    A Trust Model for Vehicular Network-Based Incident Reports

    Vehicle-to-Vehicle (V2V) and Vehicle-to-Infrastructure (V2I) networks are ephemeral, short-duration wireless networks that have the potential to improve the overall driving experience through the exchange of information between vehicles. V2V and V2I networks operate primarily by distributing real-time incident reports regarding potential traffic problems such as traffic jams, accidents, bad roads and so on to other vehicles in their vicinity over a multi-hop network. However, given the presence of malicious entities, blindly trusting such incident reports (even the one received through a cryptographically secure channel) can lead to undesirable consequences. In this paper, we propose an approach to determine the likelihood of the accuracy of V2V incident reports based on the trustworthiness of the report originator and those vehicles that forward it. The proposed approach takes advantage of existing road-side units (RSU) based V2I communication infrastructure deployed and managed by central traffic authorities, which can be used to collect vehicle behavior information in a crowd-sourcedfashion for constructing a more comprehensive view of vehicle trustworthiness. For validating our scheme, we implemented a V2V/V2I trust simulator by extending an existing V2V simulator with trust management capabilities. Preliminary analysis of the model shows promising results. By combining our trust modeling technique with a threshold-based decision strategy, we observed on average 85% accuracy

    Inquiry web-based learning to enhance information problem solving competences in science

    Early research on using web information indicates that secondary students fail to explore much web tools, use them naively and have serious difficulties to understand and integrate web information. In response to these challenges, the main goal of this research has been to design, implement and evaluate an instructional approach that helps students learn from web information. We have developed on-line learning materials which focus on specific curricular contents and provide specific scaffolds to help students accomplish web-based tasks and develop specific information problem-solving competencies. These scaffolds have intended to give support to students involved in information-seeking activities as they were asked questions, searched for information, organised and assessed their findings, and created rich representations of their newly-constructed understandings. We have designed a one year long study to investigate the depth and accuracy of 127 secondary students, as regards their content understanding as well as their development of information problem-solving competencies when using on-line resources to solve instructional tasks. Our research demonstrates that the experimental group performed computer-based activities statistically better than the control group. Our findings also suggest that students were able to develop accurate and in-depth understanding from web information if they could appropriately use search and managerial strategies. This research lends evidence to questions regarding the value of students engaging in on-line inquiry web-based learning to enhance content understanding and to develop more efficient information problem-solving competencies in secondary education

    Mi-2β promotes immune evasion in melanoma by activating EZH2 methylation

    Recent development of new immune checkpoint inhibitors has been particularly successfully in cancer treatment, but still the majority patients fail to benefit. Converting resistant tumors to immunotherapy sensitive will provide a significant improvement in patient outcome. Here we identify Mi-2β as a key melanoma-intrinsic effector regulating the adaptive anti-tumor immune response. Studies in genetically engineered mouse melanoma models indicate that loss of Mi-2β rescues the immune response to immunotherapy in vivo. Mechanistically, ATAC-seq analysis shows that Mi-2β controls the accessibility of IFN-γ-stimulated genes (ISGs). Mi-2β binds to EZH2 and promotes K510 methylation of EZH2, subsequently activating the trimethylation of H3K27 to inhibit the transcription of ISGs. Finally, we develop an Mi-2β-targeted inhibitor, Z36-MP5, which reduces Mi-2β ATPase activity and reactivates ISG transcription. Consequently, Z36-MP5 induces a response to immune checkpoint inhibitors in otherwise resistant melanoma models. Our work provides a potential therapeutic strategy to convert immunotherapy resistant melanomas to sensitive ones

    Noncanonical function of DGCR8 controls mESC exit from pluripotency

    Mouse embryonic stem cells (mESCs) deficient for DGCR8, a key component of the microprocessor complex, present strong differentiation defects. However, the exact reasons impairing their commitment remain elusive. The analysis of newly generated mutant mESCs revealed that DGCR8 is essential for the exit from the pluripotency state. To dissociate canonical versus noncanonical functions of DGCR8, we complemented the mutant mESCs with a phosphomutant DGCR8, which restored microRNA levels but did not rescue the exit from pluripotency defect. Integration of omics data and RNA immunoprecipitation experiments established DGCR8 as a direct interactor of Tcf7l1 mRNA, a core component of the pluripotency network. Finally, we found that DGCR8 facilitated the splicing of Tcf7l1, an event necessary for the differentiation of mESCs. Our data reveal a new noncanonical function of DGCR8 in the modulation of the alternative splicing of Tcf7l1 mRNA in addition to its established function in microRNA biogenesis
