St. John’s Wort Regulates Proliferation and Apoptosis in MCF-7 Human Breast Cancer Cells by Inhibiting AMPK/mTOR and Activating the Mitochondrial Pathway
St. John’s Wort (SJW) has been used as an estrogen agonist in the systems affected by menopause. Also, hypericin, a bioactive compound of SJW, has been used as a photosensitizer in photodynamic therapy. In the present study, we investigate the anti-proliferative and pro-apoptotic effects of SJWto demonstrate the chemo-preventive effect in human breast cancer cells. MCF-7 cellswere culturedwith DMSO or various concentrations of SJWethanol extract (SJWE). Cell viability, proliferation, apoptosis, the expression of proteins involved in cell growth and apoptosis, and caspase-3/7 activity were examined. SJWE dose-dependently suppressed cell growth and induced apoptosis ofMCF-7 cells. Mechanistically, SJWE enhanced the phosphorylation of AMP-activated protein kinase (AMPK) and decreased the expression of p-mammalian target of rapamycin (p-mTOR) and p-eukaryotic translation initiation factor 4E (eIF4E)-binding protein 1 (4E-BP1). Also, SJWE inhibited the phosphorylation of protein kinase B (Akt) and showed increases in the expression of pro-apoptotic proteins Bax and Bad with decreases in the expression of anti-apoptotic proteins including B-cell lymphoma 2 (Bcl-2), B-cell lymphoma-extra large (Bcl-xL), and p-Bcl-2-associated death promoter (p-Bad). SJWE at 50 µg/mL showed markedly enhanced caspase-7 activation. Taken together, our results provide evidence that SJWE shows anti-proliferative and pro-apoptotic effects via inhibition of AMPK/mTOR and activation of a mitochondrial pathway. Therefore, SJWE can be used as a chemo-preventive agent without photo-activation

Design of temperature insurance index and risk zonation for single-season rice in response to high-temperature and low-temperature damage: a case study of Jiangsu Province, China.
Disaster insurance is an important tool for achieving sustainable development in modern agriculture. However, in China, the design of such insurance indexes is far from sufficient. In this paper, the single-season rice in Jiangsu Province of China is taken as an example to design the high-temperature damage index in summer and the low-temperature damage index in autumn to constructtheformulacalculatingtheweatheroutputandsingle-seasonriceyieldreduction. Thedaily highest, lowest and average temperatures between 1999 and 2015 are selected as main variables for the temperature disaster index to quantitatively analyze the relationship between the temperature indexandtheyieldreductionrateofthesingle-seasonrice. Thetemperaturedisasterindexcanbeput into the relevant model to obtain the yield reduction rate of the year and determine whether to pay the indemnity. Then, the burn analysis is used to determine the insurance premium rate for all cities in Jiangsu Province under four-level deductibles, and the insurance premium rate can be used for the risk division of the Province. The research provides some insights for the design of agricultural weather insurance products, and the empirical results provide a reference for the design of similar single-season rice temperature index insurance products
THiFLY Research at SemEval-2023 Task 7: A Multi-granularity System for CTR-based Textual Entailment and Evidence Retrieval
The NLI4CT task aims to entail hypotheses based on Clinical Trial Reports
(CTRs) and retrieve the corresponding evidence supporting the justification.
This task poses a significant challenge, as verifying hypotheses in the NLI4CT
task requires the integration of multiple pieces of evidence from one or two
CTR(s) and the application of diverse levels of reasoning, including textual
and numerical. To address these problems, we present a multi-granularity system
for CTR-based textual entailment and evidence retrieval in this paper.
Specifically, we construct a Multi-granularity Inference Network (MGNet) that
exploits sentence-level and token-level encoding to handle both textual
entailment and evidence retrieval tasks. Moreover, we enhance the numerical
inference capability of the system by leveraging a T5-based model, SciFive,
which is pre-trained on the medical corpus. Model ensembling and a joint
inference method are further utilized in the system to increase the stability
and consistency of inference. The system achieves f1-scores of 0.856 and 0.853
on textual entailment and evidence retrieval tasks, resulting in the best
performance on both subtasks. The experimental results corroborate the
effectiveness of our proposed method. Our code is publicly available at

Intrasellar abscess after transsphenoidal pituitary surgery
Sellar abscess is extremely rare complication that could occur after a transphenoidal surgery (TSS). Here we report 45-year-old female patient with left cheek numbness for one month. The Magnetic resonance imaging (MRI) report suggested a pituitary adenoma. Therefore, transphenoidal surgery was performed. During follow up, the patient complained of headache for few days. Subsequent MRI revealed recurrent or residual pituitary tumor. We attempted to excise the lesion using a transfrontal approach. However, pus in the sellar region was found at operation, and no tumor tissue was identified. After the surgery, the patient received antibiotic therapy and fully recovered. Sellar abscess is a serious condition with a high mortality rate. However, a combination of surgical resection and specific antibiotic therapy seems to be very promising

