    Implementação estadual do edTPA em preparação para testes de alto risco: Um estudo de métodos mistos das preocupações dos coordenadores do edTPA

    This study examined the implementation of high-stakes adoption of edTPA® in one state in the year prior to consequential use of edTPA scores for teacher licensure. Using a mixed methods design, we investigated concerns of coordinators who were responsible for edTPA implementation in their institutions. We utilized the Concerns Based Adoption Model (CBAM) to understand edTPA coordinators’ Stages of Concern, the nature of the challenges they faced, and the professional development opportunities that alleviated their concerns. Based on the CBAM survey, the most common Stage of Concernfor edTPA coordinators was Management.Coordinators’ interviews revealed the nature of their concerns at different stages and how the size of their institution and supportive resources at particular times may have played a crucial role in shaping the edTPA roll-out in their institutions. The use of the CBAM framework enabled edTPA coordinators (a) to understand their own concerns about the high-stakes policy, (b) to articulate the complexities involved in implementing edTPA initiatives, and (c) to underscore the importance of relating concerns to appropriate professional development opportunities and support for themselves as well as their faculty. Este estudio examinó la implementación de la adopción de edTPA® de alto riesgo en un estado previo al uso consecuente de los puntajes de edTPA para la licencia de maestros. Usando un diseño de método mixto, investigamos las preocupaciones de los coordinadores responsables de implementar edTPA en sus instituciones. Utilizamos el Modelo de adopción basado en la preocupación (CBAM) para comprender las etapas de preocupación para los coordinadores de edTPA, la naturaleza de los desafíos que enfrentaron y las oportunidades de desarrollo profesional que alivian sus preocupaciones. Según la investigación de CBAM, la etapa de preocupación más común para los coordinadores de edTPA fue la gestión. Las entrevistas con los coordinadores revelaron la naturaleza de sus preocupaciones en diferentes etapas y cómo el tamaño de su institución y los recursos de apoyo en ciertos momentos pueden haber jugado un papel crucial en la definición de la implementación de edTPA en sus instituciones. El uso del marco CBAM ha permitido a los coordinadores de edTPA (a) comprender sus propias preocupaciones sobre la política de alto riesgo, (b) articular las complejidades involucradas en la implementación de iniciativas de edTPA, y (c) subrayar la importancia de relacionar las preocupaciones con oportunidades apropiadas para el desarrollo profesional y el apoyo para ellos mismos y la facultad.Este estudo examinou a implementação da adoção de edTPA® de alto risco em um estado do ano anterior ao uso consequente das pontuações de edTPA para licenciamento de professores. Usando um design de métodos mistos, investigamos as preocupações dos coordenadores responsáveis pela implementação do edTPA em suas instituições. Utilizamos o Modelo de Adoção com Base em Preocupações (CBAM) para entender os Estágios de Preocupação dos coordenadores da edTPA, a natureza dos desafios que enfrentaram e as oportunidades de desenvolvimento profissional que atenuaram suas preocupações. Com base na pesquisa da CBAM, o estágio de preocupação mais comum para os coordenadores da edTPA foi o gerenciamento. As entrevistas dos coordenadores revelaram a natureza de suas preocupações em diferentes estágios e como o tamanho de sua instituição e os recursos de suporte em determinados momentos podem ter desempenhado um papel crucial na definição da implementação do edTPA em suas instituições. O uso da estrutura CBAM permitiu aos coordenadores da edTPA (a) entender suas próprias preocupações sobre a política de altos riscos, (b) articular as complexidades envolvidas na implementação de iniciativas da edTPA, e (c) sublinhar a importância de relacionar as preocupações às oportunidades apropriadas de desenvolvimento profissional e apoiar a si e ao corpo docente

    Reversible Plasticity of Fear Memory-Encoding Amygdala Synaptic Circuits Even after Fear Memory Consolidation

    It is generally believed that after memory consolidation, memory-encoding synaptic circuits are persistently modified and become less plastic. This, however, may hinder the remaining capacity of information storage in a given neural circuit. Here we consider the hypothesis that memory-encoding synaptic circuits still retain reversible plasticity even after memory consolidation. To test this, we employed a protocol of auditory fear conditioning which recruited the vast majority of the thalamic input synaptic circuit to the lateral amygdala (T-LA synaptic circuit; a storage site for fear memory) with fear conditioning-induced synaptic plasticity. Subsequently the fear memory-encoding synaptic circuits were challenged with fear extinction and re-conditioning to determine whether these circuits exhibit reversible plasticity. We found that fear memory-encoding T-LA synaptic circuit exhibited dynamic efficacy changes in tight correlation with fear memory strength even after fear memory consolidation. Initial conditioning or re-conditioning brought T-LA synaptic circuit near the ceiling of their modification range (occluding LTP and enhancing depotentiation in brain slices prepared from conditioned or re-conditioned rats), while extinction reversed this change (reinstating LTP and occluding depotentiation in brain slices prepared from extinguished rats). Consistently, fear conditioning-induced synaptic potentiation at T-LA synapses was functionally reversed by extinction and reinstated by subsequent re-conditioning. These results suggest reversible plasticity of fear memory-encoding circuits even after fear memory consolidation. This reversible plasticity of memory-encoding synapses may be involved in updating the contents of original memory even after memory consolidation

