma donors (FPDs) in rural Central China in the early-mid 1990s is likely the largest known HIV-infected cohort in the world related to commercial plasma donation but has never been fully described. The objectives of this study are to estimate the timing and geographic spread of HIV infection in this cohort and to demonstrate the impact of antiretroviral therapy on survival outcomes. METHODOLOGY/PRINCIPAL FINDINGS: HIV-infected FPDs were identified using the national HIV epidemiology and treatment databases. Locations of subjects were mapped. Dates of infection and survival were estimated using the midpoint date between initial-final plasma donation dates from 1985-2008 among those with plasma donation windows ≤2 years. Among 37,084 FPDs in the two databases, 36,110 were included. 95% were located in focal areas of Henan Province and adjacent areas of surrounding provinces. Midpoint year between initial-final plasma donation dates was 1994 among FPDs with known donation dates. Median survival from infection to AIDS was 11.8 years and, among those not treated, 1.6 years from AIDS to death. Among those on treatment, 71% were still alive after five years. Using Cox proportional hazard modeling, untreated AIDS patients were 4.9 times (95% confidence interval 4.6-5.2) more likely to die than those on treatment. CONCLUSIONS/SIGNIFICANCE: The epidemic of HIV-infected FPD in China was not widespread throughout China but rather was centered in Henan Province and the adjacent areas of surrounding provinces. Even in these areas, infections were concentrated in focal locations. Overall, HIV infections in this cohort peaked in 1994, with median survival of 13.4 years from infection to death among those not treated. Among AIDS patients on treatment, 71% were still alive after five years

MicroRNA Transcriptomic Analysis of Heterosis during Maize Seed Germination

    Get PDF
Heterosis has been utilized widely in the breeding of maize and other crops, and plays an important role in increasing yield, improving quality and enhancing stresses resistance, but the molecular mechanism responsible for heterosis is far from clear. To illustrate whether miRNA-dependent gene regulation is responsible for heterosis during maize germination, a deep-sequencing technique was applied to germinating embryos of a maize hybrid, Yuyu22, which is cultivated widely in China and its parental inbred lines, Yu87-1 and Zong3. The target genes of several miRNAs showing significant expression in the hybrid and parental lines were predicted and tested using real-time PCR. A total of 107 conserved maize miRNAs were co-detected in the hybrid and parental lines. Most of these miRNAs were expressed non-additively in the hybrid compared to its parental lines. These results indicated that miRNAs might participate in heterosis during maize germination and exert an influence via the decay of their target genes. Novel miRNAs were predicted follow a rigorous criterion and only the miRNAs detected in all three samples were treated as a novel maize miRNA. In total, 34 miRNAs belonged to 20 miRNA families were predicted in germinating maize seeds. Global repression of miRNAs in the hybrid, which might result in enhanced gene expression, might be one reason why the hybrid showed higher embryo germination vigor compared to its parental lines
