    Effect of storage temperature and wrapping treatments on the keeping quality of tender king coconut

    King coconut (Cocos nucifera var.aurantiaca) has a good export market as a natural beverage. Howerver the quality of the nut has to be maintained for several weeks to meet export market requirements. Therefore, the present study was undertaken to develop techniques to imporve the keeping quality of tender king coconuts. Seven -month -old nuts collected from different locations were given wrapping and waxing treatments and stored at low temperature to improve the keeping quality up to four weeks. Invert sugars and total sugars were analyzed. Observations on cosmetic appearance and organoleptic tests were made at initial stage and after storage. The study revealed that the quality of tender kind coconuts could be maintained for a period of four weeks when the whole nuts were wrapped with cling film and stored at 14-15C in order to meet export market requirements

    Aspects Associated with Nesting Ecology of Indian Blackbird (Turdus merula kinnisii) in Tropical Montane Cloud Forests of Horton Plains National Park of Sri Lanka

    Breeding ecology of Indian Blackbird (Turdus merula kinnisii) was studied at the tropical montane cloud forests of Horton Plains National Park, situated in the highland plateau of the central highlands, from September 2015 to August 2018 within three consecutive days per month. This sub species is endemic to montane forests of Sri Lanka. There are two breeding seasons from March to April and August to September. There was a research gap about the breeding ecology of Blackbird in Sri Lanka. The objective was to conduct a detailed study about breeding ecology. Three habitats were selected within HPNP as cloud forest, cloud forest die-back and grassland habitat. Nests were located by following individuals and searching the vegetation. The location of the nests were marked by a GPS (Garmin eTrex 10). Tree species, nest height, diameter at breast height (DBH) was measured after the birds leaving the nest. Nest concealment, distance from the nest tree to the adjacent tree, nearest road or walking path and water source, was measured. Disturbances for the nests were observed and recorded. Nests accessible from the ground were only taken for inner nest parameters. In this study 30 nests were observed. Most of the nests were located at cloud forest habitat (n=26).They preferred seven plant species to construct their nests. Neolitsea fuscata was their major nesting plant. The nest height was 3.87±1.42 m and the nesting tree height was 7.01±1.71 m. The nests were located in the middle of the trees (relative height=0.56±0.18). Nest concealment was 14.33±4.30%. They highly preferred medium sized trees which were situated at shady habitats near water sources. The nest was a densely built cup with a volume of 936.4±157.5 cm3. Sometimes there were dummy nests around the original nest to protect from predator attacks. They spent 7 to 9 days to complete the nest. The incubation ranged from 16 to 18 days. Nestling period ranged from 13 to 15 days. The adults considered sole care of the young until it become an adult. Nesting success was 73%. Corvus splendens was recorded as the major predator. Present study reveals that cloud forest habitat as the most suitable habitat to construct the nests of Blackbird.     EQ-5D-3L Derived Population Norms for Health Related Quality of Life in Sri Lanka

    Background Health Related Quality of Life (HRQoL) is an important outcome measure in health economic evaluation that guides health resource allocations. Population norms for HRQoL are an essential ingredient in health economics and in the evaluation of population health. The aim of this study was to produce EQ-5D-3L-derived population norms for Sri Lanka. Method A population sample (n =  780) was selected from four districts of Sri Lanka. A stratified cluster sampling approach with probability proportionate to size was employed. Twenty six clusters of 30 participants each were selected; each participant completed the EQ-5D-3L in a face-to-face interview. Utility weights for their EQ-5D-3L health states were assigned using the Sri Lankan EQ-5D-3L algorithm. The population norms are reported by age and socio-economic variables. Results The EQ-5D-3L was completed by 736 people, representing a 94% response rate. Sixty per cent of the sample reported being in full health. The percentage of people responding to any problems in the five EQ-5D-3L dimensions increased with age. The mean EQ-5D-3L weight was 0.85 (SD 0.008; 95%CI 0.84-0.87). The mean EQ-5D-3L weight was significantly associated with age, housing type, disease experience and religiosity. People above 70 years of age were 7.5 times more likely to report mobility problems and 3.7 times more likely to report pain/discomfort than those aged 18-29 years. Those with a tertiary education were five times less likely to report any HRQoL problems than those without a tertiary education. A person living in a shanty was 4.3 more likely to have problems in usual activities than a person living in a single house. Conclusion The population norms in Sri Lanka vary with socio-demographic characteristics. The socioeconomically disadvantaged have a lower HRQoL. The trends of population norms observed in this lower middle income country were generally similar to those previously reported in high income countries

