    SummarySelf-localization and navigation are critical functions for the survival of mobile animals. By processing sensory information such as landmarks and environmental features, as well as keeping track of the path they have taken, animals are able to remain oriented as they explore the world, learning what resources are where and planning how to reach them. This ancient capacity for self-orientation has, through evolution, become intimately entwined with the ability to remember the events of daily life — an ability known as episodic memory. Spatial and episodic memory involve the interaction of many cognitive faculties and brain circuits, and so are fascinating subjects for study that reveal much about how the brain works

    The White Dwarf Age of NFC 2477

    We present deep photometric observations of the open cluster NGC 2477 using HST/WFPC2. By identifying seven cluster white dwarf candidates, we present an analysis of the white dwarf age of this cluster, using both the traditional method of fitting isochrones to the white dwarf cooling sequence, and by employing a new Bayesian statistical technique that has been developed by our group. This new method performs an objective, simultaneous model fit of the cluster and stellar parameters (namely, age, metallicity, distance, reddening, as well as individual stellar masses, mass ratios, and cluster membership) to the photometry. Based on this analysis, we measure a white dwarf age of 1.035 ± 0.054 ± 0.087 Gyr (uncertainties represent the goodness of model fits and discrepancy among models, respectively) in good agreement with the cluster\u27s main-sequence turnoff age. This work is part of our ongoing work to calibrate main-sequence turnoff and white dwarf ages using open clusters, and to improve the precision of cluster ages to the ~5% level

    Integrated Administrative Data for Early Childhood Iowa: A Governance Model to inform Policy and Program Collaboration

    In response to demands on public systems to do more, do better, and cost less, the value of integrated administrative data systems (IDS) for social policy is increasing (Fantuzzo & Culhane, 2016). This is particularly relevant in programming for young children where services are historically fragmented, disconnected from systems serving school-aged children, and siloed among health, human services, and education agencies. Guided by the vision that Iowa’s early childhood system will be effectively and efficiently coordinated to support healthy families, we are developing an early childhood IDS to address this disconnection and facilitate relevant and actionable social policy research. Iowa’s IDS is a state-university partnership that acknowledges the need for agencies to retain control of their data while enabling it to be integrated across systems for social policy research. The innovative governance model deliberately incorporates procedures for stakeholder engagement at critical tension points between executive leaders, program managers, researchers, and practitioners. Standing committees (Governance Board, Data Stewardship, and Core team) authorize and implement the work of the IDS, while ad-hoc committees are solicited for specific projects to advise and translate research into practice. This paper will articulate the Iowa IDS governance model that was informed by means tested principles articulated by the Actionable Intelligence for Social Policy Network. It will include our collaborative development process; articulated mission and principles that guided discussions about legal authorization, governance, and use cases; and the establishment of governance committees to implement our vision for ethical and efficient use of administrative data for social policy

    Fibroblast Growth Factor-2 Enhances Expansion of Human Bone Marrow-Derived Mesenchymal Stromal Cells without Diminishing Their Immunosuppressive Potential

    Allogeneic hematopoietic stem cell transplantation is the main curative therapy for many hematologic malignancies. Its potential relies on graft-versus-tumor effects which associate with graft-versus-host disease. Mesenchymal stromal cells (MSCs) possess immunomodulatory properties that make them attractive therapeutic alternatives. We evaluated the in vitro immunosuppressive activity of medium conditioned by human MSCs from 5 donors expanded 13 passages with or without FGF-2. FGF-2 supplementation increased expansion 3,500- and 240,000-fold by passages 7 and 13, respectively. There were no differences in immunosuppressive activity between media conditioned by passage-matched cells expanded under different conditions, but media conditioned by FGF-treated MSCs were superior to population doubling-matched controls. The immunosuppressive activity was maintained in three of the preparations but decreased with expansion in two. The proliferation induced by FGF-2 did not result in loss of immunosuppressive activity. However, because the immunosuppressive activity was not consistently preserved, caution must be exercised to ensure that the activity of the cells is sufficient after extensive expansion

    Purely translational realignment in grid cell firing patterns following nonmetric context change

    Grid cells in entorhinal and parahippocampal cortices contribute to a network, centered on the hippocampal place cell system, that constructs a representation of spatial context for use in navigation and memory. In doing so, they use metric cues such as the distance and direction of nearby boundaries to position and orient their firing field arrays (grids). The present study investigated whether they also use purely nonmetric “context” information such as color and odor of the environment. We found that, indeed, purely nonmetric cues—sufficiently salient to cause changes in place cell firing patterns—can regulate grid positioning; they do so independently of orientation, and thus interact with linear but not directional spatial inputs. Grid cells responded homogeneously to context changes. We suggest that the grid and place cell networks receive context information directly and also from each other; the information is used by place cells to compute the final decision of the spatial system about which context the animal is in, and by grid cells to help inform the system about where the animal is within it

    Statistical Viewer: a tool to upload and integrate linkage and association data as plots displayed within the Ensembl genome browser

