    Elaboración y validación de un video educativo para prevenir la violencia sexual en adolescentes

    Objective: to evaluate the elaboration and validation process of an educational video for the prevention of sexual violence in school-age adolescents. Method: this is a methodological study, developed from September 2018 to February 2021 in Recife-PE. Production of the educational video was carried out according to the methodological path proposed by Kindem and Musberg. The video was submitted to content validation and face evaluation by expert judges and schoolage adolescents, respectively. In the data analysis, absolute frequency, Content Validity Index and binomial test were used. Results: in content validation, agreement was satisfactory and the mean validity index for all video items was 0.92. In the face evaluation, the adolescents considered the video understandable, with a minimum agreement level of 92.3%. Conclusion: the educational video elaborated was considered by the judges as valid to contribute to the knowledge and attitude of school-age adolescents regarding the prevention of sexual violence.Objetivo: avaliar o processo de construção e validação de um vídeo educacional para prevenção da violência sexual em adolescentes em idade escolar. Métodos: trata-se de um estudo metodológico, desenvolvido durante o período de setembro de 2018 a fevereiro de 2021 em Recife-PE. Foi realizada a produção de vídeo educacional de acordo com a trajetória metodológica proposta por Kindem e Musberg. O vídeo foi submetido a validação de conteúdo e avaliação de aparência, por juízes especialistas e adolescentes em idade escolar, respectivamente. Na análise dos dados, utilizou-se frequência absoluta, índice de validade de conteúdo e teste binomial. Resultados: na validação de conteúdo, a concordância foi satisfatória e a média do índice de validade para todos os itens do vídeo foi de 0,92. Na avaliação de aparência, os adolescentes consideraram o vídeo compreensível, com nível de concordância mínima de 92,3%. Resultados: en la validación del contenido, la concordancia fue satisfactoria y la media del índice de validez correspondiente a todos los ítems del video fue de 0,92. En la evaluación del aspecto, los adolescentes consideraron que el video es comprensible, con un nivel de concordancia mínima del 92,3%.     Bonding performance of universal adhesives with concomitant use of silanes to CAD/CAM blocks

    ABSTRACT Objective: This study assessed whether the use of a silane coupling agent influence the bond strength of two universal adhesives to ceramic or resin CAD/CAM blocks. Methods: Forty-eight samples were obtained from each resin nano-ceramic hybrid block (RCBs) and lithium disilicate ceramic blocks (LD). Samples were treated with silane with MDP (Monobond - S-MDP), silane without MDP (Prosil - PS), and no silane application (Control - Ctr) followed by universal adhesive with silane (UAS) and without silane (UA) (n=8). Three polyurethane tubes (1.5 mm of internal diameter) were positioned in each sample treated surface and filled with a dual cured resin cement. Bond strength was assessed by microshear bond strength test and failure analysis was performed for all samples. Results: For the RCBs, UAS presented the highest bond strength values (p=0.004). Silane application was not significant in bond strength values (p=0.444). For LD, silane application was significant in bond strength values (p<0.001), but the adhesive was not (p=0.066). Failure analysis showed high prevalence of adhesive failures for both substrates. Conclusion: A silane-containing universal adhesive promoted the best bond strength results to the resin nano-ceramic hybrid block. For bonding to a glass-ceramic CAD/CAM material, additional silane (without MDP) application presented the best results

