    Epidemiological review and clinicopathological study of gastric adenocarcinoma cases in a tertiary care center in North Chennai, India

    Background: Gastric carcinoma is the third most common cause of cancer-related mortality worldwide. The Incidence of gastric carcinoma shows wide geographic and regional variation as well. The clinical presentation varies with the individual and hence histopathological evaluation plays a major role in the diagnosis and management of the patients. This study aims to evaluate the epidemiological and clinicopathological profile of gastric adenocarcinoma cases encountered in a tertiary care center in North Chennai where only limited statistical data available in literature.Methods: All the gastric adenocarcinoma cases diagnosed by histopathology who underwent total/subtotal gastrectomy were retrospectively collected for clinical as well as histopathological details for a year. Descriptive statistics were used for analysis.Results: The mean age of the study population is 59.3±11.3 with male: female sex ratio of 2.1:1. Higher prevalence noted in lower socioeconomic status (80%) and alcoholics (65%) with a history of high salted diet (62%). Most common presenting symptom was dyspepsia (94%). Upper GI endoscopy revealed antrum (42%) as the commonest site of tumor and majority were more than 5 cm size (75%). 62% of tumors were of moderately differentiated, predominantly intestinal type (86%) and mostly found to be in stage 3 (50%).Conclusions: Gastric adenocarcinoma was more common in elderly males and majority presented in advanced stage at the time of diagnosis. The General public needs to be creating awareness about variable nonspecific symptoms of an early stage of gastric carcinoma as well as the risk of poor dietary habits (high salted diet) and can enforce screening of high-risk category people

    Detection of illegal traffic pattern using Hybrid Improved CART and Multiple Extreme Learning Machine Approach

    In the proposed hybrid intrusion detection process, misuse detection and anomaly detection model is integrated to detect the attack in traffic pattern. In misuse detection model, the traffic pattern is classified into known attack and not known attack. Each extracted normal data set does not have known attack and it contains small amount of varied connection patterns than overall normal data set. Anomaly detection model classifies the not known attack as normal data set and unknown attack thus improving the performance of normal traffic behavior. Experiment is carried out using NSL –KDD dataset and performance of proposed approach is compared with traditional learning approaches in terms of training time, testing time, false positive ratio and detection ratio. The proposed method detects the known attacks and unknown attacks with ratio of 99.8 % and 52% respectively

    Antidiabetic activity of Manomani chooranam aqueous extract on female wistar albino rats

    Background: The aim of the present study was to evaluate the antidiabetic potential of Manomani chooranam (MMC), an indigenous polyherbal siddha formulation in Streptozotocin (STZ) induced diabetic female Wistar albino rats.Methods: Aqueous extract of MMC was prepared. Wistar albino rats were divided into six groups (n=6). Group 1 was kept as normal control, Group 2, 3, 4, 5 and 6 were induced diabetes. After induction, the group 2 was kept as diabetes control; Group 3 received the standard drug metformin (100 mg/kg), whereas Groups 4, 5 and 6 were treated with the aqueous extract of MMC at 500 mg/kg, 1000 mg/kg and 1250 mg/kg doses, respectively for the 21 days. Blood sugar was estimated at the end of each week. At the end of the study, rats were sacrificed and the pancreas was analyzed for histopathological changes. Data expressed as mean±standard error of the mean. Statistical analysis was done using one-way ANOVA followed by post hoc Tukey's test. p<0.05 considered statistically significant.Results: The groups which received aqueous extract of MMC at 500 mg/kg, 1000 mg/kg and 1250 mg/kg showed a significant decrease in the mean blood sugar level when compared to normal level. The groups which received MMC shows significant reduction in blood sugar level in comparison with standard drug metformin 100 mg/kg.Conclusions: The aqueous extract of MMC was able to decrease the elevated blood sugar levels in dose dependent manner

    Maternal Manganese Restriction Increases Susceptibility to High-Fat Diet-Induced Dyslipidemia and Altered Adipose Function in WNIN Male Rat Offspring

    Growth in utero is largely a reflection of nutrient and oxygen supply to the foetus. We studied the effects of Mn restriction per se, maternal Mn restriction, and postnatal high-fat feeding in modulating body composition, lipid metabolism and adipocyte function in Wistar/NIN (WNIN) rat offspring. Female weanling, WNIN rats received ad libitum for 4 months, a control or Mn-restricted diet and were mated with control males. Some restricted mothers were rehabilitated with control diet from conception (MnRC) or parturition (MnRP), and their offspring were raised on control diet. Some restricted offspring were weaned onto control diet (MnRW), while others continued on restricted diet throughout (MnR). A set of offspring from each group was fed high-fat diet from 9 months onwards. Body composition, adipocytes function, and lipid metabolism were monitored in male rat offspring at regular intervals. Maternal manganese restriction increased the susceptibility of the offspring to high-fat-induced adiposity, dyslipidaemia, and a proinflammatory state but did not affect their glycemic or insulin status