The Development of Web3D-based Open-pit Mine Monitoring System
Large-scale open-pit mines are critical infrastructure for acquiring natural resources. However, this type of mine can experience environmental and safety problems during operations and thus requires continuous monitoring. In this study, a web three-dimensional(3D)-based monitoring system is constructed using geospatial information open platform and open-source geospatial information software which targets open-pit mines in Gangwon-do, Korea. The purpose is to develop a monitoring system of open-pit mines that enables any person to monitor the topographic and environmental changes caused by mine operations and to develop and restore the area’s ecology. Open-pit mines were classified into active or inactive mines and monitoring items and methodologies were established for each type of mine. Cesium which is a WebGL-based open-source platform was chosen as it supports dynamic data visualization and hardware-accelerated graphics related to elapsed time which is the essential factor in the monitoring. The open-pit mine monitoring system was developed based on the geospatial database which contains information required for mine monitoring as time elapses, and by developing the open-source-based system software. The geospatial information database for monitoring consists of digital imagery and terrain data and also includes vector data and the restoration plan datas. The basic geospatial information used in the monitoring includes high resolution ortho image(GSD 0.5 m or above) for all areas of the mines. This is acquired by periodically using an airborne laser scanning system and a LiDAR DEM (grid size 1m × 1 m). In addition, geospatial information data were acquired by using an UAV and terrestrial LiDAR for small-scale areas; these tools are frequently used for rapid and irregular data acquisition. The geospatial information acquired for the monitoring of the open-pit mines represents various spatial resolutions and different terrain data. The database was constructed by converging this geospatial information with the Cesium-based geospatial information open platform of the ESRI World Imagery map and with SDK World Terrain meshes. The problems that resulted from the process of fusing aerial imagery and terrain data were solved in the Cesium-based open source environment. The prototype menu for the monitoring system was designed according to the monitoring item which was determined by the type of mine. The scene of the mine and changes in terrain were controlled and analyzed using the raster function of PostGIS according to the elapsed time. Using the GeoServer, the aerial imagery, terrain and restoration information for each period were serviced using the web standard interface, and the monitoring system was completed by visualizing these elements in Cesium in 3D format according to the elapsed time. This study has established a monitoring methodology for open-pit mines according to the type of mine and proposes a method for upgrading the imagery and terrain data required for monitoring. The study also showed the possibility of developing a Web3D-based open-pit mine monitoring system that is applicable to a wide range of mashup service developments
A sulfated polysaccharide of Ecklonia cava inhibits the growth of colon cancer cells by inducing apoptosis
We investigated anticancer effects of the crude polysaccharides (CPs) isolated from Ecklonia cava enzymatic extracts using AMG, Viscozyme, Protamex, and Alcalase enzyme against a colon cancer cell line, CT26 cells. Among them, the CP of Protamex extract (PCP) contained the highest fucose and sulfated group contents and showed the highest growth inhibitory effect against CT-26 cells. In addition, PCP dose-dependently increased the formation of apoptotic body and the percentage of Sub-G1 DNA contents. Also, PCP activated caspase 9 and PARP as regulating the expressions of Bax and Bcl-2. Moreover, PPP2, a fraction purified from PCP showed the highest growth inhibitory effect against CT 26 cells with the increased fucose and sulfated group contents. The results demonstrate that the isolated SP containing plentiful fucose and sulfated group contents has the anticancer effect on colon cancer cells via regulation of Bcl-2/Bax signal pathway
Anchoring Competition on Nanosurface Boundaries with Conflicting Mixed Nematic Anchoring Properties
We propose a technique for continuously controlling the full range of pretilt angles with a high process margin. The proposed method is characterized by tuning the thickness of a heterogeneous polyimide layer that consists of homeotropic and planar polyimides. The thickness of the mixture can be controlled by varying the concentration of the solvent. The liquid crystal (LC) pretilt generated at the very thin mixture film is insensitive to some incorrect mixing ratio, since the segregation of the depth direction of the mixture including the van der Waals effect in interactions with LCs decreases ultimately. Consequently, we can obtain continuous LC pretilt angles with a high process margin by controlling mixing ratio in a very thin heterogeneous polyimide film. Furthermore, it is possible to achieve excellent reliability, uniformity, and productivity using this technique. A simple mathematical model based on van der Waals interaction provides a good description of the experimental results
CD4+ T cells from MHC II-dependent thymocyte–thymocyte interaction provide efficient help for B cells
Recently, a novel CD4+ T-cell developmental pathway was reported that generates thymocyte–thymocyte (T–T) CD4+ T cells. We established a mouse system (CIITAtgCIITApIV−/−) where thymic positive selection occurred only by major histocompatibility complex (MHC) class II+ thymocytes. T–T CD4+ T cells selected via MHC class II-dependent T–T interaction are comprised of PLZF-negative and innate PLZF-positive populations. Until recently, the functional role of the PLZF-negative population was unclear. In this study, we demonstrate that naïve T–T CD4+ T cells provide B-cell help to a level comparable with that of naïve conventional CD4+ T cells. Considering the absence of PLZF expression in naïve T–T CD4+ T cells, these results suggest that PLZF-negative naïve T–T CD4+ T cells are functionally equivalent to conventional naïve CD4+ T cells in terms of B-cell help
Propofol Inhibits Lipopolysaccharide-Induced Tumor Necrosis Factor-Alpha Expression and Myocardial Depression through Decreasing the Generation of Superoxide Anion in Cardiomyocytes
TNF-α has been shown to be a major factor responsible for myocardial depression in sepsis. The aim of this study was to investigate the effect of an anesthetic, propofol, on TNF-α expression in cardiomyocytes treated with LPS both in vivo and in vitro. In cultured cardiomyocytes, compared with control group, propofol significantly reduced protein expression of gp91phox and phosphorylation of extracellular regulated protein kinases 1/2 (ERK1/2) and p38 MAPK, which associates with reduced TNF-α production. In in vivo mice studies, propofol significantly improved myocardial depression and increased survival rate of mice after LPS treatment or during endotoxemia, which associates with reduced myocardial TNF-α production, gp91phox, ERK1/2, and p38 MAPK. It is concluded that propofol abrogates LPS-induced TNF-α production and alleviates cardiac depression through gp91phox/ERK1/2 or p38 MAPK signal pathway. These findings have great clinical importance in the application of propofol for patients enduring sepsis