    Short term effects on liver and renal functions following chemotherapy treatment for breast cancer patients in oncology clinic, university hospital Kotelawala Defence University in Sri Lanka

    Background: Breast cancer tops the global cancer incidence rates, having the highest rate of death among women. The primary objective of this study was to assess the impact of standard chemotherapy treatment dose adjusted for the Sri Lankan population, on hepatic and kidney function of breast cancer patients. Methods: The study conducted a cross-sectional, retrospective and prospective analysis of 75 breast cancer patients who received doxorubicin, cyclophosphamide, and paclitaxel chemotherapy regimen with normal liver and renal function at baseline at UHKDU oncology clinic. The study population had a mean age and BMI of 54.04±11.33 years and 26.7±3.89, respectively. Prior to starting the 16-cycle chemotherapy treatment, mean serum SGOT, SGPT, Creatinine, and eGFR values were 27.57 U/l, 31.32 U/l, 0.71 mg/dl, and 99.07 ml/minute/1.73 m2 respectively. Results: During the treatment, there was a statistically significant increase in the mean values of SGOT and SGPT (p<0.05), whereas there was no significant variation in the mean values of creatinine and eGFR (p>0.05) compared to the baseline results. The study identified a significant positive correlation in SGOT (r=0.793) and SGPT (r=0.872) values, while there was a noteworthy negative correlation (r=-0.757) between eGFR and chemotherapy cycle. Furthermore, there was a positive significant correlation between serum creatinine levels and chemotherapy cycle (r=0.579). Conclusions: The dosed adjusted chemotherapy regimen had a significant impact on hepatic function but had no statistically significant impact on renal function among the study population. Further research is recommended to evaluate the long-term effects of standard chemotherapy treatment on liver and kidney functions

    Odour-mediated orientation of beetles is influenced by age, sex and morph

    The behaviour of insects is dictated by a combination of factors and may vary considerably between individuals, but small insects are often considered en masse and thus these differences can be overlooked. For example, the cowpea bruchid Callosobruchus maculatus F. exists naturally in two adult forms: the active (flight) form for dispersal, and the inactive (flightless), more fecund but shorter-lived form. Given that these morphs show dissimilar biology, it is possible that they differ in odour-mediated orientation and yet studies of this species frequently neglect to distinguish morph type, or are carried out only on the inactive morph. Along with sex and age of individual, adult morph could be an important variable determining the biology of this and similar species, informing studies on evolution, ecology and pest management. We used an olfactometer with motion-tracking to investigate whether the olfactory behaviour and orientation of C. maculatus towards infested and uninfested cowpeas and a plant-derived repellent compound, methyl salicylate, differed between morphs or sexes. We found significant differences between the behaviour of male and female beetles and beetles of different ages, as well as interactive effects of sex, morph and age, in response to both host and repellent odours. This study demonstrates that behavioural experiments on insects should control for sex and age, while also considering differences between adult morphs where present in insect species. This finding has broad implications for fundamental entomological research, particularly when exploring the relationships between physiology, behaviour and evolutionary biology, and the application of crop protection strategies

    Cre-dependent DNA recombination activates a STING-dependent innate immune response

    Gene-recombinase technologies, such as Cre/loxP-mediated DNA recombination, are important tools in the study of gene function, but have potential side effects due to damaging activity on DNA. Here we show that DNA recombination by Cre instigates a robust antiviral response in mammalian cells, independent of legitimate loxP recombination. This is due to the recruitment of the cytosolic DNA sensor STING, concurrent with Cre-dependent DNA damage and the accumulation of cytoplasmic DNA. Importantly, we establish a direct interplay between this antiviral response and cell-cell interactions, indicating that low cell densities in vitro could be useful to help mitigate these effects of Cre. Taking into account the wide range of interferon stimulated genes that may be induced by the STING pathway, these results have broad implications in fields such as immunology, cancer biology, metabolism and stem cell research. Further, this study sets a precedent in the field of gene-engineering, possibly applicable to other enzymatic-based genome editing technologies
