    Effect of the clinical nursing pathway on clinical symptoms, hip joint function, and nursing satisfaction in male patients with artificial hip arthroplasty

    The purpose of this study is to explore the effect of clinical nursing pathway on the clinical symptoms, hip function and nursing satisfaction of male patients who undergo artificial hip replacement. A total of 102 male patients who had artificial hip arthroplasty and were admitted to the West China Hospital of Sichuan University were recruited. Participants were randomly divided into the study group (given clinical nursing pathway) and the control group (given routine nursing intervention). The hospitalization time, treatment cost, symptom checklist, nursing satisfaction and hip function score of the two groups were observed and compared. The hospitalization time and treatment cost of the study group were significantly lesser than those of the control group (p < 0.05). Both groups showed significant improvement in the scores of the symptom checklist, but the study group had significantly lower scores than the control group. Similarly, on the day of discharge, there was no significant difference in hip joint function scores between the two groups of patients. However, 30 days after surgery, the hip joint function scores of both groups of patients was remarkably improved (p < 0.05), yet, this improvement is significantly higher in the study group. The nursing satisfaction of the study group was higher than that of the control group, and the difference was significant (p < 0.05). The total incidence of complications in the study group was lower than the control group (5.88%, vs. 23.53%, p < 0.05). The clinical nursing pathway could effectively improve the clinical symptoms of male patients undergoing artificial hip arthroplasty, promote the recovery of hip joint function, and significantly improve the clinical nursing satisfaction. Therefore, it is one of the reliable schemes in the clinical practice of patients undergoing artificial hip arthroplasty

    MAPK-activated transcription factor PxJun suppresses PxABCB1 expression and confers resistance to Bacillus thuringiensis Cry1Ac toxin in Plutella xylostella (L.)

    Deciphering the molecular mechanisms underlying insect resistance to Cry toxins produced by Bacillus thuringiensis (Bt) is pivotal for the sustainable utilization of Bt biopesticides and transgenic Bt crops. Previously, we identified that MAPK-mediated reduced expression of the PxABCB1 gene is associated with Bt Cry1Ac resistance in the diamondback moth, Plutella xylostella (L.). However, the underlying transcriptional regulation mechanism remains enigmatic. Herein, the PxABCB1 promoter in Cry1Ac-susceptible and Cry1Ac-resistant P. xylostella strains was cloned and analyzed and found to contain a putative Jun binding site (JBS). A dual-luciferase reporter assay and yeast one-hybrid assay (Y1H) demonstrated that the transcription factor PxJun repressed PxABCB1 expression by interacting with this JBS. The expression levels of PxJun were increased in the midguts of all resistant strains compared to the susceptible strain. Silencing of PxJun expression significantly elevated PxABCB1 expression and Cry1Ac susceptibility in the resistant NIL-R strain, and silencing of PxMAP4K4 expression decreased PxJun expression and also increased PxABCB1 expression. These results indicate that MAPK-activated PxJun suppresses PxABCB1 expression to confer Cry1Ac resistance in P. xylostella, deepening our understanding of the transcriptional regulation of midgut Cry receptor genes and the molecular basis of insect resistance to Bt Cry toxins.ImportanceThe transcriptional regulation mechanisms underlying reduced expression of Bt toxin receptor genes in Bt-resistant insects remain elusive.     The Rat IgGFcÎłBP and Muc2 C-Terminal Domains and TFF3 in Two Intestinal Mucus Layers Bind Together by Covalent Interaction

    The secreted proteins from goblet cells compose the intestinal mucus. The aims of this study were to determine how they exist in two intestinal mucus layers.The intestinal mucosa was fixed with Carnoy solution and immunostained. Mucus from the loose layer, the firm layer was gently suctioned or scraped, respectively, lysed in SDS sample buffer with or without DTT, then subjected to the western blotting of rTFF3, rIgGFcÎłBP or rMuc2. The non-reduced or reduced soluble mucus samples in RIPA buffer were co-immunoprecipitated to investigate their possible interactions. Polyclonal antibodies for rTFF3, the rIgGFcÎłBP C-terminal domain and the rMuc2 C-terminal domain confirmed their localization in the mucus layer and in the mucus collected from the rat intestinal loose layer or firm layer in both western blot and immunoprecipitation experiments. A complex of rTFF3, which was approximately 250 kDa, and a monomer of 6 kDa were present in both layers of the intestinal mucus; rIgGFcÎłBP was present in the complex (250-280 kDa) under non-reducing conditions, but shifted to 164 kDa under reducing conditions in both of the layers. rMuc2 was found mainly in a complex of 214-270 kDa under non-reducing conditions, but it shifted to 140 kDa under reducing conditions. The co-immunoprecipitation experiments showed that binding occurs among rTFF3, rIgGFcÎłBP and rMuc2 in the RIPA buffer soluble intestinal mucus. Blocking the covalent interaction by 100 mM DTT in the RIPA buffer soluble intestinal mucus disassociated their binding.Rat goblet cell-secreted TFF3, IgGFcÎłBP and Muc2, existing in the two intestinal mucus layers, are bound together by covalent interactions in the soluble fraction of intestinal mucus and form heteropolymers to be one of the biochemical mechanisms of composing the net-like structure of mucus

    Ascl2 Knockdown Results in Tumor Growth Arrest by miRNA-302b-Related Inhibition of Colon Cancer Progenitor Cells

