    Development genetic and stability classification of seasonal glacial lakes in a tectonically active area—A case study in Niangmuco, east margin of the Eastern Himalayan Syntaxis

    The Niangmuco region on the east margin of the Eastern Himalayan Syntaxis features a large number of glacial lakes. The development process and stability classification of glacial lakes is of great significance to the study of seasonal glaciers in the eastern Himalayan margin, with implications for economic development and disaster prevention. Based on Landsat remote sensing image data from 2000 to 2021, this study analyzed the development and change characteristics of glacial lakes in the Niangmuco region during the past 21 years, and classified the stability of lakes with areas >0.02 km2 using the fuzzy consistent matrix method. In this area, 126 glacial lakes were identified within an elevation range of 3044–4156 m with a total area of 10.94 km2. These lakes primarily included glacial erosion lakes, valley lakes, tectonic lakes, and landslide dam lakes. Specifically, glacial erosion lakes accounted for 88.9% of the total number of lakes and 60.3% of the total lake area, followed by valley lakes with 6.3% and 23.7%, respectively. From 2000 to 2010, the total area of glacial lakes decreased from 10.53 km2 to 10.09 km2, which may be attributable to climate fluctuations. Subsequently, the area of lakes increased significantly to 10.94 km2 in 2021, an increase of 0.41 km2. Compared with 2000, among the lakes with a growth rate of 0.019 km2/a in 21 years, glacial erosion lakes exhibited the largest change. Among the classified glacial lakes in the study area, 95.7% were stable and relatively stable, mostly comprising glacial erosion lakes at high altitudes between 3468 and 4156 m. Only 4 unstable and extremely unstable glacial lakes were identified, and they were located near a fault zone. The findings show that the development and the change of glacial lakes in the area are primarily controlled by temperature and precipitation, and the topography and fault activity have important influences on the stability of glacial lakes

    Evaluation of LOXL1 polymorphisms in exfoliation syndrome in a Chinese population

    Purpose: To evaluate the association profiles of the lysyl oxidase-like 1 (LOXL1) gene polymorphisms with exfoliation syndrome in a Chinese population. Methods: Fifty unrelated patients with exfoliation syndrome and 125 control subjects were included. Genotypes of the three single nucleotide polymorphisms (SNPs) of LOXL1 (rs1048661, rs3825942, and rs2165241) were analyzed by direct sequencing, and a case-control association study was performed. Results: The three SNPs were significantly associated with exfoliation syndrome (XFS) and exfoliation glaucoma (XFG) individually. After controlling for rs3825942 and rs2165241, the association between rs1048661 and XFS/XFG remained significant (p=3.6x10(-7)). At this SNP, the T allele and TT genotype conferred a 7.59-(95% confidence interval [CI]: 3.87-14.89, p=6.95x10(-11)) and 8.69-(95% CI: 4.15-18.20, p<1.00x10(-7)) fold increased risk to the disease. The alleles of T at rs1048661 and C at rs2165241 were found to be risk alleles in Chinese subjects, which were opposite to Caucasian individuals. The haplotypes T-G, defined by SNPs rs1048661 and rs3825942, and T-C by SNPs rs1048661 and rs2165241, were also significantly associated with the disorder. However when the genotypic or allelic frequencies of the three SNPs were compared between XFS and XFG, no significant difference was detected. Conclusions: LOXL1 is a susceptibility gene of XFS/XFG in the Chinese population, and the association is mainly attributed to SNP rs1048661. The risk alleles of rs1048661 and rs2165241 in Chinese subjects were found to be opposite to that of Caucasians.     Investigation into the Influence of Physician for Treatment Based on Syndrome Differentiation

