    Alterations of the CARD15/NOD2 gene and the impact on management and treatment of Crohn's disease patients

    The recent identification of the CARD15/NOD2 gene as a susceptibility locus for Crohn's disease represents an important step towards the delineation of the immuno-pathogenesis of inflammatory bowel disease. CARD15 functions as an intracellular receptor for bacterial components and thus represents an important link between inflammatory bowel disease and innate immunity. Three major CARD15/NOD2 gene mutations have been associated with Crohn's disease in Caucasians in several independent studies. Together, they explain about 20% of the genetic susceptibility for Crohn's disease. Genotype-phenotype analyses demonstrated an association of these mutations with ileum-specific disease, an increased incidence of the fibrostenotic phenotype and an earlier age of disease onset. Beside these associations, no other relationship between the CARD15/NOD2 genotype and disease behavior or response to treatment has been detailed so far. Thus, the clinical impact of knowing the patient's genotype is limited at this time. Screening for CARD15 mutations in order to identify high-risk individuals or to introduce an individualized disease management is therefore currently not recommended. Copyright (C) 2003 S.     Variants of the human PPARG locus and the susceptibility to chronic periodontitis

    Apart from its regulatory function in lipid and glucose metabolism, peroxisome proliferator-activated receptor (PPAR)γ has impact on the regulation of inflammation and bone metabolism. The aim of the study was to investigate the association of five polymorphisms (rs10865710, rs2067819, rs3892175, rs1801282, rs3856806) within the PPARG gene with chronic periodontitis. The study population comprised 402 periodontitis patients and 793 healthy individuals. Genotyping of the PPARG gene polymorphisms was performed by PCR and melting curve analysis. Comparison of frequency distribution of genotypes between individuals with periodontal disease and healthy controls for the polymorphism rs3856806 showed a P-value of 0.04 but failed to reach significance after correction for multiple testing (P  0.90). A 3-site analysis (rs2067819-rs1801282-rs3856860) revealed five haplotypes with a frequency of ≥1% among cases and controls. Following adjustment for age, gender and smoking, none of the haplotypes was significantly different between periodontitis and healthy controls after Bonferroni correction.     Anti-Saccharomyces cerevisiae antibodies in patients with inflammatory bowel disease and their first-degree relatives: Potential clinical value

    Anti-Saccharomyces cerevisiae antibodies (ASCA) have been described as specific markers in Crohn's disease and their healthy first-degree relatives. 171 patients with Crohn's disease, their 105 first-degree relatives, 145 patients with ulcerative colitis and 101 first-degree relatives of patients with ulcerative colitis, 50 patients with infectious enterocolitis and 100 healthy controls were tested for ASCA employing the ELISA technique. When compared with the healthy controls (p < 0.0001) and patients with infectious enterocolitis (p < 0.0001) the prevalence of ASCA was significantly increased in patients with Crohn's disease and their first-degree relatives (p < 0.01). Further significant differences concerning the frequency of ASCA within the different groups of our study population were not observed. In particular, ASCA were not found in increased prevalence in infectious enterocolitis. These observations are compatible with a role of ASCA as a marker of genetic predisposition to Crohn's disease.     Adaptive mastery testing using the Rasch model and Bayesian sequential decision theory

    A version of sequential mastery testing is studied in which response behavior is modeled by an item response theory (IRT) model. First, a general theoretical framework is sketched that is based on a combination of Bayesian sequential decision theory and item response theory. A discussion follows on how IRT based sequential mastery testing can be generalized to adaptive item and testlet selection rules; i.e., to a situation in which the choice of the next item or testlet to be administered is optimized using the information from previous responses. The performance of IRT based sequential and adaptive sequential mastery testing is studied in a number of simulations using the Rasch model. The possibilities and difficulties of application of the approach in the framework of the two-parameter logistic and three-parameter logistic models is also discussed

    Determination of the local concentrations of Mn interstitials and antisite defects in GaMnAs

    We present a method for the determination of the local concentrations of interstitial and substitutional Mn atoms and As antisite defects in GaMnAs. The method relies on the sensitivity of the structure factors of weak reflections to the concentrations and locations of these minority constituents. High spatial resolution is obtained by combining structure factor measurement and X-ray analysis in a transmission electron microscope. We demonstrate the prevalence of interstitials with As nearest neighbors in as-grown layers.Comment: 10 pages, 4 figure

    El Plan de Desarrollo Académico de la Universidad Nacional de Avellaneda consta de diferentes programas y proyectos que se pusieron en marcha en el 2011 como resultado del análisis de los factores indispensables a atender por la Secretaría Académica en el marco de una política de inclusión social. Con el objeto de cumplir con la gestión en el primer año de constitución de la Universidad de acuerdo con el Proyecto Institucional Universitario, los cursos de acción propuestos se hallan atravesados por los principios institucionales: Promoción de la calidad, entendida como inclusión, la mejora continua, la flexibilidad y la innovación. En esta comunicación desarrollamos tres de los siete programas implementados, por ser éstos los que atienden al acceso y permanencia de una población heterogénea, mayoritariamente constituida por estudiantes que son primera generación de universitarios. Los programas en cuestión son: Ingreso y nivelación, apoyo a los estudiantes de grado y pregrado y apoyo, seguimiento y evaluación de la enseñanza

    Capitalization on item calibration error in computer adaptive testing

    In adaptive testing, item selection is sequentially optimized during the test. Since the optimization takes place over a pool of items calibrated with estimation error, capitalization on these errors is likely to occur. How serious the consequences of this phenomenon are depends not only on the distribution of the estimation errors in the pool or the ratio of the test length to the pool size, but also on the structure of the item-selection criteria used. A simulation study demonstrated the existence of the phenomenon empirically. It also showed that its effect on the errors in the ability estimates interacts strongly with the distribution of the items in the pool
