    Organic Cation Transporter 3: A Cellular Mechanism Underlying Rapid, Non-Genomic Glucocorticoid Regulation of Monoaminergic Neurotransmission, Physiology, and Behavior

    Corticosteroid hormones act at intracellular glucocorticoid receptors (GR) and mineralocorticoid receptors (MR) to alter gene expression, leading to diverse physiological and behavioral responses. In addition to these classical genomic effects, corticosteroid hormones also exert rapid actions on physiology and behavior through a variety of non-genomic mechanisms, some of which involve GR or MR, and others of which are independent of these receptors. One such GR-independent mechanism involves corticosteroid-induced inhibition of monoamine transport mediated by “uptake2” transporters, including organic cation transporter 3 (OCT3), a low-affinity, high-capacity transporter for norepinephrine, epinephrine, dopamine, serotonin and histamine. Corticosterone directly and acutely inhibits OCT3-mediated transport. This review describes the studies that initially characterized uptake2 processes in peripheral tissues, and outlines studies that demonstrated OCT3 expression and corticosterone-sensitive monoamine transport in the brain. Evidence is presented supporting the hypothesis that corticosterone can exert rapid, GR-independent actions on neuronal physiology and behavior by inhibiting OCT3-mediated monoamine clearance. Implications of this mechanism for glucocorticoid-monoamine interactions in the context-dependent regulation of behavior are discussed

    Identification of the YfgF MASE1 domain as a modulator of bacterial responses to aspartate

    Complex 3'-5'-cyclic diguanylic acid (c-di-GMP) responsive regulatory networks that are modulated by the action of multiple diguanylate cyclases (DGC; GGDEF domain proteins) and phosphodiesterases (PDE; EAL domain proteins) have evolved in many bacteria. YfgF proteins possess a membrane-anchoring domain (MASE1), a catalytically inactive GGDEF domain and a catalytically active EAL domain. Here, sustained expression of the Salmonella enterica spp. Enterica ser. Enteritidis YfgF protein is shown to mediate inhibition of the formation of the aspartate chemotactic ring on motility agar under aerobic conditions. This phenomenon was c-di-GMP-independent because it occurred in a Salmonella strain that lacked the ability to synthesize c-di-GMP and also when PDE activity was abolished by site-directed mutagenesis of the EAL domain. YfgF-mediated inhibition of aspartate chemotactic ring formation was impaired in the altered redox environment generated by exogenous p-benzoquinone. This ability of YfgF to inhibit the response to aspartate required a motif, (213)Lys-Lys-Glu(215), in the predicted cytoplasmic loop between trans-membrane regions 5 and 6 of the MASE1 domain. Thus, for the first time the function of a MASE1 domain as a redox-responsive regulator of bacterial responses to aspartate has been shown

Methods of improving efficiency of ground operations were assessed and technology elements that could reduce cost identified. Emphasis is on: (1) specific technology items and (2) management approaches required to develop and support efficient ground operations.     Increases in the size, complexity, and life requirements of satellites and space vehicles have put increasing demands on the lubrication requirements for trouble-free service. Since the development costs of large systems are high, long lives with minimum maintenance are dictated. The Space Station represents the latest level of size and complexity in satellite development; it will be nearly 100 meters in major dimensions and will have a life requirement of thirty years. It will have numerous mechanisms critical to its success, some of which will be exposed to the space environment. Designing long-life lubrication systems and choosing appropriate lubricants for these systems will be necessary for their meeting the requirements and for avoiding failures with associated dependent mechanisms. The purpose of this program was to identify the various critical mechanisms and review their designs during the overall design and development stage so that problem areas could be avoided or minimized prior to the fabrication of hardware. The specific objectives were fourfold: (1) to perform a tribology survey of the Space Station for the purpose of documenting each wear point as to materials involved, environmental conditions, and operating characteristics; (2) to review each wear point (point of relative motion) as to the lubrication used and substrate materials selected in the context of its operating characteristics and the environmental conditions imposed; (3) to make recommendations for improvement in areas where the lubricant chosen and/or where the substrate (materials of the wear couple) are not considered optimum for the application; and (4) to make or recommend simulated or full scale tests in tribological areas where the state-of-the-art is being advanced, in areas where new designs are obviously being employed and a critical review would indicate that problems are a strong possibility, and/or where excessive wear, a malfunction, or excessive leakage would create fluid systems problems or contamination of exposed optical equipment

    This chapter looks at the regulatory environment for privacy in the open data environment in the United Kingdom, and civil society work on privacy in the same context. The temporal coverage of the chapter is 2011, when the open data movement began to accelerate in the UK, to 2019. The chapter begins by acknowledging the political nature of data, framing the discussion in terms of data politics and notions of open societies and environments. Next, it surveys the privacy laws, regulations, policies, and resources issued by the state (“privacy from above”) before looking at the resources and campaigns for privacy coming out of civil society (“privacy from below”). The authors suggest that civil society action is less concerned with data release than with methods of data collection and sharing between public and private sector actors
