    Avaliaram-se os resultados clínicos após realização de cirurgia descompressiva em 45 cães com doença do disco intervertebral cervical ou toracolombar. Após a cirurgia, 35 cães (77,8%) recuperaram-se totalmente, oito (17,8%) parcialmente e dois (4,4%) não apresentaram alteração do quadro inicial. Em oito cães com paraplegia e perda da sensibilidade dolorosa profunda houve completa melhora do quadro clínico, com recuperação total em 62,5% dos casos. Em quatro cães com tetraparesia, a cirurgia foi eficaz. A cirurgia descompressiva (slot cervical e hemilaminectomia toracolombar), com a retirada do material do disco do interior do canal vertebral, foi uma forma efetiva de gerar melhora do quadro funcional.<br>Clinical results after decompressive surgery were evaluated in 45 dogs with cervical or thoracolumbar intervertebral disk disease. After surgery, 35 dogs recovered totally, eight (17.8%) partially, and two (4.4%) did not present any change in clinical findings. Eight dogs with paraplegy and loss of deep pain perception showed improvement, with total recovering in 62.5% of cases. Surgery was effective in four dogs with tetraparesy. Decompressive surgery (cervical slot or hemilaminectomy), with removal of disk material from inside the vertebral canal, was an effective form to produce functional improvement in dogs with this disease

    Adolescent substance use in the multimodal treatment study of attention-deficit/hyperactivity disorder (ADHD) (MTA) as a function of childhood ADHD, random assignment to childhood treatments, and subsequent medication

    Objective: To determine long-term effects on substance use and substance use disorder (SUD), up to 8 years after childhood enrollment, of the randomly assigned 14-month treatments in the multisite Multimodal Treatment Study of Children with Attention-Deficit/Hyperactivity Disorder (MTA; n = 436); to test whether medication at follow-up, cumulative psychostimulant treatment over time, or both relate to substance use/SUD; and to compare substance use/SUD in the ADHD sample to the non-ADHD childhood classmate comparison group (n = 261). Method: Mixed-effects regression models with planned contrasts were used for all tests except the important cumulative stimulant treatment question, for which propensity score matching analysis was used. Results: The originally randomized treatment groups did not differ significantly on substance use/SUD by the 8-year follow-up or earlier (mean age = 17 years). Neither medication at follow-up (mostly stimulants) nor cumulative stimulant treatment was associated with adolescent substance use/SUD. Substance use at all time points, including use of two or more substances and SUD, were each greater in the ADHD than in the non-ADHD samples, regardless of sex. Conclusions: Medication for ADHD did not protect from, or contribute to, visible risk of substance use or SUD by adolescence, whether analyzed as randomized treatment assignment in childhood, as medication at follow-up, or as cumulative stimulant treatment over an 8-year follow-up from childhood.     Radiation-induced Activation of Nuclear Factor-κB Involves Selective Degradation of Plasma Membrane-associated IκBα

    In contrast to nuclear factor-κB (NF-κB) activation by tumor necrosis factor-α (TNF-α), the specific processes involved in the activation of this transcription factor by ionizing radiation (IR) have not been completely defined. According to the classical paradigm, a critical event in NF-κB activation is the degradation of IκBα. Data presented herein show that, in contrast to treatment with TNF-α, IR-induced NF-κB activation was not accompanied by degradation of IκBα in the U251 glioblastoma cell line as determined in whole cell lysates. However, treatment with the proteosome inhibitor MG-132 inhibited NF-κB activation induced by IR, suggesting that IκBα degradation was a critical event in this process. To reconcile these results, U251 cell lysates were separated into soluble and insoluble fractions and IκBα levels evaluated. Although IκBα was found in both subcellular fractions, treatment with IR resulted in the degradation of IκBα only in the insoluble fraction. Further subcellular fractionation suggested that the IR-sensitive, insoluble pool of IκBα was associated with the plasma membrane.     Em 10 cães com peso médio de 14,6kg, as vértebras cervicais 5 e 6 foram cirurgicamente desestabilizadas através da secção do disco intervertebral e, em seguida, estabilizadas com placas ortopédicas confeccionadas com PVC de 2mm de espessura, para após 180 dias, proceder-se ao estudo histológico do tecido ósseo e conjuntivo circunvizinho. Constatou-se que o PVC causou alterações ósseas que podem ter favorecido o afrouxamento dos parafusos e a falha do implante. O material induziu ainda à formação de granuloma de corpo estranho e a reações inflamatórias locais que podem ter causado degradação do material implantado. Assim, placas de PVC, apesar de proporcionarem estabilidade e alinhamento da coluna vertebral, não satisfazem a maioria das propriedades necessárias a um biomaterial, não sendo recomendadaa a sua utilização em ortopedia veterinária.<br>In ten dogs with an average mean weight of 14,6 kg, the cervical vertebra 5 and 6 were destabilized and fixed with plates of 2mm of thickness. The purpose of this work was to verify the effect of orthopedic PVC plates on the internal stabilization of the caudal cervical spine of dogs by studying the occurrence of alterations in the bone tissue and fibrous tissue adjacent to the plate after 180 days of permanence of the material in the dogs' organism. PVC causes progressive bone alterations, which, in the long term, could promote the loosening of the screws and failure of the implant. It also induces the formation of foreign body granuloma and inflammatory reactions which could cause degradation of the implant. Thus, PVC plates do not satisfy the majority of properties required of a biomaterial, its use not being recommended in veterinary orthopedics
