    A Critical Policy Analysis of Kentucky School Districts’ Trauma-Informed Education Plans

    The Commonwealth of Kentucky has taken its first steps in creating trauma-informed K-12 schools through passing the School Safety and Resiliency Act of 2019. School districts were tasked with developing trauma-informed education plans by July 2021. However, the Commonwealth’s mandate gave broad autonomy to school districts in shaping their trauma-informed education plans’ processes and content. Through a critical policy analysis design, this study examined the availability and quality of trauma-informed education plans from each school district in Kentucky and compared them to the trauma-informed framework developed by the National Child Traumatic Stress Network. Findings indicate that only 19 out of 171 school districts published their trauma-informed education plans publicly. Out of those 19, only three met all the criteria recommended by the National Child Traumatic Stress Network. There is a discussion of how a lack of state-level guidance impacts the quality of trauma-informed education plans followed by recommendations for improving these plans

    This study explored Kentucky school administrators’ perceptions, knowledge, and beliefs about trauma-informed practices, competence in promoting these practices, and the extent to which school administration preparation programs provided relevant training. Participants reported they were not adequately trained in their school administration programs and believed they needed additional training and support to implement trauma-informed practices. The Culturally Responsive, Trauma-informed Educator Identity framework outlines essential knowledge and skills needed to promote trauma-informed practices, and implications for critical supervision are explored

    Pre-service Teachers’ Experiences with Multi-Tiered Systems of Support: Universal Screening & Intervention Planning

    Assessment plays a key role in the multi-tiered system of support (MTSS) model. Universal screening is an assessment component embedded into MTSS, specifically within the response to intervention framework. The process of administering, scoring, and analyzing data from a universal screener is an essential skill that pre-service teachers need to gain in their educator preparation programs. In this qualitative, case study approach, we examined the ways an experimental learning project in implementing universal screeners impact pre-service teachers’ beliefs and practices for Tier 1 of MTSS. The project was provided to pre-service teachers to give them familiarization with the implementation of universal screeners and how to use data to make informed instructional decisions regarding interventions. Results from participants indicate four themes including: 1) confidence in administrating a universal screener, 2) belief that no student is left behind, 3) benefits in knowing student progress, and 4) the value of hands-on experience

    PRESERVICE TEACHERS’ PERCEPTIONS AND KNOWLEDGE OF RESPONSE TO INTERVENTION/MULTITIERED SYSTEMS OF SUPPORT

    There has been considerable research that establishes the need to improve teachers’ knowledge of and ability to effectively implement response to intervention (RtI)/multi-tiered systems of support (MTSS), and there is a scarcity of research examining interventions addressing these concerns. In a mixed methods study, we examined the perceptions and knowledge of the RtI/MTSS frameworks of undergraduate preservice teaching candidates enrolled in a dual certification program at a small, private Catholic university in Kentucky, before and after participating in a semester-long, experiential learning project. The project involved monitoring both the reading and mathematics progress of struggling elementary or middle school-aged students using validated Curriculum Based Measures. Preservice teachers worked in concert with the cooperating classroom teacher and their professor to score, graph, and interpret CBM results and make recommendations for high-quality, research-based intervention. Results from participants indicate their concern that not all teachers will enter the profession with knowledge of RtI/MTSS. Further, the experiential learning project intervention enhanced preservice teachers’ perceptions and knowledge of RtI/MTSS frameworks

    The signature sequence region of the human drug transporter organic anion transporting polypeptide 1B1 Is important for protein surface expression

    The organic anion transporting polypeptides (OATPs) encompass a family of membrane transport proteins responsible for the uptake of xenobiotic compounds. Human organic anion transporting polypeptide 1B1 (OATP1B1) mediates the uptake of clinically relevant compounds such as statins and chemotherapeutic agents into hepatocytes, playing an important role in drug delivery and detoxification. The OATPs have a putative 12-transmembrane domain topology and a highly conserved signature sequence (human OATP1B1: DSRWVGAWWLNFL), spanning the extracellular loop 3/TM6 boundary. The presence of three conserved tryptophan residues at the TM interface suggests a structural role for the sequence. This was investigated by site-directed mutagenesis of selected amino acids within the sequence D251E, W254F, W258/259F, and N261A. Transport was measured using the substrate estrone-3-sulfate and surface expression detected by luminometry and confocal microscopy, facilitated by an extracellular FLAG epitope. Uptake of estrone-3-sulfate and the surface expression of D251E, W254F, and W258/259F were both significantly reduced from the wild type OATP1B1-FLAG in transfected HEK293T cells. Confocal microscopy revealed that protein was produced but was retained intracellularly. The uptake and expression of N261A were not significantly different. The reduction in surface expression and intracellular protein retention indicates a structural and/or membrane localization role for these signature sequence residues in the human drug transporter OATP1B1

    Ubicomp for animal welfare: envisioning smart environments for kenneled dogs

    Whilst the ubicomp community has successfully embraced a number of societal challenges for human benefit, including healthcare and sustainability, the well-being of other animals is hitherto underrepresented. We argue that ubicomp technologies, including sensing and monitoring devices as well as tangible and embodied interfaces, could make a valuable contribution to animal welfare. This paper particularly focuses on dogs in kenneled accommodation, as we investigate the opportunities and challenges for a smart kennel aiming to foster canine welfare. We conducted an in-depth ethnographic study of a dog rehoming center over four months; based on our findings, we propose a welfare centered framework for designing smart environments, integrating monitoring and interaction with information management. We discuss the methodological issues we encountered during the research and propose a smart ethnographic approach for similar projects