    Interplay between chronic inflammation and clonal haematopoiesis of indeterminate potential in Behçets disease

    Background Clonal haematopoiesis of indeterminate potential (CHIP) is a predisposition to haematological malignancy whose relationship with chronic inflammatory diseases, such as cardiovascular diseases, has been highlighted. Here, we aimed to investigate the CHIP emergence rate and its association with inflammatory markers in Behçets disease (BD). Methods We performed targeted next-generation sequencing to detect the presence of CHIP using peripheral blood cells from 117 BD patients and 5004 healthy controls between March 2009 and September 2021 and analysed the association between CHIP and inflammatory markers. Results CHIP was detected in 13.9% of patients in the control group and 11.1% of patients in the BD group, indicating no significant intergroup difference. Among the BD patients of our cohort, five variants (DNMT3A, TET2, ASXL1, STAG2, and IDH2) were detected. DNMT3A mutations were the most common, followed by TET2 mutations. CHIP carriers with BD had a higher serum platelet count, erythrocyte sedimentation rate, and C-reactive protein level; older age; and lower serum albumin level at diagnosis than non-CHIP carriers with BD. However, the significant association between inflammatory markers and CHIP disappeared after the adjustment for various variables, including age. Moreover, CHIP was not an independent risk factor for poor clinical outcomes in patients with BD. Conclusions Although BD patients did not have higher CHIP emergence rates than the general population, older age and degree of inflammation in BD were associated with CHIP emergence

    The Six1 oncoprotein downregulates p53 via concomitant regulation of RPL26 and microRNA-27a-3p

    TP53 is mutated in 50% of all cancers, and its function is often compromised in cancers where it is not mutated. Here we demonstrate that the pro-tumorigenic/metastatic Six1 homeoprotein decreases p53 levels through a mechanism that does not involve the negative regulator of p53, MDM2. Instead, Six1 regulates p53 via a dual mechanism involving upregulation of microRNA-27a and downregulation of ribosomal protein L26 (RPL26). Mutation analysis confirms that RPL26 inhibits miR-27a binding and prevents microRNA-mediated downregulation of p53. The clinical relevance of this interaction is underscored by the finding that Six1 expression strongly correlates with decreased RPL26 across numerous tumour types. Importantly, we find that Six1 expression leads to marked resistance to therapies targeting the p53–MDM2 interaction. Thus, we identify a competitive mechanism of p53 regulation, which may have consequences for drugs aimed at reinstating p53 function in tumours

    Meaning of Parental Involvement Among Korean Immigrant Parents: A Mixed-Methods Approach

    The authors\u27 goal was to identify ways in which Korean immigrant parents define the concept of parental involvement and to examine the statistical significances of interrelationships among these meanings. Seventy-seven parents responded to an open-ended question that asked them to define the meaning of parental involvement; 141 responses were analyzed. Qualitative analysis resulted in four distinct categories: (a) support at home (68.8%), (b) home–school connection (17.7%), (c) participation in school (5.0%), and (d) duty (8.5%). The category of support at home was divided into three subcategories: Support of nonacademic development (31.9%), support of emotional psychological development (29.1%), and support of academic success (7.8%). A correlational analysis indicated that support of emotional psychological development was negatively correlated with home–school connection and support of academic success. The parents who considered parental involvement to be their duty did not perceive their roles as participating in home–school connections and supporting nonacademic development activities

    Korean Immigrant Parents’ Perceptions of Library Service in Support of Their Own and Their Children’s Learning

    Even though local libraries are one of the most easily available and accessible community organizations for immigrant parents to seek assistance, little is known about the ways in which immigrant parents use local libraries to enhance their own learning and their children’s learning. A questionnaire was developed to study the current use of library programs and services by Korean immigrant parents and their children as well as their and their children’s need for library programs and services. One forty-one completed questionnaires were analyzed. The more time parents spent at the libraries, the more they came to believe that the libraries offered programs for immigrant families, �s = .25, p \u3c .01. Almost half of the participants (43%) reported dissatisfaction with library services because of the lack of Korean resources (16%), linguistic barriers (16%), and the lack of organized cultural events (11%). Results on the frequency of time spent at libraries, reasons to use library resources, and their awareness of programs for immigrants offered by libraries will be shared. The importance of this study is that the parents stressed contextual barriers such as the lack of resources in immigrants’ native languages as a strong barrier for immigrants to not use library services
