    Identifying genetic variants associated with multiple correlated traits and the use of an ensemble of genetic risk models for phenotype prediction and classification

    Sickle cell disease is a monogenic blood disorder in which the clinical course and disease severity vary widely among patients. In order for physicians to make more informed decisions regarding the treatment and management of disease, it would be useful to be able to predict disease severity. We focus on two primary modulators of disease severity in sickle cell patients, hemolysis and fetal hemoglobin (HbF). This dissertation evaluates methodology to identify genetic variants associated with severity of sickle cell disease and develops new methodology of genetic risk prediction to predict disease severity in sickle cell patients based on levels of HbF. Hemolysis is a trait that is influenced by multiple correlated phenotypes (lactate dehydrogenase, reticulocytes, bilirubin and aspartate transaminase). There are several approaches to statistical analyses of multiple correlated phenotypes. The first part of this dissertation evaluates the use of principal component analysis (PCA) and compares it to the alternative approach of examining the results of multiple univariate phenotypes individually. We will focus on the question of if and under what conditions we gain more power using a summarized phenotype from PCA in a genome wide association study (GWAS) rather than conducting multiple individual GWAS. We find that the there is more power gained from the PCA approach when there is a strong intercorrelation between the phenotypes. The second part of this dissertation proposes a novel method of genetic risk prediction for continuous traits using an ensemble of genetic models. We aim to show through a simulation and prediction of HbF that the proposed method is more robust to the inclusion of false positives and yields more stable predictions than computing a GRS and 10 fold cross validation. The third part of this dissertation introduces a Bayesian-based clustering approach to produce clusters of sickle cell anemia patients based on their "predicted genetic profiles" of HbF. We then examine the genetic profiles of individuals in the extreme clusters to determine which genes contribute more prominently to the genetic profile so that we may potentially identify genes that are highly influential in the regulation of extremely high and low values of HbF

    El presente trabajo de investigación lleva por título Evasión Tributaria y la Recaudación de Impuestos en el Emporio Comercial De Gamarra, La Victoria – 2020. Esta investigación tiene por finalidad saber de qué manera la evasión tributaria se relaciona con la recaudación de impuestos en el emporio comercial de Gamarra, La Victoria -2020. Tuvo como objetivo general determinar de qué manera la evasión tributaria se relaciona con la recaudación de impuestos en el emporio comercial de Gamarra, La Victoria -2020. La metodología que se uso fue de tipo aplicada, con un diseño no experimental, con un enfoque cuantitativo. Su población estuvo conformada por los comerciantes del emporio de Gamarra, la técnica utilizada fue la encuesta, para la comprobación de la hipótesis se utilizó la correlación de Pearson, donde se logró encontrar una relación positiva, entre las variables de estudio (Evasión tributaria y Recaudación de impuestos). Finalmente, se concluye que existe relación entre la evasión tributaria y la recaudación de impuestos en el emporio comercial de Gamarra, La Victoria -2020

    En el presente trabajo se desarrolla la eficacia de la imprescriptibilidad de la cuota de alimentos a partir de la reforma del artículo 261 del Código de Familia teniendo en consideración el ámbito histórico del derecho de alimentos, lo establecido por la doctrina en virtud del derecho de alimentos, como también en el término de prescripción del derecho de alimentos, realizando a su vez un estudio en el ámbito legal y jurisprudencial para tener de esa manera conocimiento en cuanto a la aplicación de esta reforma, de esta manera al considerar el ámbito de desarrollo se tiene en cuenta los incidentes que se obtienen en cuanto a la eficacia de esta nueva reforma y el estudio del derecho comparado teniendo de esa manera un conocimiento amplio de la aplicación del Derecho a nivel internacional. Después de conocer de manera singular cada apartado del presente trabajo se tiene en consideración que la imprescriptibilidad en la cuota de alimentos resulta ineficaz ya que brinda inseguridad jurídica al alimentante estableciendo una deuda perpetua para su persona, siendo está una prohibición establecida en nuestra Constitución de la República

    Assessing the effect of adverse economic events on severity of hunger among food pantry clients

    This study assessed relationship between adverse economic events (AEE) and hunger level (i.e., little to no, moderate, severe). A cross-sectional survey was conducted from June to August 2018 in 10 food pantries with 616 food pantry users. Hunger level was assessed by the Household Hunger Scale. AEE were evaluated over the past 3 months. Participants (60.55%) experienced unexpected or increased medical expenses (17.69%), job loss (13.64%), pay reduction (11.85%), and death of a family member (9.09%). Pay reduction (OR = 1.87, 95% CI: 1.12, 3.14) and increased debt (OR = 2.71, 95% CI: 1.92, 3.84) were associated with moderate hunger; death of a family member (OR = 2.43, 95% CI: 1.21, 4.90), pay reduction (OR = 2.95, 95% CI: 1.24, 7.04), and increased debt (OR = 3.46, 95% CI: 1.98, 6.04) were associated with severe hunger. Awareness of AEE can inform public health programs and policies for people in need of additional resources, which is essential in times of increased economic instability
