    Are Asian Thinking Styles Different? Acculturation and Thinking Styles in a Chinese-Canadian Population

    This research examined the theory of mental self-government and its various thinking styles. The theory of mental self-government (Sternberg, 1988a) has established its utility in verbal contexts including education, occupation, partner choices, and cross-culturally. The Associated Thinking Style Inventory (TSI; Sternberg & Wagner, 1991; 1992) has also demonstrated its reliability and validity in a Hong Kong population (Zhang, 1999; Zhang & Lcks, 1997; Zhang & Sternberg, 1998), but it has not been used with Chinese-Canadians. It was hypothesized that as Chinese individuals become more acculturated to the North American cultural system, their thinking styles approach European-American norms. This study found the a to be an internally consistent measure of thinking styles in a Chinese-Canadian population; ! median alpha reliability was .82. Observed intercorrelations between the various thinking variables were in the anticipated directions and three factors were found to account for 55% of the variance. Significant relationships were found between some thinking styles and socialization variables such as gender, age, amount of education, and years lived in North America. However : present data do not support the hypothesis that Chinese-Canadian acculturation was related to : theory of mental self-government. Although the TSI and Suinn-Lew Asian Self-Identity co-culturation Scale (SL-ASIA; Suinn, Rikard-Figueroa, Lew, & Vigil, 1987) were both reliable, here were no significant correlations between the two measures. The absence of a relationship 1etween the TSI and the SL-ASIA is understandable considering the complexity of these constructs; acculturation has been shown to be multi-faceted and there are numerous possibilities f thinking style profiles. In addition, individuals may diversify their thinking styles to better adapt to an environment, further complicating the matter. Perhaps the complexity of c0-culturation and thinking style profiles does not allow for simple categorization. However, this study was able to demonstrate that Chinese-Canadians think somewhat differently from two Hong Kong populations (Zhang, 1999; Zhang & Sacks, 1997).     This dissertation presents three chapters addressing issues pertaining to monetary policy, information, and central bank communication. The first chapter studies optimal monetary policy in an environment where policy actions provide a signal of economic fundamentals to imperfectly informed agents. I derive the optimal discretionary policy in closed form and show that, in contrast to the perfect information case, the signaling channel leads the policymaker to be tougher on inflation. The strength of the signaling effect of policy depends on relative uncertainty levels. As the signaling effect strengthens, the optimal policy under discretion approaches that under commitment to a forward-looking linear rule, thereby decreasing the stabilization bias. This contributes to the central bank finding it optimal to withhold its additional information from private agents. Under a general linear policy rule, inflation and output forecasts can respond positively to a positive interest rate surprise when the signaling channel is strong. This positive response is the opposite of what standard perfect information New Keynesian models predict and it matches empirical patterns found by previous studies. Chapter 2 provides new empirical evidence supporting the predictions of the model presented in Chapter 1. More specifically, I find that the responses of inflation forecasts to interest rate surprises is especially positive when there is greater uncertainty regarding the previous forecast. Finally, Chapter 3 examines whether communications by the Federal Open Market Committee might have the ability to influence financial market responses to macroeconomic news. In particular, I am able to relate labor-related word use in FOMC statements and meeting minutes to the amount by which interest rates' response to labor-related news exceeds their response to other news.Economic

    Biochemical and Genetic Conservation of Fission Yeast DSK1 and Human SRPK1

    Arginine/serine-rich (RS) domain-containing proteins and their phosphorylation by specific protein kinases constitute control circuits to regulate pre-mRNA splicing and coordinate splicing with transcription in mammalian cells. We present here the finding that similar SR networks exist in Schizosaccharomyces pombe. We previously showed that Dsk1 protein, originally described as a mitotic regulator, displays high activity in phosphorylating S. pombe Prp2 protein (spU2AF59), a homologue of human U2AF65. We now demonstrate that Dsk1 also phosphorylates two recently identified fission yeast proteins with RS repeats, Srp1 and Srp2, in vitro. The phosphorylated proteins bear the same phosphoepitope found in mammalian SR proteins. Consistent with its substrate specificity, Dsk1 forms kinase-competent complexes with those proteins. Furthermore, dsk1+ gene determines the phenotype of prp2+ overexpression, providing in vivo evidence that Prp2 is a target for Dsk1. The dsk1-null mutant strain became severely sick with the additional deletion of a related kinase gene. Significantly, human SR protein-specific kinase 1 (SRPK1) complements the growth defect of the double-deletion mutant. In conjunction with the resemblance of dsk1+ and SRPK1 in sequence homology, biochemical properties, and overexpression phenotypes, the complementation result indicates that SRPK1 is a functional homologue of Dsk1. Collectively, our studies illustrate the conserved SR networks in S. pombe consisting of RS domain-containing proteins and SR protein-specific kinases and thus establish the importance of the networks in eucaryotic organisms