    Background: Achaete scute-like 2 (Ascl2), a basic helix-loop-helix (bHLH) transcription factor, controls the fate of intestinal stem cells. However, the role of Ascl2 in colon cancer progenitor cells remains unknown. The cell line HT-29 (47.5–95 % of CD133 + population) and LS174T (0.45 % of CD133 + population) were chosen for functional evaluation of Ascl2 in colon cancer progenitor cells after gene knockdown by RNA interference. Methodology/Principal Findings: Immunohistochemistry demonstrated that Ascl2 was significantly increased in colorectal adenocarcinomas. Downregulation of Ascl2 using RNA interference in cultured colonic adenocarcinoma HT-29 and LS174T cells reduced cellular proliferation, colony-forming ability, invasion and migration in vitro, and resulted in the growth arrest of tumor xenografts in vivo. The Ascl2 protein level in CD133 + HT-29 cells was significantly higher than in CD133 2 HT-29 cells. Ascl2 blockade via shRNA interference in HT-29 cells (shRNA-Ascl2/HT-29 cells) resulted in 26.2 % of cells staining CD133 + compared with 54.7 % in control shRNA-Ctr/HT-29 cells. The levels of ‘stemness ’ associated genes, such as CD133, Sox2, Oct4, Lgr5, Bmi1, and C-myc, were significantly decreased in shRNA-Ascl2/HT-29 and shRNA-Ascl2/LS174T cells in vitro as well as in the corresponding tumor xenograft (CD133 was not performed in shRNA-Ascl2/LS174T cells). The shRNA-Ascl2/ HT-29 cells had inhibited abilities to form tumorspheres compared with control.     Short-term Breakdown and Long-term Failure in Nanodielectrics: A Review

    Nanodielectrics, which are concentrated in polymer matrix incorporating nanofillers, have received considerable attention due to their potential benefits as dielectrics. In this paper, short-term breakdown and long-term failure properties of nanodielectrics have been reviewed. The characteristics of polymer matrix, types of nanoparticle and its content, and waveforms of the applied voltage are fully evaluated. In order to effectively comment on the published experimental data, a ratio k has been proposed to compare the electric properties of the nanodielectrics with the matrix and assess the effect for nanoparticles doping. There is evidence that the short-term breakdown properties of nanodielectrics show a strong dependence on the applied voltage waveforms. The polarity and the cohesive energy density (CED) of polymer matrix have a dramatic influence on the properties of nanodielectrics. Nanoparticle doped composites show a positive effect on the long-term failure properties, such as ageing resistance and partial discharge (PD) properties of nanocomposites are superior than microcomposites and the matrix. The larger the dielectric constant and CED of the matrix become, the more significant improvements in long-term performance appear. Based on the reported experimental results, we also present our understandings and propose some suggestions for further work

    Robust size illusion produced by expanding and contracting flow fields

    A new illusion is described. Randomly positioned dots moved radially within an imaginary annular window. The dots&#39; motion periodically changed the direction, leading to an alternating percept of expanding and contracting motion. Strikingly, the apparent size of the enclosed circular region shrank during the dots&#39; expanding phases and dilated during the contracting phases. We quantitatively measured the illusion, and found that the presence of energy at the local kinetic edge could not account for the illusion. Besides, we reproduced the illusion on a natural scene background seen from a first-person point of view that moved forward and backward periodically. Blurring the boundaries of motion areas could not reverse the illusion in all subjects. Taken together, our observed illusion is likely induced by optic flow processing with some components of motion contrast. Expanding or contracting dots may induce the self-motion perception of either approaching or leaving way from the circle. These will make the circle appear smaller or larger since its retinal size remains constant. (C) 2017 Elsevier Ltd. All rights reserved.</p

    Impact of acute pancreatitis during pregnancy in Chinese women: a meta-analysis

    A random-effects meta-analysis was performed in English and Chinese databases since its inception to August 2020 to assess the incidence, causes and severity of acute pancreatitis (AP) at various stages of pregnancy, maternal and foetal mortality. A total of 154 articles representing 4034 patients with AP during pregnancy in China were included for the analysis. The incidence of AP during pregnancy was 0.0469 (95% confidence interval [CI], 0.0349; 0.0627) in the first trimester, whereas it was 0.2518 (95% CI, 0.2210; 0.2854) and 0.6323 (95% CI, 0.5870; 0.6753) in the second and third trimester, respectively. The major causes of AP were hypertriglyceridaemia (0.351 [95% CI, 0.3202; 0.3834]) and biliary pancreatitis (0.424 [95% CI, 0.4094; 0.5002]). The severity of AP was mild in majority of the patients. The incidence of AP at maternal mortality was 0.0184 (95% CI, 0.0126; 0.0269) and foetal mortality was 0.1018 (95% CI, 0.0867; 0.1192). Our meta-analysis revealed that hypertriglyceridaemia and biliary pancreatitis remain the major causes of AP during pregnancy. Foetal mortality requires further investigation. IMPACT STATEMENT What is already known on this subject? Acute pancreatitis (AP) in pregnant women is characterised by acute onset and delay in understanding the interaction of the metabolic changes with pancreatic pathophysiology, and thus becomes difficult to diagnose the disease and provide timely treatment to the patients. This poses a greater health risk among women and their foetus by increasing their chances of mortality. What the results of this study add? We performed an exhaustive, random-effects meta-analysis involving 154 articles representing 4034 patients to assess the incidence of AP at various stages of pregnancy, the causes of AP and the severity of AP during pregnancy, maternal and foetal mortality. What are the implications of these findings for clinical practice and/or further research? Our meta-analysis revealed that hypertriglyceridaemia and biliary pancreatitis remain the major causes of AP during pregnancy. Although the rates of maternal mortality have decreased in the recent years, foetal mortality still remains high and requires further investigation