    Background. The characteristics of treatment based on syndrome differentiation (TBSD) cause great challenges to evaluate the effectiveness of the clinical methods. Objectives. This paper aims to evaluate the influence of physician to personalized medicine in the process of TBSD. Methods. We performed a randomized, triple-blind trial involving patients of primary insomnia treated by 3 physicians individually and independently. The patients (n=30) were randomly assigned to receive treatments by the 3 physicians for every visit. However, they always received the treatment, respectively, prescribed by the physician at the first visit. The primary outcome was evaluated, respectively, by the Pittsburgh Sleep Quality Index (PSQI) and the TCM symptoms measuring scale. The clinical practices of the physicians were recorded at every visit including diagnostic information, syndrome differentiation, treating principles, and prescriptions. Results. All patients in the 3 groups (30 patients) showed significant improvements (>66%) according to the PSQI and TCM symptoms measuring scale. Conclusion. The results indicate that although with comparable effectiveness, there exist significant differences in syndrome differentiation, the treating principles, and the prescriptions of the approaches used by the 3 physicians. This means that the physician should be considered as an important factor for individualized medicine and the related TCM clinical research

    Assessing Heavy Metal Pollution of the Largest Nature Reserve in Tianjin City, China

    Embargo until June 10, 2023Beidagang Wetland (BW) Nature Reserve is centrally situated in Tianjin City, experiencing an extreme industrial development. This study uses index characteristic analysis systems for assessing the individual and combined heavy metal pollution loading in the water during the spring and autumn seasons. By combining the pollution level of single pollutant, a more comprehensive evaluation of water quality in BW was achieved. Water quality was worst during autumn due to high level of Cd and Pb, which indicate the type of anthropogenic activities have a serious effect on heavy metal pollution in BW. In addition, high exchangeable amounts of Cd (> 40%) were found in the sediments of BW, indicating Cd pollution has emerged. There is a need for appropriate abatement actions curbing heavy metal loading and improving water quality of the BW Nature Reserve, thereby ensuring a sustainable management of its ecosystem services.acceptedVersio

    ADAR2-dependent RNA editing of GluR2 is involved in thiamine deficiency-induced alteration of calcium dynamics

    BACKGROUND: Thiamine (vitamin B1) deficiency (TD) causes mild impairment of oxidative metabolism and region-selective neuronal loss in the central nervous system (CNS). TD in animals has been used to model aging-associated neurodegeneration in the brain. The mechanisms of TD-induced neuron death are complex, and it is likely multiple mechanisms interplay and contribute to the action of TD. In this study, we demonstrated that TD significantly increased intracellular calcium concentrations [Ca2+]i in cultured cortical neurons. RESULTS: TD drastically potentiated AMPA-triggered calcium influx and inhibited pre-mRNA editing of GluR2, a Ca2+-permeable subtype of AMPA receptors. The Ca2+ permeability of GluR2 is regulated by RNA editing at the Q/R site. Edited GluR2 (R) subunits form Ca2+-impermeable channels, whereas unedited GluR2 (Q) channels are permeable to Ca2+ flow. TD inhibited Q/R editing of GluR2 and increased the ratio of unedited GluR2. The Q/R editing of GluR2 is mediated by adenosine deaminase acting on RNA 2 (ADAR2). TD selectively decreased ADAR2 expression and its self-editing ability without affecting ADAR1 in cultured neurons and in the brain tissue. Over-expression of ADAR2 reduced AMPA-mediated rise of [Ca2+]i and protected cortical neurons against TD-induced cytotoxicity, whereas down-regulation of ADAR2 increased AMPA-elicited Ca2+ influx and exacerbated TD-induced death of cortical neurons. CONCLUSIONS: Our findings suggest that TD-induced neuronal damage may be mediated by the modulation of ADAR2-dependent RNA Editing of GluR2

    Comparison of a solvent mixture assisted dilute acid and alkali pretreatment in sugar production from hybrid Pennisetum

    Abstract(#br)The effects of an acetone-butanol-ethanol (ABE) mixture on dilute H 2 SO 4 and NaOH pretreatment for enzymatic saccharification of hybrid Pennisetum (HP) were investigated. The results showed that ABE assisted the removal of xylan and lignin during H 2 SO 4 and NaOH pretreatment, respectively. The glucose yield of HP increased from 33.6% to 52.9% with the assistance of a relatively higher concentration of ABE mixture (ABE4) during H 2 SO 4 pretreatment, and during NaOH pretreatment, a lower concentration of ABE (ABE2) increased the glucose yield from 64.6% to 80.2%. The hydrolysis yield increases were related to the compositional change and surface characteristics of the pretreated materials. As observed by X-ray photoelectron spectroscopy, ABE4 resulted in a greater lignin content on the surface of materials than that produced by ABE2 during NaOH pretreatment, which possibly increased the non-productive adsorption of cellulase, thus decreasing the hydrolysis yield. The results suggested that an ABE mixture could be used as an auxiliary agent for further increasing of the digestibility of acid- and alkali-pretreated lignocellulosic materials. However, the digestibility was different depending on the concentrations of ABE during acid and alkali pretreatments

