    Lighting-Up Tumor for Assisting Resection via Spraying NIR Fluorescent Probe of γ-Glutamyltranspeptidas

    For the precision resection, development of near-infrared (NIR) fluorescent probe based on specificity identification tumor-associated enzyme for lighting-up the tumor area, is urgent in the field of diagnosis and treatment. Overexpression of γ-glutamyltranspeptidase, one of the cell-membrane enzymes, known as a biomarker is concerned with the growth and progression of ovarian, liver, colon and breast cancer compared to normal tissue. In this work, a remarkable enzyme-activated NIR fluorescent probe NIR-SN-GGT was proposed and synthesized including two moieties: a NIR dicyanoisophorone core as signal reporter unit; γ-glutamyl group as the specificity identification site. In the presence of γ-GGT, probe NIR-SN-GGT was transformed into NIR-SN-NH2, the recovery of Intramolecular Charge Transfer (ICT), liberating the NIR fluorescence signal, which was firstly employed to distinguish tumor tissue and normal tissues via simple “spraying” manner, greatly promoting the possibility of precise excision. Furthermore, combined with magnetic resonance imaging by T2 weight mode, tumor transplanted BABL/c mice could be also lit up for first time by NIR fluorescence probe having a large stokes, which demonstrated that probe NIR-SN-GGT would be a useful tool for assisting surgeon to diagnose and remove tumor in clinical practice

    Anaplasma, Bartonella, and Rickettsia infections in Daurian ground squirrels (Spermophilus dauricus), Hebei, China

    Rodents have been confirmed as hosts of various vector-borne zoonotic pathogens and are important for the maintenance of these microbes in nature. However, surveillance for zoonotic pathogens is limited for many wild rodent species in China, so our knowledge of pathogen ecology, genetic diversity, and the risk of cross-species transmission to humans is limited. In this study, 165 spleen samples of Daurian ground squirrels (Spermophilus dauricus) were collected from Weichang Manchu and the Mongolian Autonomous County of Hebei Province, China, and Rickettsia, Bartonella, and Anaplasma were identified by DNA detection using polymerase chain reaction (PCR). Sequence analysis identified eight bacterial pathogens: R. raoultii, R. sibirica, Candidatus R. longicornii, B. washoensis, B. grahamii, B. jaculi, A. capra, and Candidatus Anaplasma cinensis. Co-infection of B. grahamii and R. raoultii in one sample was observed. Our results demonstrated the genetic diversity of bacteria in Daurian ground squirrels and contributed to the distribution of these pathogens. Six species, A. capra, R. raoultii, R. sibirica, Candidatus R. longicornii, B. washoensis, and B. grahamii, are known to be pathogenic to humans, indicating a potential public health risk to the local human population, especially to herders who frequently have close contact with Daurian ground squirrels and are thus exposed to their ectoparasites

    [Objectives] To enrich the species resources of Tianjin, Aornia melanocarpa was introduced and cultivated in 4 districts of Tianjin. [Methods] Two consecutive years of observation in 2018-2019 and research on the phenological period, high yield and fruit economic traits in 4 districts of Tianjin were carried out. [Results] The A. melanocarpa of the Linhai Jiayuan Experimental Site in Jinghai District has larger single fruit weight, higher nutrient composition, stronger plant growth, higher yield and storage stability, so this site can be used as a main planting base for large-scale planting of A. melanocarpa; the experimental site of Tianjin Agricultural University East Campus and the experimental site of Cuiping Lake Science Park of Jizhou District have a sour and astringent taste but relatively low yield, need to be processed as by-products in combination with market prices to determine whether to plant; the experimental site of Tianjin Agricultural University West Campus has low yield, small fruit and poor taste, and the planting effect is the worst among the four experimental sites. [Conclusions] From the overall economic traits of fruits, the fruits produced in the experimental site of Jinghai District have sufficient weight, high solid-acid ratio, and relatively high nutrient content, thus this experimental site can be used as large-scale cultivation base for A. melanocarpa

    The Impact of Insect-Proof Screen on Microclimate, Reference Evapotranspiration and Growth of Chinese Flowering Cabbage in Arid and Semi-Arid Region