    NGAL is downregulated in oral squamous cell carcinoma and leads to increased survival, proliferation, migration and chemoresistance

    Oral cancer is a major public health burden worldwide. The lack of biomarkers for early diagnosis has increased the difficulty in managing this disease. Recent studies have reported that neutrophil gelatinase-associated lipocalin (NGAL), a secreted glycoprotein, is upregulated in various tumors. In our study, we found that NGAL was significantly downregulated in primary malignant and metastatic tissues of oral cancer in comparison to normal tissues. The downregulation of NGAL was strongly correlated with both degree of differentiation and stage (I–IV); it can also serve as a prognostic biomarker for oral cancer. Additionally, tobacco carcinogens were found to be involved in the downregulation of NGAL. Mechanistic studies revealed that knockdown of NGAL increased oral cancer cell proliferation, survival, and migration; it also induced resistance against cisplatin. Silencing of NGAL activated mammalian target of rapamycin (mTOR)signaling and reduced autophagy by the liver kinase B1 (LKB1)-activated protein kinase (AMPK)-p53-Redd1 signaling axis. Moreover, cyclin-D1, Bcl-2, and matrix metalloproteinase-9 (MMP-9) were upregulated, and caspase-9 was downregulated, suggesting that silencing of NGAL increases oral cancer cell proliferation, survival, and migration. Thus, from our study, it is evident that downregulation of NGAL activates the mTOR pathway and helps in the progression of oral cancer

    Whitebacked planthopper Sogatella furcifera (Horváth) (Homoptera: Delphacidae) resistance in rice variety Sinna Sivappu

    Whitebacked planthopper (WBPH) along with brown planthopper (BPH) has emerged as a major pest of rice in several Asian countries. Development and cultivation of varieties resistant to both planthoppers is an ecologically acceptable strategy to manage these pests. Sinna Sivappu, a Sri Lankan landrace, was reported to be resistant to both planthoppers. While inheritance of BPH resistance has been reported, the genetics of WBPH resistance in this variety is not known. Using a mapping population of 255 F2:3 families from Taichung Native (TN)1/Sinna Sivappu cross and 128 polymorphic simple sequence repeat (SSR) markers, genes or quantitative trait loci (QTLs) for WBPH resistance quantified in ten phenotypic tests were identified, adopting classical Mendelian segregation, correlation and QTL analyses. The inheritance pattern suggested that a single recessive gene controlled regulation of seedling damage score. Antixenosis or nymphal preference was influenced by two complementary recessive genes, whereas tolerance in terms of days to wilt was under the influence of a single dominant gene. Several of these phenotypic tests recorded high degree of positive or negative correlation between them, suggesting dependence or redundancy of the tests. QTL analysis revealed 13 loci associated with nine traits. Five major-effect QTLs were detected for damage score (chromosome 6), nymphal survival (chromosome 12), and days to wilt (three QTLs on chromosome 4). We suggest involvement of four WBPH resistance genes in Sinna Sivappu, designated as wbph9(t), wbph10(t), wbph11(t), and Wbph12(t). One of the recessive genes could be allelic to any of the recessive genes reported in cluster C on chromosome 6 which might confer resistance to both BPH and WBPH

The tumor necrosis factor-a-induced protein 8-like (TIPE/TNFAIP8) family is a recently identified family of proteins that is strongly associated with the regulation of immunity and tumorigenesis. This family is comprised of four members, namely, tumor necrosis factor-a-induced protein 8 (TIPE/TNFAIP8), tumor necrosis factor-a-induced protein 8-like 1 (TIPE1/TNFAIP8L1), tumor necrosis factor-a-induced protein 8-like 2 (TIPE2/TNFAIP8L2), and tumor necrosis factor-a-induced protein 8-like 3 (TIPE3/TNFAIP8L3). Although the proteins of this family were initially described as regulators of tumorigenesis, inflammation, and cell death, they are also found to be involved in the regulation of autophagy and the transfer of lipid secondary messengers, besides contributing to immune function and homeostasis. Interestingly, despite the existence of a significant sequence homology among the four members of this family, they are involved in different biological activities and also exhibit remarkable variability of expression. Furthermore, this family of proteins is highly deregulated in different human cancers and various chronic diseases. This review summarizes the vivid role of the TIPE family of proteins and its association with various signaling cascades in diverse chronic diseases