    Diversity of classic and novel human astrovirus in outpatient children with acute gastroenteritis in Shanghai, China

    IntroductionHuman astrovirus (HAstV) is an important pathogen of acute gastroenteritis (AGE) in children. This study was aimed at investigating the diversity and epidemiology of classic and novel HAstV in outpatient children aged 0–16 years old with AGE in Shanghai.MethodsFrom May 2020 to December 2022, a total of 1,482 stool samples were collected from children diagnosed as AGE from the Children’s Hospital of Fudan University. HAstV was identified using pan-astrovirus consensus primers by Reverse transcription PCR.ResultsDuring the study period, 3.3% (49/1,482) of specimens were identified as HAstV, with a detection rate of 2.5% (37/1,482) for classic HAstV and 0.8% (12/1,482) for novel HAstV. Among the 12 novel HAstV strains, 11 (91.7%) belonged to the HAstV-MLB and 1 (8.3%) was HAstV-VA. Genotyping revealed six circulating genotypes. Strain HAstV-1 was predominant in the study population with a detection rate of 1.8% (26/1,482) followed by HAstV-MLB1 (0.7%, 10/1,482) and HAstV-4 (0.6%, 9/1,482). Of note, all the HAstV-4 strains detected in this study were close to one astrovirus strain isolated from Bactrian camels with 99.0-100.0% amino acid sequences identity. In this study, HAstV was detected in all age groups with the highest detection rate of HAstV-positive specimens observed in children older than 73 months (5.7%, 12/209).DiscussionThis study provided useful information and contributed to the molecular epidemiology of both classic and novel HAstV, which were simultaneously characterized and reported for the first time in Shanghai

    Holocene sediment source analysis and paleoclimatic significance of core KZK01 from the eastern part of the Beibu Gulf

    Identifying the sources of sediments is of great significance in reconstructing Holocene paleoclimate evolution in the Beibu Gulf and in understanding the characteristics of regional responses to changes in global climate. The Holocene paleoclimatic evolutionary history of the Beibu Gulf was investigated by chronological, geochemical, and mineralogical means using the sediments of Core KZK01 from the eastern part of the Beibu Gulf. The rare earth element (REE) distribution curves, (Gd/Yb)N and (La/Yb)N discriminant diagram, and (Gd/Lu)N and ∑LREE/∑HREE discriminant diagram indicated that the detrital materials in the eastern part of the Beibu Gulf primarily originated from Hainan Island and its proximal sources, with considerable contributions from Taiwan and Pearl River materials. Source analysis of clay minerals showed that Luzon Island was the main source of smectite, followed by Hainan Island. Rivers in Taiwan were the main sources of illite in the study area, followed by the Red River. The Red River was the main contributor of chlorite, followed by the Pearl River. Kaolinite mainly originated from Hainan Island and Guangxi. Coastal currents, surface currents, and warm currents were the main drivers of material transport. Paleoclimatic variations since the Holocene in the Beibu Gulf were divided into three stages: 12–9 cal kyr BP, 9–1.3 cal kyr BP, and 1.3 cal kyr BP to the present. During different stages of climatic evolution, drought was often accompanied by cold and humidity coexisted with warmth, and cold-dry-warm-humid alternation is characterized by significant phases. The illite crystallinity clearly recorded the extreme cold events, such as Bond Events (except Bond6) and the Younger Dryas, and the change trend was essentially consistent with the regional climate record, reflecting the control of global climate change on the process of land–sea interaction in the tropical region. Furthermore, it highlights the great potential of illite crystallinity as a proxy indicator for reconstructing the surface chemical weathering processes of the region
