    Sustainability aspects of biobased applications : comparison of different crops and products from the sugar platform BO-12.05-002-008

    In this study different uses of biomass are compared. In order to allow for a systematic comparison the study focuses on three different chemicals that can be produced from sugar. In this way it is also, in principle, possible to compare different crops for the production of the same product. The study focuses on the production of PLA (polylactic acid, a bioplastic), ethanol, and biopolyethylene (bio-PE, which is produced via ethanol). These three products can presently be produced from biomass and therefore form realistic cases. All three products are produced from sugars, and thus the systems can be decoupled at the sugar step. The sugar can be produced from different crops. In this study five different crops are compared: wheat, maize, sugar beet, sugar cane and Miscanthus. The sustainability aspects that we studied are non-renewable energy use (NREU), greenhouse gas (GHG) emission in the crop-product chain and direct land use for producing the bio-materials

    In deze studie is onderzocht hoe de duurzaamheid van verschillende biobased producten die uit fermenteerbare suikers kunnen worden gemaakt zich tot elkaar verhoudt. Daarbij is gekeken naar het gebruik van niet-hernieuwbare energie en de uitstoot van broeikasgassen tijdens het gehele productieproces, inclusief het telen van de gewassen. Er zijn vijf verschillende gewassen met elkaar vergeleken. In de studie zijn verschillende invalshoeken gekozen, waarbij in het ene geval er voor wordt gekozen de co-producten te verzamelen en om te zetten in energie, en in het andere geval de huidige landbouwpraktijk wordt aangenomen, waarbij co-producten deels op het land blijven liggen. Verder wordt het effect getoond van verschillende manieren om de data te presenteren. Drie manieren van presenteren leiden schijnbaar tot verschillende conclusies

    This study focusses on the production of vegetable oil based products. A limited number of aspacts of the sustainability of the full chain (from agriculture to product at the factory gate) was evaluated. Three different vegetable oils were taken into account: palm oil, soy oil and rapeseed oil. Also three different products made from vegetable oil were evaluated: biodiesel, polyol (a raw material for production of PU foams) and resin. In the present study, sustainability of these products was evaluated through the following parameters: Non-Renewable Energy Usage (NREU) (GJ per ton of product); Greenhouse Gas (GHG) emissions (ton CO2-equivalent per ton of product); Land use (ha/ton vegetable oil) and NREU and GHG emission avoided (GJ or ton CO2-equivalent per hectare of land)

    Dit boek gaat in op een vergelijking van producten uit natuurlijke olie. Drie oliegewassen zijn in de studie vergeleken: oliepalm, koolzaad en soja. Uit deze gewassen kan een veelheid van biobased producten gemaakt worden: biodiesel, polyol voor polyurethaan en harsen zijn in de studie als eindproduct omschreven. Voor alle onderzochte producten geldt, dat vervanging van een fossiel product door een vergelijkbaar biobased product leidt tot een verlaging van de uitstoot van broeikasgassen en van het gebruik van fossiele energie

    Resonant transmission through an open quantum dot

    We have measured the low-temperature transport properties of a quantum dot formed in a one-dimensional channel. In zero magnetic field this device shows quantized ballistic conductance plateaus with resonant tunneling peaks in each transition region between plateaus. Studies of this structure as a function of applied perpendicular magnetic field and source-drain bias indicate that resonant structure deriving from tightly bound states is split by Coulomb charging at zero magnetic field.Comment: To be published in Phys.     Co-activation of NF-ÎșB and MYC renders cancer cells addicted to IL6 for survival and phenotypic stability

    NF-ÎșB and MYC are found co-deregulated in human B and plasma-cell cancers. In physiology, NF-ÎșB is necessary for terminal B-to-plasma cell differentiation, whereas MYC repression is required. It is thus unclear if NF-ÎșB/MYC co-deregulation is developmentally compatible in carcinogenesis and/or impacts cancer cell differentiation state, possibly uncovering unique sensitivities. Using a mouse system to trace cell lineage and oncogene activation we found that NF-ÎșB/MYC co-deregulation originated cancers with a plasmablast-like phenotype, alike human plasmablastic-lymphoma and was linked to t(8;14)[MYC-IGH] multiple myeloma. Notably, in contrast to NF-ÎșB or MYC activation alone, co-deregulation rendered cells addicted to IL6 for survival and phenotypic stability. We propose that conflicting oncogene-driven differentiation pressures can be accommodated at a cost in poorly-differentiated cancers. SIGNIFICANCE: Our studies improve the understanding of cancer pathogenesis by demonstrating that co-deregulation of NF-ÎșB and MYC synergize in forming a cancer with a poorly-differentiated state. The cancers in the mouse system share features with human Plasmablastic lymphoma that has a dismal prognosis and no standard of care, and with t(8;14)[MYC-IGH] Multiple myeloma, which is in overall resistant to standard therapy. Notably, we found that NF-ÎșB and MYC co-deregulation uniquely render cells sensitive to IL6 deprivation, providing a road-map for patient selection. Because of the similarity of the cancers arising in the compound mutant mouse model with that of human Plasmablastic lymphoma and t(8;14)[MYC-IGH] Multiple myeloma, this model could serve in preclinical testing to investigate novel therapies for these hard-to-treat diseases

    VEGAS as a Platform for Facile Directed Evolution in Mammalian Cells

    Directed evolution, artificial selection toward designed objectives, is routinely used to develop new molecular tools and therapeutics. Successful directed molecular evolution campaigns repeatedly test diverse sequences with a designed selective pressure. Unicellular organisms and their viral pathogens are exceptional for this purpose and have been used for decades. However, many desirable targets of directed evolution perform poorly or unnaturally in unicellular backgrounds. Here, we present a system for facile directed evolution in mammalian cells. Using the RNA alphavirus Sindbis as a vector for heredity and diversity, we achieved 24-h selection cycles surpassing 10−3 mutations per base. Selection is achieved through genetically actuated sequences internal to the host cell, thus the system's name: viral evolution of genetically actuating sequences, or “VEGAS.” Using VEGAS, we evolve transcription factors, GPCRs, and allosteric nanobodies toward functional signaling endpoints each in less than 1 weeks’ time. © 2019 Elsevier Inc.The VEGAS system is a platform for directed evolution, a method for engineering DNA sequences, in mammalian cells. The system is highly mutagenic, facile, and self-contained, requiring no in vitro handling during evolution cycles. As a result, robust evolution campaigns can be run within the context of a mammalian cell signaling environment. We perform three such campaigns as a proof-of-concept: evolving a transcription factor, a G-protein coupled receptor, and llama-derived nanobodies toward specific in vivo activities. © 2019 Elsevier Inc

    “A few sparks of inspiration”?: Analysing the outcomes of European Union cultural policy coordination

    This article examines the outcomes of cultural policy coordination in the European Union using a case study of one policy priority in the 2011–2014 Work Plan for Culture. The Open Method of Coordination brings Member States together to exchange and cooperate on key policy priorities. Drawing on interviews with key actors as well as participant observation material, the article demonstrates the limited influence of the culture OMC on domestic policy, showing that domestic usage tends to be on the scale of individuals and organisations rather than Member State-wide. The article finishes by contextualising the outcomes, highlighting the constraints and challenges of intergovernmental coordination in fields where the EU holds a supporting competence
