    Addendum: "The Dynamics of M15: Observations of the Velocity Dispersion Profile and Fokker-Planck Models" (ApJ, 481, 267 [1997])

    It has recently come to our attention that there are axis scale errors in three of the figures of Dull et al. (1997, hereafter D97). D97 presented Fokker-Planck models for the collapsed-core globular cluster M15 that include a dense, centrally concentrated population of neutron stars and massive white dwarfs, but do not include a central black hole. In this Addendum, we present corrected versions of Figures 9, 10, and 12, and an expanded version of Figure 6. This latter figure, which shows the full run of the velocity dispersion profile, indicates that the D97 model predictions are in good agreement with the moderately rising HST-STIS velocity dispersion profile for M15 reported by Gerssen et al. (2002, astro-ph/0209315). Thus, a central black hole is not required to fit the new STIS velocity measurements, provided that there is a sufficient population of neutron stars and massive white dwarfs. This conclusion is consistent with the findings of Gerssen et al. (2002, astro-ph/0210158), based on a reapplication of their Jeans equation analysis using the corrected mass-to-light profile (Figure 12) for the D97 models.Comment: 4 pages, 4 figures, submitted to Ap

    External Mass Accumulation onto Core Potentials: Implications for Star Clusters, Galaxies and Galaxy Clusters

    Accretion studies have been focused on the flow around bodies with point mass gravitational potentials, but few general results are available for non-point mass distributions. Here, we study the accretion flow onto non-divergent, core potentials moving through a background medium. We use Plummer and Hernquist potentials as examples to study gas accretion onto star clusters, dwarf and large galaxy halos and galaxy clusters in a variety of astrophysical environments. The general conditions required for a core potential to collectively accrete large quantities of gas from the external medium are derived using both simulations and analytic results. The consequences of large mass accumulation in galaxy nuclei, dwarf galaxies and star clusters are twofold. First, if the gas cools effectively star formation can be triggered, generating new stellar members in the system. Second, if the collective potential of the system is able to alter the ambient gas properties before the gas is accreted onto the individual core members, the augmented mass supply rates could significantly alter the state of the various accreting stellar populations and result in an enhanced central black hole accretion luminosity.Comment: 24 pages, 15 figures, accepted to Ap

    Measuring gravitational waves from binary black hole coalescences: I. Signal to noise for inspiral, merger, and ringdown

    We estimate the expected signal-to-noise ratios (SNRs) from the three phases (inspiral,merger,ringdown) of coalescing binary black holes (BBHs) for initial and advanced ground-based interferometers (LIGO/VIRGO) and for space-based interferometers (LISA). LIGO/VIRGO can do moderate SNR (a few tens), moderate accuracy studies of BBH coalescences in the mass range of a few to about 2000 solar masses; LISA can do high SNR (of order 10^4) high accuracy studies in the mass range of about 10^5 to 10^8 solar masses. BBHs might well be the first sources detected by LIGO/VIRGO: they are visible to much larger distances (up to 500 Mpc by initial interferometers) than coalescing neutron star binaries (heretofore regarded as the "bread and butter" workhorse source for LIGO/VIRGO, visible to about 30 Mpc by initial interferometers). Low-mass BBHs (up to 50 solar masses for initial LIGO interferometers; 100 for advanced; 10^6 for LISA) are best searched for via their well-understood inspiral waves; higher mass BBHs must be searched for via their poorly understood merger waves and/or their well-understood ringdown waves. A matched filtering search for massive BBHs based on ringdown waves should be capable of finding BBHs in the mass range of about 100 to 700 solar masses out to 200 Mpc (initial LIGO interferometers), and 200 to 3000 solar masses out to about z=1 (advanced interferometers). The required number of templates is of order 6000 or less. Searches based on merger waves could increase the number of detected massive BBHs by a factor of order 10 or more over those found from inspiral and ringdown waves, without detailed knowledge of the waveform shapes, using a "noise monitoring" search algorithm. By exploring the concept of a circular supply chain, the paper illustrates the role of supply chain professionals in operationalising circular economy initiatives within their organisations, as well as the opportunities and challenges they may encounter along the way. The paper also provides initialrecommendations for and examples of companies overcoming some of these challenges, based on the experiences of supply chain professionals involved in the research

    Content and Quality of Motor Initiatives in the Support of People With Profound Intellectual and Multiple Disabilities

    Motor activation is rarely integrated into the support of people with profound intellectual and multiple disabilities (PIMD), which might be the result of the limited evidence‐based knowledge in this field. Practitioners have recently been developing several motor initiatives for people with PIMD, but it remains unclear about what core elements the motor initiatives actually consist of and to what level of quality it is implemented in practice. This study aims to offer an overview and analysis of the content and quality of motor initiatives actually in use for people with PIMD. Motor initiatives were explored by asking practitioners to complete an online inventory form. Documents, expert knowledge, and observations were used to collect data about the characteristics of the motor initiatives. The quality of the motor initiatives which met our eligibility criteria, was analyzed on the basis of the level of evidence for their effectiveness. The inventory yielded 118 motor initiatives of which 17 met the eligibility criteria. We identified four motor initiatives reflecting an approach to motorically activate people with PIMD within various activities, three including power‐assisted exercises, three with aquatic exercises, two frameworks which integrated motor activities into their daily programs, two methods which included small‐scale activities, two rhythmic movement therapies, and one program including gross motor activities. We found limited indications for descriptive evidence from 17 initiatives, limited or no indications for theoretical evidence from 12 and five initiatives respectively, and none of the initiatives provided a causal level of evidence for effectiveness. A wide variety of motor initiatives is used in current practice to activate persons with PIMD, although their effectiveness is actually unproven. Science and practice should cooperate to develop an evidence‐based understanding to ensure more evidence‐based support for the motor activation of people with PIMD in the future

