    Results of abundance surveys of juvenile Atlantic and gulf Menhaden, Brevoortia tyrannus and B. patronus

    The estuarine populations of juvenile Atlantic and gulf menhaden (Brevoortia tyrannus and B. patronus) were sampled during two-boat, surface-trawl, abundance surveys extensively conducted in the 1970s. Juvenile Atlantic menhaden were sampled in 39 estuarine streams along the U.S. Atlantic coast from northern Florida into Massachusetts. Juvenile gulf menhaden were sampled in 29 estuarine streams along the Gulf of Mexico from southeast Texas into western Florida. A stratified, two-stage, cluster sampling design was used. Annual estimates of relative juvenile abundance for each species of menhaden were obtained from catch-effort data from the surveys. There were no significant correlations, for either species, between the relative juvenile abundance estimates and fishery-dependent estimates of year-class strength. From 1972 to 1975, the relative abundance of juvenile Atlantic menhaden in north Atlantic estuaries decreased to near zero. (PDF file contains 22 pages.

    Urine Specimens from Pregnant and Nonpregnant Women Inhibitory to Amplification of \u3cem\u3eChlamydia trachomatis\u3c/em\u3e Nucleic Acid by PCR, Ligase Chain Reaction, and Transcription-Mediated Amplification: Identification of Urinary Substances Associated with Inhibition and Removal of Inhibitory Activity

    The presence of endogenous amplification inhibitors in urine may produce false-negative results for the detection of Chlamydia trachomatis nucleic acids by tests such as PCR, ligase chain reaction (LCR), and transcription-mediated amplification (TMA). Consecutive urine specimens from 101 pregnant women and 287 nonpregnant women submitted for urinalysis were processed for C. trachomatis detection. Aliquots were spiked with the equivalent of one C. trachomatis elementary body and were tested by three commercial assays: AMPLICOR CT/NG, Chlamydia LCX, and Chlamydia TMA. The prevalence of inhibitors resulting in complete inhibition of amplification was 4.9% for PCR, 2.6% for LCR, and 7.5% for TMA. In addition, all three assays were partially inhibited by additional urine specimens. Only PCR was more often inhibited by urine from pregnant women than by urine from nonpregnant women (9.9 versus 3.1%; P = 0.011). A complete urinalysis including dipstick and a microscopic examination was performed. Logistic regression analysis revealed that the following substances were associated with amplification inhibition: beta-human chorionic gonadotropin (odd ratio [OR], 3.3) and crystals (OR, 3.3) for PCR, nitrites for LCR (OR, 14.4), and hemoglobin (OR, 3.3), nitrites (OR, 3.3), and crystals (OR, 3.3) for TMA. Aliquots of each inhibitory urine specimen were stored at 4 and -70°C and a dilution of 1:10 (84% for PCR, 100% for LCR, and 92% for TMA). Five urine specimens (three for PCR and two for TMA) required phenol-chloroform extraction to remove inhibitors. The results indicate that the prevalence of nucleic acid amplification inhibitors in female urine is different for each technology, that this prevalence may be predicted by the presence of urinary factors, and that storage and dilution remove most of the inhibitors

    Passive transfer and rate of decay of maternal antibody against African horse sickness virus in South African throughbred foals

    REASONS FOR PERFORMING STUDY : African horse sickness is an insect transmitted, non-contagious disease of equids caused by African horse sickness virus (AHSV). Mortality can exceed 90% in fully susceptible horse populations. A live-attenuated (modified-live) cell culture-adapted (MLV) polyvalent AHSV vaccine is widely used to control AHS in endemic areas in southern Africa. Field studies detailing antibody responses of vaccinated horses are lacking. OBJECTIVES : To determine antibody titres to the 9 known serotypes of AHSV in a cohort of brood mares that were regularly vaccinated with the MLV AHSV vaccine, and to measure the passive transfer and rate of decay of maternal antibody to the individual virus serotypes in foals. METHODS : Serum was collected from 15 mares before foaling and from their foals after foaling and monthly thereafter for 6 months. Antibody titres to each of the 9 AHSV serotypes were determined by serum-virus neutralisation assay. RESULTS : There was marked variation in the antibody response of the mares to individual AHSV serotypes even after repeated vaccination, with consistently higher titre responses to some virus serotypes. Similarly, duration of maternally-derived antibodies in foals differed among serotypes. CONCLUSIONS : Data from this study confirm variation of the neutralising antibody response of individual mares to repeated vaccination with polyvalent AHSV vaccine. Virus strains of individual AHSV serotypes included in the vaccine may vary in their inherent immunogenicity. Passivelyacquired maternal antibodies to AHSV vary markedly among foals born to vaccinated mares, with further variation in the duration of passive immunity to individual AHSV serotypes. POTENTIAL RELEVANCE : These data are relevant to the effective utilization of live-attenuated AHSV vaccines in endemic regions, and potentially to the use of vaccines in response to future incursions of AHSV into previously free regions.     Global Positioning System Derived Performance Measures Are Responsive Indicators of Physical Activity, Disease and the Success of Clinical Treatments in Domestic Dogs

    <div><p>Objective</p><p>To assess the use of Global Positioning System receiver (GPS) derived performance measures for differentiating between: 1) different outdoor activities in healthy dogs; 2) healthy dogs and those with osteoarthritis; 3) osteoarthritic dogs before and after treatment with non-steroidal anti-inflammatory analgesia.</p><p>Design</p><p>Prospective study.</p><p>Animals</p><p>Ten healthy dogs and seven dogs with osteoarthritis of the elbow joint (OA dogs).</p><p>Procedure</p><p>Healthy dogs were walked on a standard route on-lead, off-lead and subjected to playing activity (chasing a ball) whilst wearing a GPS collar. Each dog was walked for five consecutive days. Dogs with OA were subjected to a single off-lead walk whilst wearing a GPS collar, and then administered oral Carprofen analgesia daily for two weeks. OA dogs were then subjected to the same walk, again wearing a GPS collar.</p><p>Results</p><p>GPS derived measures of physical performance could differentiate between on-lead activity, off-lead activity and playing activity in healthy dogs, and between healthy dogs and OA dogs. Variation in the performance measures analysed was greater between individual dogs than for individual dogs on different days. Performance measures could differentiate healthy dogs from OA dogs. OA Dogs treated with Carprofen analgesia showed improvements in their physical performance, which returned to values indistinguishable from those of healthy dogs on nearly all the measures assessed.</p><p>Conclusions and Clinical Relevance</p><p>GPS derived measures of physical performance in dogs are objective, easy to quantify, and can be used to gauge the effects of disease and success of clinical treatments. Specific stimuli can be used to modulate physical performance beyond the self-governed boundaries that dogs will naturally express when allowed to exercise freely without stimulation.</p></div

    African horse sickness in naturally infected, immunised horses

    To determine whether subclinical cases, together with clinical cases, of African horse sickness (AHS) occur in immunised horses in field conditions, whole blood samples were collected and rectal temperatures recorded weekly from 50 Nooitgedacht ponies resident in open camps at the Faculty of Veterinary Science, University of Pretoria, Onderstepoort, during 2008–2010. The sampleswere tested for the presence of African horse sickness virus (AHSV) RNAby a recently developed real-time RT-PCR. Itwas shown that 16% of immunised horses in an AHS endemic areawere infected with AHSV over a 2 year period, with half of these (8%) being subclinically infected.