    Active Q-switched X-Ray Regenerative Amplifier Free-Electron Laser

    Despite tremendous progress in x-ray free-electron laser (FEL) science over the last decade, future applications still demand fully coherent, stable x-rays that have not been demonstrated in existing X-ray FEL facilities. In this Letter, we describe an active Q-switched x-ray regenerative amplifier FEL scheme to produce fully coherent, high-brightness, hard x rays at a high-repetition rate. By using simple electron-beam phase space manipulation, we show this scheme is flexible in controlling the x-ray cavity quality factor Q and hence the output radiation. We report both theoretical and numerical studies on this scheme with a wide range of accelerator, x-ray cavity, and undulator parameters

    Automated Detection and Characterization of Cracks on Concrete using Laser Scanning

    Accurate crack detection and characterization on concrete are essential for the maintenance, safety, and serviceability of various infrastructures. In this paper, an innovative approach was developed to automatically measure the cracks from 3D point clouds collected by a phase-shift terrestrial laser scanner (TLS) (FARO Focus3D S120). The approach integrates several techniques to characterize the cracks, which include the deviation on point normal determined using k-nearest neighbor (kNN) and principal components analysis (PCA) algorithms to identify the cracks, and principal axes and curve skeletons of cracks to determine the projected and real dimensions of cracks, respectively. The coordinate transformation was then performed to estimate the projected dimensions of cracks. Curve skeletons and cross sections of cracks were extracted to represent the real dimensions. Two cases of surface cracks were used to validate the developed approach. Because of the differences in definitions of the crack dimension in the three methods and due to the curve shape of the crack, the width and depth of cracks obtained from the cross-section method and manual measurement were close but slightly smaller than those measured by the projection algorithm; whereas the length of cracks determined by the curve-skeletons method was slightly larger than those obtained by the manual measurement and projection method. The real dimension of a crack has good agreements with real situations when compared with the results of the manual measurement and projection method

    Transcriptional regulation of FoxO3 gene by glucocorticoids in murine myotubes.

    Glucocorticoids and FoxO3 exert similar metabolic effects in skeletal muscle. FoxO3 gene expression was increased by dexamethasone (Dex), a synthetic glucocorticoid, both in vitro and in vivo. In C2C12 myotubes the increased expression is due to, at least in part, the elevated rate of FoxO3 gene transcription. In the mouse FoxO3 gene, we identified three glucocorticoid receptor (GR) binding regions (GBRs): one being upstream of the transcription start site, -17kbGBR; and two in introns, +45kbGBR and +71kbGBR. Together, these three GBRs contain four 15-bp glucocorticoid response elements (GREs). Micrococcal nuclease (MNase) assay revealed that Dex treatment increased the sensitivity to MNase in the GRE of +45kbGBR and +71kbGBR upon 30- and 60-min Dex treatment, respectively. Conversely, Dex treatment did not affect the chromatin structure near the -17kbGBR, in which the GRE is located in the linker region. Dex treatment also increased histone H3 and/or H4 acetylation in genomic regions near all three GBRs. Moreover, using chromatin conformation capture (3C) assay, we showed that Dex treatment increased the interaction between the -17kbGBR and two genomic regions: one located around +500 bp and the other around +73 kb. Finally, the transcriptional coregulator p300 was recruited to all three GBRs upon Dex treatment. The reduction of p300 expression decreased FoxO3 gene expression and Dex-stimulated interaction between distinct genomic regions of FoxO3 gene identified by 3C. Overall, our results demonstrate that glucocorticoids activated FoxO3 gene transcription through multiple GREs by chromatin structural change and DNA looping

    Effect of Ca2+ on the Steady-State and Time-Resolved Emission Properties of the Genetically Encoded Fluorescent Sensor CatchER

    We previously designed a calcium sensor CatchER (a GFP-based Calcium sensor for detecting high concentrations in the high calcium concentration environment such as ER) with a capability for monitoring calcium ion responses in various types of cells. Calcium binding to CatchER induces the ratiometric changes in the absorption spectra, as well as an increase in fluorescence emission at 510 nm upon excitation at both 395 and 488 nm. Here, we have applied the combination of the steady-state and time-resolved optical methods and Hydrogen/Deuterium isotope exchange to understand the origin of such calcium-induced optical property changes of CatchER. We first demonstrated that calcium binding results in a 44% mean fluorescence lifetime increase of the indirectly excited anionic chromophore. Thus, CatchER is the first protein-based calcium indicator with the single fluorescent moiety to show the direct correlation between the lifetime and calcium binding. Calcium exhibits a strong inhibition on the excited-state proton transfer nonadiabatic geminate recombination in protic (vs deuteric) medium. Analysis of CatchER crystal structures and the MD simulations reveal the proton transfer mechanism in which the disrupted proton migration path in CatchER is rescued by calcium binding. Our finding provides important insights for a strategy to design calcium sensors and suggests that CatchER could be a useful probe for FLIM imaging of calcium in situ
