    Meteorological and operational aspects of 46 clear air turbulence sampling missions with an instrument B-57B aircraft. Volume 1: Program summary

    The results of 46 clear air turbulence (CAT) probing missions conducted with an extensively instrumented B-57B aircraft are summarized. Turbulence samples were obtained under diverse conditions including mountain waves, jet streams, upper level fronts and troughs, and low altitude mechanical and thermal turbulence. CAT was encouraged on 20 flights comprising 77 data runs. In all, approximately 4335 km were flown in light turbulence, 1415 km in moderate turbulence, and 255 km in severe turbulence during the program.     An ultrahigh-speed digitizer for the Harvard College Observatory astronomical plates

    A machine capable of digitizing two 8 inch by 10 inch (203 mm by 254 mm) glass astrophotographic plates or a single 14 inch by 17 inch (356 mm by 432 mm) plate at a resolution of 11 microns per pixel or 2309 dots per inch (dpi) in 92 seconds is described. The purpose of the machine is to digitize the \~500,000 plate collection of the Harvard College Observatory in a five year time frame. The digitization must meet the requirements for scientific work in astrometry, photometry, and archival preservation of the plates.     Development and flight tests of vortex-attenuating splines

    The ground tests and full-scale flight tests conducted during development of the vortex-attenuating spline are described. The flight tests were conducted using a vortex generating aircraft with and without splines; a second aircraft was used to probe the vortices generated in both cases. The results showed that splines significantly reduced the vortex effects, but resulted in some noise and climb performance penalties on the generating aircraft

    Flight tests evaluating the effects of altered span loading, turbulence ingestion, combinations of mass and turbulence ingestion, and combinations of altered span loading turbulance ingestion on trailed wake vortex attenuation were conducted. Span loadings were altered in flight by varying the deflections of the inboard and outboard flaps on a B-747 aircraft. Turbulence ingestion was achieved in flight by mounting splines on a C-54G aircraft. Mass and turbulence ingestion was achieved in flight by varying the thrust on the B-747 aircraft. Combinations of altered span loading and turbulence ingestion were achieved in flight by installing a spoiler on a CV-990 aircraft and by deflecting the existing spoilers on a B-747 aircraft. The characteristics of the attenuated and unattenuated vortexes were determined by probing them with smaller aircraft. Acceptable separation distances for encounters with the attenuated and unattenuated vortexes are presented

    Lifetimes in \u3csup\u3e124\u3c/sup\u3eTe: Examining Critical-Point Symmetry in the Te Nuclei

    The Doppler-shift attenuation method following inelastic neutron scattering was used to determine the lifetimes of nuclear levels to 3.3-MeV excitation in 124Te. Level energies and spins, γ-ray energies and branching ratios, and multipole-mixing ratios were deduced from measured γ-ray angular distributions at incident neutron energies of 2.40 and 3.30 MeV, γ-ray excitation functions, and γγ coincidence measurements. The newly obtained reduced transition probabilities and level energies for 124Te were compared to critical-point symmetry model predictions. The E(5) and β4 potential critical-point symmetries were also investigated in 122Te and 126Te

    Tissue-Specific Remodeling of the Mitochondrial Proteome in Type 1 Diabetic Akita Mice

    ObjectiveTo elucidate the molecular basis for mitochondrial dysfunction, which has been implicated in the pathogenesis of diabetes complications.Research design and methodsMitochondrial matrix and membrane fractions were generated from liver, brain, heart, and kidney of wild-type and type 1 diabetic Akita mice. Comparative proteomics was performed using label-free proteome expression analysis. Mitochondrial state 3 respirations and ATP synthesis were measured, and mitochondrial morphology was evaluated by electron microscopy. Expression of genes that regulate mitochondrial biogenesis, substrate utilization, and oxidative phosphorylation (OXPHOS) were determined.ResultsIn diabetic mice, fatty acid oxidation (FAO) proteins were less abundant in liver mitochondria, whereas FAO protein content was induced in mitochondria from all other tissues. Kidney mitochondria showed coordinate induction of tricarboxylic acid (TCA) cycle enzymes, whereas TCA cycle proteins were repressed in cardiac mitochondria. Levels of OXPHOS subunits were coordinately increased in liver mitochondria, whereas mitochondria of other tissues were unaffected. Mitochondrial respiration, ATP synthesis, and morphology were unaffected in liver and kidney mitochondria. In contrast, state 3 respirations, ATP synthesis, and mitochondrial cristae density were decreased in cardiac mitochondria and were accompanied by coordinate repression of OXPHOS and peroxisome proliferator-activated receptor (PPAR)-gamma coactivator (PGC)-1alpha transcripts.ConclusionsType 1 diabetes causes tissue-specific remodeling of the mitochondrial proteome. Preservation of mitochondrial function in kidney, brain, and liver, versus mitochondrial dysfunction in the heart, supports a central role for mitochondrial dysfunction in diabetic cardiomyopathy

Wrote the paper: JJP MEP PC BCW.BackgroundHorizontal gene transfer (HGT) plays a major role in speciation and evolution of bacteria and archaea by controlling gene distribution within an environment. However, information that links HGT to a natural community using relevant population-genetics parameters and spatial considerations is scarce. The Great Salt Lake (Utah, USA) provides an excellent model for studying HGT in the context of biogeography because it is a contiguous system with dispersal limitations due to a strong selective salinity gradient. We hypothesize that in spite of the barrier to phylogenetic dispersal, functional characteristics—in the form of HGT—expand beyond phylogenetic limitations due to selective pressure.Methodology and ResultsTo assay the functional genes and microorganisms throughout the GSL, we used a 16S rRNA oligonucleotide microarray (Phylochip) and a functional gene array (GeoChip) to measure biogeographic patterns of nine microbial communities. We found a significant difference in biogeography based on microarray analyses when comparing Sørensen similarity values for presence/absence of function and phylogeny (Student's t-test; p = 0.005).Conclusion and SignificanceBiogeographic patterns exhibit behavior associated with horizontal gene transfer in that informational genes (16S rRNA) have a lower similarity than functional genes, and functional similarity is positively correlated with lake-wide selective pressure. Specifically, high concentrations of chromium throughout GSL correspond to an average similarity of chromium resistance genes that is 22% higher than taxonomic similarity. This suggests active HGT may be measured at the population level in microbial communities and these biogeographic patterns may serve as a model to study bacteria adaptation and speciation.Yeshttp://www.plosone.org/static/editorial#pee

    Acetate-grown GS-15 whole-cell suspensions were disrupted with detergent and assayed for enzymes associated with acetate catabolism. Carbon monoxide dehydrogenase and formate dehydrogenase were not observed in GS-15. Catabolic levels of acetokinase and phosphotransacetylase were observed. Enzyme activities of the citric acid cycle, i.e., isocitrate dehydrogenase, 2-oxoglutarate sythase, succinate dehydrogenase, fumarase, and malate dehydrogenase, were observed
