    A 2-chain can interlock with an open 10-chain

    It is an open problem, posed in \cite{SoCG}, to determine the minimal kk such that an open flexible kk-chain can interlock with a flexible 2-chain. It was first established in \cite{GLOSZ} that there is an open 16-chain in a trapezoid frame that achieves interlocking. This was subsequently improved in \cite{GLOZ} to establish interlocking between a 2-chain and an open 11-chain. Here we improve that result once more, establishing interlocking between a 2-chain and a 10-chain. We present arguments that indicate that 10 is likely the minimum.Comment: 9 pages, 6 figure

    Research on Coordinating Cloud Service Supply Chain Considering Service Disruption

    The risk of the implementation of cloud service and the worry about the failure of projects or strategies caused by service disruption is an important reason of low adoption rates of the cloud service. Service disruption not only directly affects the cloud service free trial results, but also leads to compensation to the consumers. The coordination problem between a CFP (cloud function provider) and a CIP (cloud integration provider) in a cloud supply chain is investigated, in which service demand is determined by the application free trial. Coordination Contracts are discussed in two kinds of situations, linked respectively to the information symmetry and information asymmetry. The results show that the cost and risk-sharing coordination contracts we proposed can realize optimal supply chain performance, and Pareto improvement of supply chain members’ profits. Reducing the service disruption probability and improving the level of service reliability are the key to the free trial. Besides, the compensation cost allocation enhances the scalability of cost allocation. Through numerical exploration analysis, effectiveness of the model is demonstrated and some managerial insights are obtained

    17-beta-hydroxysteroid dehydrogenase 10 (HSD17B10) plays an important role in mitochondrial fatty acid metabolism and is also involved in mitochondrial tRNA maturation. HSD17B10 missense mutations cause HSD10 mitochondrial disease (HSD10MD). HSD17B10 with mutations identified from cases of HSD10MD show loss of function in dehydrogenase activity and mitochondrial tRNA maturation, resulting in mitochondrial dysfunction. It has also been implicated to play roles in the development of Alzheimer disease (AD) and tumorigenesis. Here, we found that HSD17B10 is a new substrate of NAD-dependent deacetylase Sirtuin 3 (SIRT3). HSD17B10 is acetylated at lysine residues K79, K99 and K105 by the acetyltransferase CBP, and the acetylation is reversed by SIRT3. HSD17B10 acetylation regulates its enzymatic activity and the formation of mitochondrial RNase P. Furthermore, HSD17B10 acetylation regulates the intracellular functions, affecting cell growth and cell resistance in response to stresses. Our results demonstrated that acetylation is an important regulation mechanism for HSD17B10 and may provide insight into interrupting the development of AD

    Acox2 is a regulator of lysine crotonylation that mediates hepatic metabolic homeostasis in mice

    Acyl-CoA oxidase 2 (Acox2) is an enzyme involved in peroxisomal bile acid synthesis and branched-chain fatty acid degradation. Acox2 knockout (−/−) mice spontaneously developed liver cancer with marked lymphocytic infiltrate. Tandem-affinity purification coupled with mass spectrometry analysis revealed that Acox2 interacted with methylcrotonoyl-CoA carboxylase followed by co-immunoprecipitation confirmation. Here we reported that non-histone lysine crotonylation (Kcr) levels were downregulated in Acox2 −/− mice livers. Interestingly, Kcr signals were concentrated in the nucleus of tumor cells but mostly located in the cytoplasm of adjacent normal liver cells of Acox2 −/− mice. Quantitative analysis of the global crotonylome further revealed that 54% (27/50) of downregulated non-histone Kcr sites were located in mitochondrial (11/50) and peroxisomal (17/50) enzymes including Ehhadh, Scp2, Hsd17b4, Crot, Etfa, Cpt1a, Eci1/2, Hadha, Etfdh, and Idh2. Subsequent site-directed mutagenesis and transcriptome analysis revealed that Ehhadh K 572 cr might have site-specific regulatory roles by downregulating TOP3B expression that lead to increased DNA damage in vitro. Our findings suggested Acox2 is a regulator of Kcr that might play critical role on hepatic metabolic homeostasis

    Case report: Misdiagnosis of primary mucinous cystadenoma of the testicle by ultrasound

    Testicular mucinous cystadenoma is a rare benign testicular tumor with the characteristics of being potentially malignant and showing atypical clinical symptoms; this article reports a case of a primary testicular mucinous cystadenoma misdiagnosed as testicular teratoma by ultrasound. A 69-year-old man was admitted to the hospital because of a 1-year history of left-sided testicular enlargement with scrotal swelling and no obvious abnormalities on laboratory tests. Ultrasound examination revealed solid-mass lesions in the left testicle, suggesting a high probability of teratoma, and contrast-enhanced magnetic resonance imaging (MRI) examination suggested an increased possibility of epidermoid cysts. Contrast-enhanced computed tomography (CT) and contrast-enhanced MRI of the preoperative abdomen and pelvis showed no other primary adenocarcinoma. Doctors performed radical resection of the left testicle. The postoperative pathological diagnosis was mucinous cystadenoma, and calcification as well as partially mildly atypical epithelial cells were discovered. Furthermore, there were no neoplastic lesions in the epididymis or spermatic cord. No metastatic lesions were found after 6 months postoperative follow-up, and the patient’s prognosis remains good