    BACKGROUND: To facilitate efficient selection and the prioritization of candidate complex disease susceptibility genes for association analysis, increasingly comprehensive annotation tools are essential to integrate, visualize and analyze vast quantities of disparate data generated by genomic screens, public human genome sequence annotation and ancillary biological databases. We have developed a plug-in package for Ensembl called "Statistical Viewer" that facilitates the analysis of genomic features and annotation in the regions of interest defined by linkage analysis. RESULTS: Statistical Viewer is an add-on package to the open-source Ensembl Genome Browser and Annotation System that displays disease study-specific linkage and/or association data as 2 dimensional plots in new panels in the context of Ensembl's Contig View and Cyto View pages. An enhanced upload server facilitates the upload of statistical data, as well as additional feature annotation to be displayed in DAS tracts, in the form of Excel Files. The Statistical View panel, drawn directly under the ideogram, illustrates lod score values for markers from a study of interest that are plotted against their position in base pairs. A module called "Get Map" easily converts the genetic locations of markers to genomic coordinates. The graph is placed under the corresponding ideogram features a synchronized vertical sliding selection box that is seamlessly integrated into Ensembl's Contig- and Cyto- View pages to choose the region to be displayed in Ensembl's "Overview" and "Detailed View" panels. To resolve Association and Fine mapping data plots, a "Detailed Statistic View" plot corresponding to the "Detailed View" may be displayed underneath. CONCLUSION: Features mapping to regions of linkage are accentuated when Statistic View is used in conjunction with the Distributed Annotation System (DAS) to display supplemental laboratory information such as differentially expressed disease genes in private data tracks. Statistic View is a novel and powerful visual feature that enhances Ensembl's utility as valuable resource for integrative genomic-based approaches to the identification of candidate disease susceptibility genes. At present there are no other tools that provide for the visualization of 2-dimensional plots of quantitative data scores against genomic coordinates in the context of a primary public genome annotation browser

    Projected social costs of CO2 emissions from forest losses far exceed the sequestration benefits of forest gains under global change

    Forest cover gains and losses occur in response to complex environmental and anthropogenic pressures. Yet the impact of forest gains and losses on the provision of ecosystem services differs markedly. Here we investigate the social costs of potential forest carbon change in Australia's intensive agricultural region from 2015 to 2050 using spatial forest cover change and forest carbon models combined with climate and socioeconomic projections. More than 24,000 possible scenarios were used to identify the trend and lower and upper bounds of forest cover/carbon change. Net deforestation (3.5 million hectares, Mha)under the lower bound forest cover (LBFC)projection was around one-third less than net reforestation (4.8 Mha)under the upper bound forest cover (UBFC)projection by 2030. However, the CO emissions (1.3 Gigatons of CO , GtCO )from deforestation were more than double the sequestration (0.5 GtCO )from reforestation. The social costs (up to 134 billion dollars)of the LBFC were almost five times the benefits of the UBFC (up to 28 billion dollars). The asymmetry decreased over time but persisted to 2050. This shows the markedly different social costs of potential forest carbon losses and gains under global change, evidence which can be useful to policymakers, stakeholders, and practitioners

    The WFC3 Galactic Bulge Treasury Program: Metallicity Estimates for the Stellar Population and Exoplanet Hosts

    We present new UV-to-IR stellar photometry of four low-extinction windows in the Galactic bulge, obtained with the Wide Field Camera 3 on the Hubble Space Telescope (HST). Using our five bandpasses, we have defined reddening-free photometric indices sensitive to stellar effective temperature and metallicity. We find that the bulge populations resemble those formed via classical dissipative collapse: each field is dominated by an old (~10 Gyr) population exhibiting a wide metallicity range (-1.5 < [Fe/H] < 0.5). We detect a metallicity gradient in the bulge population, with the fraction of stars at super-solar metallicities dropping from 41% to 35% over distances from the Galactic center ranging from 0.3 to 1.2 kpc. One field includes candidate exoplanet hosts discovered in the SWEEPS HST transit survey. Our measurements for 11 of these hosts demonstrate that exoplanets in the distinct bulge environment are preferentially found around high-metallicity stars, as in the solar neighborhood, supporting the view that planets form more readily in metal-rich environments.Comment: Accepted for publication in The Astrophysical Journal Letters.     Place field repetition and purely local remapping in a multicompartment environment

    Hippocampal place cells support spatial memory using sensory information from the environment and self-motion information to localize their firing fields. Currently, there is disagreement about whether CA1 place cells can use pure self-motion information to disambiguate different compartments in environments containing multiple visually identical compartments. Some studies report that place cells can disambiguate different compartments, while others report that they do not. Furthermore, while numerous studies have examined remapping, there has been little examination of remapping in different subregions of a single environment. Is remapping purely local or do place fields in neighboring, unaffected, regions detect the change? We recorded place cells as rats foraged across a 4-compartment environment and report 3 new findings. First, we find that, unlike studies in which rats foraged in 2 compartments, place fields showed a high degree of spatial repetition with a slight degree of rate-based discrimination. Second, this repetition does not diminish with extended experience. Third, remapping was found to be purely local for both geometric change and contextual change. Our results reveal the limited capacity of the path integrator to drive pattern separation in hippocampal representations, and suggest that doorways may play a privileged role in segmenting the neural representation of space

    Transcriptomic analysis of field-droughted sorghum from seedling to maturity reveals biotic and metabolic responses.

    Drought is the most important environmental stress limiting crop yields. The C4 cereal sorghum [Sorghum bicolor (L.) Moench] is a critical food, forage, and emerging bioenergy crop that is notably drought-tolerant. We conducted a large-scale field experiment, imposing preflowering and postflowering drought stress on 2 genotypes of sorghum across a tightly resolved time series, from plant emergence to postanthesis, resulting in a dataset of nearly 400 transcriptomes. We observed a fast and global transcriptomic response in leaf and root tissues with clear temporal patterns, including modulation of well-known drought pathways. We also identified genotypic differences in core photosynthesis and reactive oxygen species scavenging pathways, highlighting possible mechanisms of drought tolerance and of the delayed senescence, characteristic of the stay-green phenotype. Finally, we discovered a large-scale depletion in the expression of genes critical to arbuscular mycorrhizal (AM) symbiosis, with a corresponding drop in AM fungal mass in the plants' roots