    DL5o da bactéria Aeromonas hydrophila para o matrinxã, Brycon amazonicus

    In order to determine the lethal dose (96-h LD50) of the bacteria Aeromonas hydrophila to matrinxã, Brycon amazonicus, to be applied in challenge tests, 90 fish (63.23 ± 6.39 g) were divided into five treatments, with different bacterial solutionsT1 - Control (0.9% NaCl saline solution); T2 (4 x 10(11) cells/ mL); T3 (5 x 10(11) cells/ mL); T4 (1.36 x 10(12) cells/ mL) and T5 (3.06 x 10(12) cells/ mL). Fish were previously anesthetized with benzocaine (60 mg L-1), inoculated in the peritoneal cavity with the bacterial suspensions and then distributed into fifteen 80-L test chambers, where the water variables were monitored and fish mortality was observed. The experiment was randomly designed in three replicates and the 96-h LD50 was estimated according to the trimmed Spearman-Karber method. Water quality variables remained within adequate ranges for fish health and performance. Fish mortality rate increased with the bacterial concentrations of A. hydrophila (T1 = 0%; T2 = 16.66%; T3 = 44.44%; T4 = 72.22% and T5 = 100%), and the first mortalities were observed after 57 h, although the signs of the bacterial infection were already observed 24 h after the inoculation. The results indicate that the 96-h LD50 value of A. hydrophila to matrinxã is 6.66 x 10(11) cells/ mL.Para determinar a dose letal (DL50 96-h) da bactéria Aeromonas hydrophila para o matrinxã, Brycon amazonicus, com aplicabilidade para testes de desafio, foram utilizados 90 peixes (63,23 ± 6,39 g), divididos em cinco tratamentos, com diferentes soluções bacterianas: T1 - Controle (solução salina 0,9% NaCl); T2 (4 x 10(11) células/ mL); T3 (5 x 10(11) células/ mL-1); T4 (1,36 x 10(12) células/mL-1) e T5 (3,06 x 10(12) células/ mL-1). Os peixes foram previamente anestesiados com benzocaína (60 mg L-1), inoculados na cavidade peritoneal com as suspensões bacterianas e distribuídos em 15 aquários de vidro de 80 L de capacidade, com aeração constante. O experimento teve duração de 96 h, no qual foram monitoradas a mortalidade e a qualidade da água. O delineamento experimental foi inteiramente casualisado com três réplicas e a DL50 96-h foi estimada de acordo com o método Spearman-Karber. Durante o experimento os parâmetros físico-químicos da água permaneceram dentro das condições consideradas adequadas para o desenvolvimento e saúde dos organismos aquáticos. A mortalidade dos peixes aumentou nas concentrações crescentes de A. hydrophila (T1 = 0%; T2 = 16,66%; T3 = 44,44%; T4 = 72,22% e T5 = 100%), contudo, as primeiras mortalidades ocorreram em 57 h após a inoculação das concentrações bacterianas, sendo observados os primeiros sinais de infecção em 24 h após a inoculação. Os resultados indicam que o valor da DL50 96-h da bactéria A. hydrophila para o matrinxã foi igual a 6,66 x 10(11) células/mL de solução salina

    Avaliação da resposta inflamatória local após aplicação de células tronco mesenquimais alogênicas em tendão flexor digital superficial de equinos

    O tendão flexor digital superficial (TFDS) é uma estrutura importante para a espécie equina e para animais de alto desempenho atlético. Atualmente, a medicina regenerativa vem evoluindo de forma significativa no tratamento de diversas enfermidades, no entanto, o entendimento a respeito do comportamento celular quando do transplante de células tronco mesenquimais (CTMs) em tecido hígido ainda não é completamente conhecido. Sendo assim, este trabalho tem como objetivo a avaliação da resposta inflamatória local após aplicação de células tronco mesenquimais alogênicas derivadas do tecido adiposo (AdCTMs) em tendão equino comparado ao transplante autólogo e ao grupo controle. Para isso, 18 membros torácicos de 9 animais, divididos em três grupos, foram submetidos à aplicação de AdCTMs em tendão hígido, sendo que em Galog os animais receberam aplicação de AdCTMs alogênicas no membro torácico (MT); em Gauto, aplicação de células autólogas no MT e em Gcont aplicou-se o PBS como grupo controle. Esses animais foram avaliados por parâmetros físicos, exames ultrassonográficos e termográficos até o momento da biópsia, que foi realizada uma semana após aplicação das células autólogas e alogênicas para avaliar a possível reação inflamatória aguda, decorrente do implante de células no tecido tendíneo. O transplante alogênico de CTMs não resultou em reação adversa ou inflamatória que comprometesse o uso dessas células, elucidando a sua segurança neste trabalho. O seu comportamento mostrou-se semelhante ao implante autólogo, sendo assim, a formação de bancos de células alogênicas pode ser favorável e seguro.The superficial digital flexor tendon (SDFT) is an important structure for the equine species and for high performance animals. Currently, regenerative medicine has evolved significantly in the treatment of several diseases, even though the cellular behavior during the implantation of mesenchymal stem cells in healthy tissue is not completely known, despite the fact that it has been tirelessly studied in the last few years. This study aims to evaluate and compare the local inflammatory response upon injection in equine tendon of either allogeneic and autologous mesenchymal stem cells derived from adipose tissue (AdMSC). For this purpose, 18 forelimbs, 9 animals were submitted to the application of AdMSC in healthy tendon, which were divided into 3 groups: in Galog the animals received application of allogeneic AdMSC in the thoracic member; in Gauto, autologous AdMSC were injected in the thoracic member; in Gcont, PBS was implanted as control group. These animals were evaluated through physical exams, ultrasound, and thermography up until the moment of the biopsy, which was performed a week after the application of AdMSC in order to evaluate the inflammatory response due to the implantation of the cells in the tendinous tissue. Biopsy samples were taken to histopathology and immunohistochemistry in order to evaluate the actions of allogeneic AdMSC. The allogeneic MSCs implantation did not result in adverse or inflammatory reaction which could compromise the use of these cells, elucidating their safety in this work.     Ocular ultrasonography and ultrasound biomicroscopy: applicability in clinical and ophthalmic surgery

