Spastin couples microtubule severing to membrane traffic in completion of cytokinesis and secretion.
Mutations in the gene encoding the microtubule (MT)-severing protein spastin are the most common cause of hereditary spastic paraplegia, a genetic condition in which axons of the corticospinal tracts degenerate. We show that not only does endogenous spastin colocalize with MTs, but that it is also located on the early secretory pathway, can be recruited to endosomes and is present in the cytokinetic midbody. Spastin has two main isoforms, a 68 kD full-length isoform and a 60 kD short form. These two isoforms preferentially localize to different membrane traffic pathways with 68 kD spastin being principally located at the early secretory pathway, where it regulates endoplasmic reticulum-to-Golgi traffic. Sixty kiloDalton spastin is the major form recruited to endosomes and is also present in the midbody, where its localization requires the endosomal sorting complex required for transport-III-interacting MIT domain. Loss of midbody MTs accompanies the abscission stage of cytokinesis. In cells lacking spastin, a MT disruption event that normally accompanies abscission does not occur and abscission fails. We suggest that this event represents spastin-mediated MT severing. Our results support a model in which membrane traffic and MT regulation are coupled through spastin. This model is relevant in the axon, where there also is co-ordinated MT regulation and membrane traffic
Environmental Aging of Scotch-Weld(TradeMark) AF-555M Structural Adhesive in Composite to Composite Bonds
Fiber reinforced resin matrix composites have found increased usage in recent years. Due to the lack of service history of these relatively new material systems, their long-term aging performance is not well established. In this study, adhesive bonds were prepared by the secondary bonding of Scotch-Weld(TradeMark) AF-555M between pre-cured adherends comprised of T800H/3900-2 uni-directional laminate. The adherends were co-cured with wet peel-ply for surface preparation. Each bond-line of single-lap-shear (SLS) specimen was measured to determine thickness and inspected visually for voids. A three-year environmental aging plan for the SLS specimens at 82 C and 85% relative humidity was initiated. SLS strengths were measured for both controls and aged specimens at room temperature and 82 C. The aging results of strength retention and failure modes to date are reported

ESCRT-III-associated proteins and spastin inhibit protrudin-dependent polarised membrane traffic

Here, we show that HeLa cells lacking spastin and embryonic fibroblasts from a spastin knock-in mouse model become highly polarised and develop cellular protrusions. In HeLa cells, this phenotype was rescued by wild-type spastin, but not by forms unable to sever microtubules or interact with endosomal ESCRT-III proteins. Cells lacking the spastin-interacting ESCRT-III-associated proteins IST1 or CHMP1B also developed protrusions. The protrusion phenotype required protrudin, a RAB-interacting protein that interacts with spastin and localises to ER–endosome contact sites, where it promotes KIF5-dependent endosomal motility to protrusions. Consistent with this, the protrusion phenotype in cells lacking spastin also required KIF5. Lack or mutation of spastin resulted in functional consequences for receptor traffic of a pathway implicated in HSP, as Bone Morphogenetic Protein receptor distribution became polarised. Our results, therefore, identify a novel role for ESCRT-III proteins and spastin in regulating polarised membrane traffic
CAPG is required for Ebola virus infection by controlling virus egress from infected cells
The replication of Ebola virus (EBOV) is dependent upon actin functionality, especially at cell entry through macropinocytosis and at release of virus from cells. Previously, major actin-regulatory factors involved in actin nucleation, such as Rac1 and Arp2/3, were shown important in both steps. However, downstream of nucleation, many other cell factors are needed to control actin dynamics. How these regulate EBOV infection remains largely unclear. Here, we identified the actin-regulating protein, CAPG, as important for EBOV replication. Notably, knockdown of CAPG specifically inhibited viral infectivity and yield of infectious particles. Cell-based mechanistic analysis revealed a requirement of CAPG for virus production from infected cells. Proximity ligation and split-green fluorescent protein reconstitution assays revealed strong association of CAPG with VP40 that was mediated through the S1 domain of CAPG. Overall, CAPG is a novel host factor regulating EBOV infection through connecting actin filament stabilization to viral egress from cells
A perspective on using experiment and theory to identify design principles in dye-sensitized solar cells
Dye-sensitized solar cells (DSCs) have been the subject of wide-ranging studies for many
years because of their potential for large-scale manufacturing using roll-to-roll processing
allied to their use of earth abundant raw materials. Two main challenges exist for DSC
devices to achieve this goal; uplifting device efficiency from the 12 to 14% currently
achieved for laboratory-scale ‘hero’ cells and replacement of the widely-used liquid
electrolytes which can limit device lifetimes. To increase device efficiency requires optimized
dye injection and regeneration, most likely from multiple dyes while replacement
of liquid electrolytes requires solid charge transporters (most likely hole transport materials
– HTMs). While theoretical and experimental work have both been widely applied to
different aspects of DSC research, these approaches are most effective when working in
tandem. In this context, this perspective paper considers the key parameters which
influence electron transfer processes in DSC devices using one or more dye molecules
and how modelling and experimental approaches can work together to optimize electron
injection and dye regeneration.

