    Ignition characteristics of the nickel-based alloy UNS N07718 in pressurized oxygen

    The development of ignition and combustion in pressurized oxygen atmospheres was studied for the nickel based alloy UNS N07718. Ignition of the alloy was achieved by heating the top. It was found that the alloy would autoheat to destruction from temperatures below the solidus temperature. In addition, endothermic events occurred as the alloy was heated, many at reproducible temperatures. Many endothermic events occurred prior to abrupt increases in surface temperature and appeared to accelerate the rate of increase in specimen temperature. It appeared that the source of some endotherms may increase the oxidation rate of the alloy. Ignition parameters are defined and the temperatures at which these parameters occur are given for the oxygen pressure range of 1.72 to 13.8 MPa (250 to 2000 psia)

    The development of ignition and combustion in pressurized oxygen atmospheres was studied for the iron based alloy UNS S66286. Ignition of the alloy was achieved by heating the top surface of a cylindrical specimen with a continuous-wave CO2 laser. Two heating procedures were used. In the first, laser power was adjusted to maintain an approximately linear increase in surface temperature. In the second, laser power was periodically increased until autoheating (self-heating) was established. It was found that the alloy would autoheat to destruction from temperatures below the solidus temperature. In addition endothermic events occurred as the alloy was heated, many at reproducible temperatures. Many endothermic events occurred prior to abrupt increases in surface temperature and appeared to accelerate the rate of increase in specimen temperature to rates greater than what would be expected from increased temperature alone. It is suggested that the source of these endotherms may increase the oxidation rate of the alloy. Ignition parameters are defined and the temperatures at which these parameters occur are given for the oxygen pressure range of 1.72 to 13.8 MPa (25 to 2000 psia)

    Tariff-Rate Quotas : Difficult to model or plain simple?

    The difficulty of reliably and accurately incorporating tariffrate quotas (TRQs) into trade models has received a lot of attention in recent years. As a result of the Uruguay Round of GATT negotiations, TRQs replaced an assortment of tariff and nontariff instruments in an effort to standardise trade barriers, and facilitate their future liberalisation. Understanding the nuances of TRQs is now particularly crucial for New Zealand because of the preferential access arrangements that New Zealand has for a number of products in highly protected markets such as the European Union, Japan, and the United States. It has been argued that TRQs are complex instruments and are difficult to model because for any trade flow between two countries, one of three regimes may be applicable : 1. The import quota may not be binding and the within-quota tariff applies; 2. The quota may be binding, the within-quota tariff applies, and a quota rent is created; or 3. Trade occurs over and above the quota, in which case an over-quota tariff applies (although, even in this regime, someone is still able to collect the quota rent on within-quota trade). But even this characterisation, which many claim is too complex to model, is a major simplification of reality. Bilateral preferences are ubiquitous, and such preferences are usually included in the determination of multilateral market access quotas. It is usual, therefore, that the TRQ instrument has several tiers to the quota schedule, plus a number of within and over-quota tariff rates applicable on either a bilateral or a multilateral basis. Further trade liberalisation creates something of a dilemma for New Zealand. Any decrease in over-quota tariffs and/or increase in quota levels potentially reduces the value of quota rents, many of which accrue to New Zealand due to the bilateral preferences. It is important, therefore, that New Zealand trade negotiators understand how much additional trade is required to offset the loss of New Zealands quota rents. Modelling trade in the presence of TRQs is the only way to ascertain this knowledge. The purpose of this paper is to show that complex TRQs can be modelled very easily and precisely. The only catch is that the model must be formulated as a complementarity problem rather than the more conventional linear or nonlinear optimisation problem.     Consistent massive truncations of IIB supergravity on Sasaki-Einstein manifolds

    Recent work on holographic superconductivity and gravitational duals of systems with non-relativistic conformal symmetry have made use of consistent truncations of D=10 and D=11 supergravity retaining some massive modes in the Kaluza-Klein tower. In this paper we focus on reductions of IIB supergravity to five dimensions on a Sasaki-Einstein manifold, and extend these previous truncations to encompass the entire bosonic sector of gauged D=5, N=2 supergravity coupled to massive multiplets up to the second Kaluza-Klein level. We conjecture that a necessary condition for the consistency of massive truncations is to only retain the lowest modes in the massive trajectories of the Kaluza-Klein mode decomposition of the original fields.     Generation of functional CD8+ T Cells by human dendritic cells expressing glypican-3 epitopes