    Despite the steadily increasing area under protected agriculture there is a current lack of knowledge about the effects of the insect-proof screen (IPS) on microclimate and crop water requirements in arid and semi-arid regions. Field experiments were conducted in two crop cycles in Ningxia of Northwest China to study the impact of IPS on microclimate, reference evapotranspiration (ET0) and growth of Chinese Flowering Cabbage (CFC). The results showed that IPS could appreciably improve the microclimate of the CFC field in the two crop cycles. During the first crop cycle (C1), compared with no insect-proof screen (NIPS) treatment, the total solar radiation and daily wind speed under the IPS treatment were reduced by 5.73% and 88.73%. IPS increased the daily average air humidity, air, and soil temperature during C1 by 11.84%, 15.11% and 10.37%, respectively. Furthermore, the total solar radiation and daily wind speed under the IPS treatment during the second crop cycle (C2) were markedly decreased by 20.45% and 95.73%, respectively. During C2, the daily average air temperature and air humidity under the IPS treatment were increased slightly, whereas the daily average soil temperature was decreased by 4.84%. Compared with NIPS treatment, the ET0 under the IPS treatment during C1 and C2 was decreased by 6.52% and 21.20%, respectively, suggesting it had great water-saving potential when using IPS. The plant height, leaf number and leaf circumference of CFC under the IPS treatment were higher than those under the NIPS treatment. The yield under the IPS treatment was significantly increased by 36.00% and 108.92% in C1 and C2, respectively. Moreover, irrigation water use efficiency (IWUE) was significantly improved under the IPS treatment in the two crop cycles. Therefore, it is concluded that IPS can improve microclimate, reduce ET0, and increase crop yield and IWUE in arid and semi-arid areas of Northwest China

During plant growth and development, environmental stresses, such as drought, salt, cold, and heat stress are unavoidable. This raises an interesting question as to how the legumes cope with the environmental stress along with SNF. Under drought stress, dehydrin proteins are accumulated, which function as protein protector and osmotic substances. In this study, we found that the dehydrin MtCAS31 (cold-acclimation-specific 31) functions in SNF in Medicago truncatula during drought stress. We found that MtCAS31 is expressed in nodules and interacts with leghemoglobin MtLb120-1. The interaction between the two proteins protects MtLb120-1 from denaturation under thermal stress in vivo. Compared to wild type, cas31 mutants display a lower nitrogenase activity, a lower ATP/ADP ratio, higher expression of nodule senescence genes and higher accumulation of amyloplasts under dehydration conditions. The results suggested that MtCAS31 protects MtLb120-1 from the damage of drought stress. We identified a new function for dehydrins in SNF under drought stress, which enriches the understanding of the molecular mechanism of dehydrins.</p

    Immunogenicity of a Candidate DTacP-sIPV Combined Vaccine and Its Protection Efficacy against Pertussis in a Rhesus Macaque Model

    The research and development of a pertussis-combined vaccine using a novel inactivated poliovirus vaccine made from the Sabin strain (sIPV) is of great significance in the polio eradication project and to address the recent resurge in pertussis. In the present study, we compared the immunogenicity and efficacy of a candidate DTacP-sIPV with those of a commercial DTacP-wIPV/Hib, DTaP/Hib, pertussis vaccine, and aluminum hydroxide adjuvant control in the rhesus macaque model with a 0-, 1-, and 2-month immunization schedule. At day 28 after the third dose, rhesus macaques were challenged with aerosol pertussis and the antibody and cellular response together with pertussis clinical symptoms were determined. The production of anti-PT, anti-PRN, anti-FHA, anti-DT, anti-TT, and polio type I, II, III antibodies was induced by the candidate DTacP-sIPV, which was as potent as commercial vaccines. In comparison with the control group that showed typical pertussis symptoms of humans after the aerosol challenge, the DTacP-sIPV group did not exhibit obvious clinical pertussis symptoms and had higher neutralization titers of anti-PT, anti-PRN, and anti-FHA. In conclusion, the DTacP-sIPV vaccine was able to induce immunity in rhesus macaques to prevent pertussis infections after immunization. The developed vaccine was as efficient as other commercial vaccines