    The neonicotinoid imidacloprid, and the pyrethroid deltamethrin, are antagonists of the insect Rdl GABA receptor

    A mutation in the second transmembrane domain of the GABA receptor subunit, Rdl, is associated with resistance to insecticides such as dieldrin and fipronil. Molecular cloning of Rdl cDNA from a strain of the malaria mosquito, Anopheles gambiae, which is highly resistant to dieldrin revealed this mutation (A296G) as well as another mutation in the third transmembrane domain (T345M). Wild-type, A296G, T345M and A296G + T345M homomultimeric Rdl were expressed in Xenopus laevis oocytes and their sensitivities to fipronil, deltamethrin, 1,1,1-trichloro-2,2-bis(p-chlorophenyl) ethane (DDT), imidacloprid and spinosad were measured using two-electrode voltage-clamp electrophysiology. Spinosad and DDT had no agonist or antagonist actions on Rdl. However, fipronil, deltamethrin and imidacloprid decreased GABA-evoked currents. These antagonistic actions were either reduced or abolished with the A296G and the A296G + T345M mutations while T345M alone appeared to have no significant effect. In conclusion, this study identifies another mutation in the mosquito Rdl that is associated with insecticide resistance. While T345M itself does not affect insecticide sensitivity, it may serve to offset the structural impact of A296G. The present study also highlights Rdl as a potential secondary target for neonicotinoids and pyrethroids

    Dysfunction of cortical GABAergic neurons leads to sensory hyper-reactivity in a Shank3 mouse model of ASD.

    Hyper-reactivity to sensory input is a common and debilitating symptom in individuals with autism spectrum disorders (ASD), but the neural basis underlying sensory abnormality is not completely understood. Here we examined the neural representations of sensory perception in the neocortex of a Shank3B-/- mouse model of ASD. Male and female Shank3B-/- mice were more sensitive to relatively weak tactile stimulation in a vibrissa motion detection task. In vivo population calcium imaging in vibrissa primary somatosensory cortex (vS1) revealed increased spontaneous and stimulus-evoked firing in pyramidal neurons but reduced activity in interneurons. Preferential deletion of Shank3 in vS1 inhibitory interneurons led to pyramidal neuron hyperactivity and increased stimulus sensitivity in the vibrissa motion detection task. These findings provide evidence that cortical GABAergic interneuron dysfunction plays a key role in sensory hyper-reactivity in a Shank3 mouse model of ASD and identify a potential cellular target for exploring therapeutic interventions

    On Virtual Displacement and Virtual Work in Lagrangian Dynamics

    The confusion and ambiguity encountered by students, in understanding virtual displacement and virtual work, is discussed in this article. A definition of virtual displacement is presented that allows one to express them explicitly for holonomic (velocity independent), non-holonomic (velocity dependent), scleronomous (time independent) and rheonomous (time dependent) constraints. It is observed that for holonomic, scleronomous constraints, the virtual displacements are the displacements allowed by the constraints. However, this is not so for a general class of constraints. For simple physical systems, it is shown that, the work done by the constraint forces on virtual displacements is zero. This motivates Lagrange's extension of d'Alembert's principle to system of particles in constrained motion. However a similar zero work principle does not hold for the allowed displacements. It is also demonstrated that d'Alembert's principle of zero virtual work is necessary for the solvability of a constrained mechanical problem. We identify this special class of constraints, physically realized and solvable, as {\it the ideal constraints}. The concept of virtual displacement and the principle of zero virtual work by constraint forces are central to both Lagrange's method of undetermined multipliers, and Lagrange's equations in generalized coordinates.Comment: 12 pages, 10 figures.     Body shape and size in 6-year old children: assessment by three-dimensional photonic scanning.

    BACKGROUND: Body shape and size are typically described using measures such as body mass index (BMI) and waist circumference, which predict disease risks in adults. However, this approach may underestimate the true variability in childhood body shape and size. OBJECTIVE: To use a comprehensive three-dimensional photonic scan approach to describe variation in childhood body shape and size. SUBJECTS/METHODS: At age 6 years, 3350 children from the population-based 2004 Pelotas birth cohort study were assessed by three-dimensional photonic scanner, traditional anthropometry and dual X-ray absorptiometry. Principal component analysis (PCA) was performed on height and 24 photonic scan variables (circumferences, lengths/widths, volumes and surface areas). RESULTS: PCA identified four independent components of children's body shape and size, which we termed: Corpulence, Central:peripheral ratio, Height and arm lengths, and Shoulder diameter. Corpulence showed strong correlations with traditional anthropometric and body composition measures (r>0.90 with weight, BMI, waist circumference and fat mass; r>0.70 with height, lean mass and bone mass); in contrast, the other three components showed weak or moderate correlations with those measures (all r<0.45). There was no sex difference in Corpulence, but boys had higher Central:peripheral ratio, Height and arm lengths and Shoulder diameter values than girls. Furthermore, children with low birth weight had lower Corpulence and Height and arm lengths but higher Central:peripheral ratio and Shoulder diameter than other children. Children from high socio-economic position (SEP) families had higher Corpulence and Height and arm lengths than other children. Finally, white children had higher Corpulence and Central:peripheral ratio than mixed or black children. CONCLUSIONS: Comprehensive assessment by three-dimensional photonic scanning identified components of childhood body shape and size not captured by traditional anthropometry or body composition measures. Differences in these novel components by sex, birth weight, SEP and skin colour may indicate their potential relevance to disease risks.This article is based on data from the study ‘Pelotas Birth Cohort, 2004’ conducted by the Postgraduate Program in Epidemiology at Federal University of Pelotas, in collaboration with Brazilian Public Health Association (ABRASCO).