A Cognitive Model of an Epistemic Community: Mapping the Dynamics of Shallow Lake Ecosystems
We used fuzzy cognitive mapping (FCM) to develop a generic shallow lake
ecosystem model by augmenting the individual cognitive maps drawn by 8
scientists working in the area of shallow lake ecology. We calculated graph
theoretical indices of the individual cognitive maps and the collective
cognitive map produced by augmentation. The graph theoretical indices revealed
internal cycles showing non-linear dynamics in the shallow lake ecosystem. The
ecological processes were organized democratically without a top-down
hierarchical structure. The steady state condition of the generic model was a
characteristic turbid shallow lake ecosystem since there were no dynamic
environmental changes that could cause shifts between a turbid and a clearwater
state, and the generic model indicated that only a dynamic disturbance regime
could maintain the clearwater state. The model developed herein captured the
empirical behavior of shallow lakes, and contained the basic model of the
Alternative Stable States Theory. In addition, our model expanded the basic
model by quantifying the relative effects of connections and by extending it.
In our expanded model we ran 4 simulations: harvesting submerged plants,
nutrient reduction, fish removal without nutrient reduction, and
biomanipulation. Only biomanipulation, which included fish removal and nutrient
reduction, had the potential to shift the turbid state into clearwater state.
The structure and relationships in the generic model as well as the outcomes of
the management simulations were supported by actual field studies in shallow
lake ecosystems. Thus, fuzzy cognitive mapping methodology enabled us to
understand the complex structure of shallow lake ecosystems as a whole and
obtain a valid generic model based on tacit knowledge of experts in the field.Comment: 24 pages, 5 Figure
Political masculinities, crisis tendencies, and social transition: Toward an understanding of change
This introduction to the special issue on “Political Masculinities and Social Transition” rethinks the notion of “crisis in masculinity” and points to its weaknesses, such as cyclical patterns and chronicity. Rather than viewing key moments in history as points of rupture, we understand social change as encompassing ongoing transitions marked by a “fluid nature” (Montecinos 2017, 2). In line with this, the contributions examine how political masculinities are implicated within a wide range of social transitions, such as nation building after war, the founding of a new political party in response to an economic crisis, an “authoritarian relapse” of a democracy, attempts at changing society through terrorism, rapid industrialization as well as peace building in conflict areas. Building on Starck and Sauer’s definition of “political masculinities” we suggest applying the concept to instances in which power is explicitly either being (re)produced or challenged. We distinguish between political masculinities that are more readily identified as such (e.g., professional politicians) and less readily identified political masculinities (e.g., citizens), emphasizing how these interact with each other. We ask whether there is a discernible trajectory in the characteristics of political masculinities brought about by social transition that can be confirmed across cultures. The contributors’ findings indicate that these political masculinities can contribute to different kinds of change that either maintain the status quo, are progressive, retrogressive, or a mixture of these. Revolutionary transitions, it seems, often promote the adherence to traditional forms of political masculinity, whereas more reformatory transition leaves discursive spaces for argument