    With the advent of high frequency transducers, the use of ocular ultrasound has become widely used because it provides definitions of major ocular structures. This diagnostic tool is useful in the evaluation of ophthalmic injuries, and also used for qualitative and quantitative evaluation of intraocular and orbital lesions. It is a noninvasive, safe and fast, easy to use and can be performed in an awake animal. The ocular ultrasonography is indicated in cases of ocular opacity, when the ophthalmic examination cannot be done, and in eyes with ocular trauma or suspected orbital disease. Ultrasound biomicroscopy (UBM) is a diagnostic tool with high cost and sedation or general anesthesia is necessary, but has the advantage of evaluating structures poorly defined or visualized in the anterior segment of the eye. Therefore, this paper aims to show the importance of ocular ultrasonography and UBM to veterinary practitioners, and to demonstrate its advantages and indications in ophthalmic routine

    VDR polymorphisms influence immunological response in HIV-1+ individuals undergoing antiretroviral therapy

    Abstract Vitamin D exerts an immuno-modulatory activity on several immune system cells through the vitamin D receptor (VDR). Herein, we verified that age and a therapeutic regimen containing protease inhibitors are associated with failures in antiretroviral therapies (ARVs). In addition, we assessed whether a VDR SNP (rs11568820: C allele and CC genotype) and GC (rs2228570-rs11568820) allelic combinations are associated with immunological failure (p < 0.05). Our findings suggest a possible role of VDR SNPs on immunological failure in HIV-1+ individuals undergoing regular ARVs

    Culture of mesenchymal stem cells derived from equine synovial membrane in alginate hydrogel microcapsules

    Abstract Background Mesenchymal stem cells derived from the synovial membrane (MSCSM) have a greater potential for joint regeneration, besides the capacity for chondrogenic differentiation, since they are a source closer to the chondrocytes. This study aimed to cultivate and evaluate viability and differentiation of MSCSM encapsulated in a three-dimensional alginate hydrogel (HA) scaffold. Samples of the synovial membrane of the metatarsophalangeal joint of 4 horses were collected by astroscopic surgery. These were subjected to enzymatic digestion, isolated mesenchymal cells, cultured in monolayers and encapsulated at various concentrations, 104; 204; 504; 105; 205 cells in 1.5% sodium alginate solution. The gelatinization process was carried out and cultured for 4 weeks. Viability and cell proliferation were performed by dissolving the microcapsules and counting with trypan blue. The ratio of live cells and total live cells at intervals 0, 7, 14, 21 and 28 days was analyzed. Results For the evaluation of differentiation, histological sections stained with hematoxylin and eosin and toluidine blue were performed. There was no statistical difference in the proportion of live cells between groups over the 28 days. The group of 105 cells obtained a higher total number of living cells at the end of the experiment. Through the histological analysis it was possible to observe at 7 days a low amount of spherical cells with chondrocyte characteristics. On day 21, chondrogenic differentiation became evident, with pericellular and territorial matrix production. Conclusions This study demonstrated the efficiency of HA as a scaffold for MSCSM and the chondrogenic differentiation, promising for use in the treatment of joint injuries in horses

    Pleuris associada à criptococose em cão: relato de caso

    Cryptococcosis is a major infectious disease caused by a yeast from the Cryptococcus genre which affects different species including humans. There are few reports related to pulmonary cryptococcosis in dogs. A case of Cryptococcosis in a dog showing dyspnea was described. The diagnosis was done by aspiration cytology after exploratory thoracotomy, and a picture of severe pleurisy was observed. The diagnosis was done by needle aspiration cytology after thoracotomy. Severe pleuris frame was observed. Treatment with itraconazole was performed for 90 days, and at the end of the treatment the dog presented complete remission of reported symptoms.A Criptococose é uma importante doença infecciosa fúngica, causada por uma levedura do gênero Cryptococcus, que acomete diferentes espécies inclusive o homem. Há poucos relatos na literatura sobre a criptococose pulmonar em cães. O presente trabalho relata um caso de criptococose em um cão apresentando alterações respiratórias, especialmente dispneia. O diagnóstico foi realizado por meio da citologia aspirativa, após toracotomia exploratória, sendo observado um quadro de pleuris grave.