    <p>Abstract</p> <p>Background</p> <p>Glypican 3 (GPC-3) is an oncofoetal protein that is expressed in most hepatocellular carcinomas (HCC). Since it is a potential target for T cell immunotherapy, we investigated the generation of functional, GPC-3 specific T cells from peripheral blood mononuclear cells (PBMC).</p> <p>Methods</p> <p>Dendritic cells (DC) were derived from adherent PBMC cultured at 37°C for 7 days in X-Vivo, 1% autologous plasma, and 800 u/ml GM-CSF plus 500 u/ml IL-4. Immature DC were transfected with 20 μg of <it>in vitro </it>synthesised GPC-3 mRNA by electroporation using the Easy-ject plus system (Equibio, UK) (300 V, 150 μF and 4 ms pulse time), or pulsed with peptide, and subsequently matured with lipopolysaccharide (LPS). Six predicted GPC-3 peptide epitopes were synthesized using standard f-moc technology and tested for their binding affinity to HLA-A2.1 molecules using the cell line T2.</p> <p>Results</p> <p>DC transfected with GPC-3 mRNA but not control DC demonstrated strong intracellular staining for GPC-3 and <it>in vitro </it>generated interferon-gamma expressing T cells from autologous PBMC harvested from normal subjects. One peptide, GPC-3<sub>522-530 </sub>FLAELAYDL, fulfilled our criteria as a naturally processed, HLA-A2-restricted cytotoxic T lymphocyte (CTL) epitope: i) it showed high affinity binding to HLA-A2, in T2 cell binding assay; ii) it was generated by the MHC class I processing pathway in DC transfected with GPC-3 mRNA, and iii) HLA-A2 positive DC loaded with the peptide stimulated proliferation in autologous T cells and generated CTL that lysed HLA-A2 and GPC-3 positive target cells.</p> <p>Conclusions</p> <p>These findings demonstrate that electroporation of GPC-3 mRNA is an efficient method to load human monocyte-derived DC with antigen because <it>in vitro </it>they generated GPC-3-reactive T cells that were functional, as shown by interferon-gamma production. Furthermore, this study identified a novel naturally processed, HLA-A2-restricted CTL epitope, GPC-3<sub>522-530 </sub>FLAELAYDL, which can be used to monitor HLA-A2-restricted CTL responses in patients with HCC. Further studies are required to investigate whether anti-GPC-3 immunotherapy has a role in the treatment of GPC-3 dependent tumours, such as HCC.</p

    Mathematical programming models, as typically formulated for international trade applications, may contain certain implied restrictions limiting price responsiveness, intermediate product flows, and arbitrage possibilities. These restrictions are especially important in the case of dairy, and may lead to results which are technically infeasible, or if feasible, not consistent with market equilibrating behavior. The difficulties encountered when modeling dairy trade are described, and an alternative formulation of a spatial model is presented.     Ethanol Affects the Development of Sensory Hair Cells in Larval Zebrafish (Danio rerio)

    Children born to mothers with substantial alcohol consumption during pregnancy can present a number of morphological, cognitive, and sensory abnormalities, including hearing deficits, collectively known as fetal alcohol syndrome (FAS). The goal of this study was to determine if the zebrafish lateral line could be used to study sensory hair cell abnormalities caused by exposure to ethanol during embryogenesis. Some lateral line sensory hair cells are present at 2 days post-fertilization (dpf) and are functional by 5 dpf. Zebrafish embryos were raised in fish water supplemented with varying concentrations of ethanol (0.75%–1.75% by volume) from 2 dpf through 5 dpf. Ethanol treatment during development resulted in many physical abnormalities characteristic of FAS in humans. Also, the number of sensory hair cells decreased as the concentration of ethanol increased in a dose-dependent manner. The dye FM 1-43FX was used to detect the presence of functional mechanotransduction channels. The percentage of FM 1-43-labeled hair cells decreased as the concentration of ethanol increased. Methanol treatment did not affect the development of hair cells. The cell cycle markers proliferating cell nuclear antigen (PCNA) and bromodeoxyuridine (BrdU) demonstrated that ethanol reduced the number of sensory hair cells, as a consequence of decreased cellular proliferation. There was also a significant increase in the rate of apoptosis, as determined by TUNEL-labeling, in neuromasts following ethanol treatment during larval development. Therefore, zebrafish are a useful animal model to study the effects of hair cell developmental disorders associated with FAS

    Colonization of \u3ci\u3eLutzomyia verrucarum\u3c/i\u3e and \u3ci\u3eLutzomyia longipalpis\u3c/i\u3e Sand Flies (Diptera: Psychodidae) by \u3ci\u3eBartonella bacilliformis\u3c/i\u3e, the Etiologic Agent of Carrion\u27s Disease

    Bartonella bacilliformis is a pathogenic bacterium transmitted to humans presumably by bites of phlebotomine sand flies, infection with which results in a bi-phasic syndrome termed Carrion\u27s disease. After constructing a low-passage GFP-labeled strain of B. bacilliformis, we artificially infected Lutzomyia verrucarum and L. longipalpis populations, and subsequently monitored colonization of sand flies by fluorescence microscopy. Initially, colonization of the two fly species was indistinguishable, with bacteria exhibiting a high degree of motility, yet still confined to the abdominal midgut. After 48h, B. bacilliformis transitioned from bacillus-shape to a non-motile, small coccoid form and appeared to be digested along with the blood meal in both fly species. Differences in colonization patterns became evident at 72h with B. bacilliformis was observed at relatively high density outside the peritrophic membrane in the lumen of the midgut in L. verrucarum, but colonization of L. longipalpis was limited to the blood meal within the intra-peritrophic space of the abdominal midgut, and the majority of bacteria were digested along with the blood meal by day 7. The viability of B. bacilliformis in L. longipalpis was assessed by artificially infecting, homogenizing, and plating for determination of colony-forming units in individual flies over a 13-d time course. Bacteria remained viable at relatively high density for approximately seven days, suggesting that L. longipalpis couple potentially serve as a vector. The capacity of L. longipalpis to transmit viable B. bacilliformis from infected to uninfected meals was analyzed via interrupted feeds. No viable bacteria were retrieved from uninfected blood meals in these experiments. This study provides significant information toward understanding colonization of sand flies by B. bacilliformis and also demonstrates the utility of L. longipalpis as a user-friendly, live-vector model system for studying this severely neglected tropical disease