    Separating Lentiviral Vector Injection and Induction of Gene Expression in Time, Does Not Prevent an Immune Response to rtTA in Rats

    BACKGROUND: Lentiviral gene transfer can provide long-term expression of therapeutic genes such as erythropoietin. Because overexpression of erythropoietin can be toxic, regulated expression is needed. Doxycycline inducible vectors can regulate expression of therapeutic transgenes efficiently. However, because they express an immunogenic transactivator (rtTA), their utility for gene therapy is limited. In addition to immunogenic proteins that are expressed from inducible vectors, injection of the vector itself is likely to elicit an immune response because viral capsid proteins will induce "danger signals" that trigger an innate response and recruit inflammatory cells. METHODOLOGY AND PRINCIPAL FINDINGS: We have developed an autoregulatory lentiviral vector in which basal expression of rtTA is very low. This enabled us to temporally separate the injection of virus and the expression of the therapeutic gene and rtTA. Wistar rats were injected with an autoregulatory rat erythropoietin expression vector. Two or six weeks after injection, erythropoietin expression was induced by doxycycline. This resulted in an increase of the hematocrit, irrespective of the timing of the induction. However, most rats only responded once to doxycycline administration. Antibodies against rtTA were detected in the early and late induction groups. CONCLUSIONS: Our results suggest that, even when viral vector capsid proteins have disappeared, expression of foreign proteins in muscle will lead to an immune respons

    Impact of a quadrivalent meningococcal ACWY glycoconjugate or a serogroup B meningococcal vaccine on meningococcal carriage: an observer-blind, phase 3 randomised clinical trial

    Background: Meningococcal conjugate vaccines protect individuals directly, but also confer herd protection by interrupting carriage transmission. This Phase III observer-blind, randomised, controlled study evaluated the effects of meningococcal quadrivalent (ACWY) glycoconjugate (MenACWY-CRM) or serogroup B (4CMenB) vaccination on meningococcal carriage rates in young adults. Methods: University students (aged 18–24 years) from ten sites in England were randomised to receive two vaccinations one month apart: two doses of Japanese Encephalitis vaccine (controls), two doses of 4CMenB (4CMenB), or one dose of MenACWY-CRM then placebo (MenACWY-CRM). Meningococci were isolated from oropharyngeal swabs collected before vaccination and at five scheduled intervals over one year. Primary analysis was cross-sectional carriage one month after the vaccine course; secondary analyses included comparison of carriage at any time point after primary analysis until study termination. Findings: 2954 subjects were randomised (control, n=987; 4CMenB, n=988; MenACWY-CRM, n=979); approximately one-third of each group was positive for meningococcal carriage at study entry. By one month, there was no significant difference in carriage between controls and 4CMenB (Odds Ratios (OR) [95% CI]; 1·2 [0·8−1·7]) or MenACWY-CRM (OR [95% CI], 0·9 [0·6–1·3]) groups. From three months after dose two, 4CMenB vaccination resulted in significantly lower carriage of any meningococcal strain (calculated efficacy 18·2% [95% CI: 3·4–30·8]) and capsular groups BCWY (calculated efficacy 26·6% [95% CI: 10·5–39·9]) compared to control vaccination. Significantly lower carriage rates were also observed in the MenACWY-CRM group compared with controls: calculated efficacies 39·0% [95%CI: 17·3-55·0] and 36.2% [95%CI: 15·6-51·7] for serogroups Y and CWY, respectively. Interpretation: MenACWY-CRM and 4CMenB vaccines reduced meningococcal carriage rates over 12 months post-vaccination and, therefore, may affect transmission where widely implemented

    Lentiviral-Mediated Transgene Expression Can Potentiate Intestinal Mesenchymal-Epithelial Signaling

    Mesenchymal-epithelial signaling is essential for the development of many organs and is often disrupted in disease. In this study, we demonstrate the use of lentiviral-mediated transgene delivery as an effective approach for ectopic transgene expression and an alternative to generation of transgenic animals. One benefit to this approach is that it can be used independently or in conjunction with established transgenic or knockout animals for studying modulation of mesenchymal-epithelial interactions. To display the power of this approach, we explored ectopic expression of a Wnt ligand in the mouse intestinal mesenchyme and demonstrate its functional influence on the adjacent epithelium. Our findings highlight the efficient use of lentiviral-mediated transgene expression for modulating mesenchymal-epithelial interactions in vivo
